    Notes: Recent advances suggest a significant role for the HLA-C locus as a target of alloreactions after bone marrow transplantation. The biological importance of products of the HLA-C locus, both as transplant antigens and as ligands for natural killer (NK) cells, is well established. A total of 10 different serologically defined HLA-Cw antigen specificities (Cw1–Cw10) are encoded by the C locus; however, there are now 151 different alleles that can be identified by molecular methods. Serological definition of Cw alleles therefore includes 20–50% blanks, which cannot be detected by the available antisera. We used the molecular method of polymerase chain reaction (PCR)-based sequence-specific amplification and probe hybridization to define Cw alleles in 91 individuals from the Maratha community, and compared the data with data for 92 serologically typed Maratha individuals from India. We identified Cw*12, Cw*14, Cw*15, Cw*16 and Cw*18, along with the serologically identified Cw*01, Cw*02, Cw*03, Cw*04, Cw*06 and Cw*07 alleles. The HLA-Cw blank allele frequency in the Maratha was reduced from 0.5706 to 0.00. Furthermore, by using a molecular technique, it was possible to identify novel allele subtypes, such as Cw*0104, Cw*0203 and Cw*0707, and a high frequency of Cw* 1801 in the Maratha community compared with other Indian and world populations. Our results will have clinical implications in related and unrelated HLA-matched bone marrow transplantation in India.

    Notes: Abstract Imide-aryl ether ether ketone block copolymers were prepared and the adhesion characteristics with poly(ether-imide) were investigated. The copolymers were prepared via the poly(amic alkyl ester) precursor to the polyimides which is hydrolytically stable and may be isolated and characterized prior to imidization. Solutions of the copolymers were cast and cured to effect the imidization, producing clear tough films which showed two transitions, indicative of a multiphase morphology. Mixtures of the copolymers with poly(ether-imide) also produced clear films, and the shift in the Tg of the aryl ether ether ketone component of the block copolymer indicated miscibility with the poly(ether-imide) within this phase. This miscibility of the poly(ether-imide) with the aryl ether ether ketone component of the block copolymer produced significant improvements in the adhesion of the thermoplastic poly(ether-imide) with the rigid polyimide copolymer.

    Notes: The synthesis and characterization of amorphous, aromatic, polyaryl ethers has been investigated. Detailed synthesis procedures are provided. The main thrust of these activities was to synthesize polymers which could serve as castable amorphous matrix resins for carbonfiber composites and which would be highly resistant to radiation degradation. It was important that these polymers not contain any aliphatic groups in order to contrast their behavior with the commercially available Bisphenol-A polysulfones. The approach taken was to synthesize a series of statistical or random copolymers derived from hydroquinone and biphenol together with 4,4′-dichlorodiphenyl sulfone. In order to produce copolymers of high molecular weight, modified techniques were developed which utilized potassium carbonate as a weak base and N-methyl-2-pyrrolidone as the aprotic dipolar solvent. Detailed procedures are provided in this paper for the synthesis of high molecular weight copolymers of this type. In addition, stress-strain, dynamic mechanical properties and preliminary electron beam (e-beam) degradation studies are reported significantly. Improved mechanical property retention after e-beam exposure was observed with the all aromatic polyether sulfones.

    Notes: The transport properties of the poly(arylether bissulfone) based on bisphenol A (PBSF) and the poly(arylether bisketone) s based on bisphenol A (PBK) and bisphenol S (PBK-S) are reported at 35°C. Comparisons are made with the polysulfone and the polycarbonate also based on bisphenol A to determine the effect of the long, rigid bisketone and bissulfone groups on polymer properties. A direct comparison also is made between PBK and PBSF, which differ only by their ketone and sulfone groups. The bulkier sulfone group increases free volume and Tg more than the ketone group. This results in higher solubility and diffusivity coefficients for the bissulfone versus the bisketone polymer, both of which contribute to higher permeability coefficients. © 1993 John Wiley & Sons, Inc.

    Description: In view of the historically devastating earthquakes in Ghana, this study employed regional satellite gravity data to reveal the extension of the crustal network of fractures emanating from the seismically active zone in the Gulf of Guinea and its implications on Coulomb stress transfer of the June 22nd, 1939 Accra earthquake. The Accra earthquake (M=6.5), Ghana is one of the most devastating intra-plate earthquakes in sub-Sahara west African region. In order to carry out the Coulomb stress transfer study, we utilized the Fault Plane Solutions (FPS) obtained from the waveform inversion of Yarwood and Doser (1979), which revealed that the earthquake composed of two events (6.1 Mw and 6.4 Mw). This study resolved the static Coulomb Failure Stress (CFS) change onto the finite fault models of the 6.4 Mw and 6.1 Mw earthquakes and its effect on associated receiver faults. The gravity study revealed swarms of fractures emanating from the gulf of Guinea and connected with the Akwapim and Coastal boundary faults favored the enhanced CFS to generate the 6.1 Mw due to transmission of stress from the seismically active zones in the Gulf of Guinea (boundary between the African plate and the South-American plate). The 6.4 Mw main shock was explained to correlate with release of seismic energy from the associated secondarily stressed prominent strike-slip (Akwapim) fault and strike-slip (coastal boundary fault). The direction of Coulomb stress transfer and the receiver faults of the 6.4 Mw main event have been mapped for the next big magnitude earthquake preparedness.

    Description: Bacterial phosphothreonine lyases catalyze a novel posttranslational modification involving formation of dehydrobutyrine/dehyroalanine by β elimination of the phosphate group of phosphothreonine or phosphoserine residues in their substrate proteins. Though there is experimental evidence for presence of dehydro amino acids in human proteins, no eukaryotic homologs of these lyases have been identified as of today. A comprehensive genome-wide search for identifying phosphothreonine lyase homologs in eukaryotes was carried out. Our fold-based search revealed structural and catalytic site similarity between bacterial phosphothreonine lyases and BLES03 (basophilic leukemia-expressed protein 03), a human protein with unknown function. Ligand induced conformational changes similar to bacterial phosphothreonine lyases, and movement of crucial arginines in the loop region to the catalytic pocket upon binding of phosphothreonine-containing peptides was seen during docking and molecular dynamics studies. Genome-wide search for BLES03 homologs using sensitive profile-based methods revealed their presence not only in eukaryotic classes such as chordata and fungi but also in bacterial and archaebacterial classes. The synteny of these archaebacterial BLES03-like proteins was remarkably similar to that of type IV lantibiotic synthetases which harbor LanL-like phosphothreonine lyase domains. Hence, context-based analysis reinforced our earlier sequence/structure-based prediction of phosphothreonine lyase catalytic function for BLES03. Our in silico analysis has revealed that BLES03-like proteins with previously unknown function are novel eukaryotic phosphothreonine lyases involved in biosynthesis of dehydro amino acids, whereas their bacterial and archaebacterial counterparts might be involved in biosynthesis of natural products similar to lantibiotics.
