    Notes: Sugar beet (Beta vulgaris L.) root suspension-cultured cells were converted to protoplasts which responded to fusicoccin (FC) by a rise in cytoplasmic pH (pHcyt) averaging 0.25 units in the fluorimetric assay. This effect was blocked by erythrosin B, a specific inhibitor of the plasma membrane H+-ATPase. A protein kinase inhibitor, staurosporine also caused cytosolic alkalinization that was sensitive to H+-ATPase inhibitors. Most strikingly, the effect of staurosporine was suppressed by fusicoccin and vice versa. Addition of okadaic acid, entailing overall protein phosphorylation, also led to H+-ATPase activation, whereupon fusicoccin lost its effect on proton transport. In parallel, kinetic and inhibitor analyses demonstrated that FC binding to the protoplast plasma membrane involved two sites with dissociation constants of 1 nM and 0.2 μM and was indifferent to phosphorylation and dephosphorylation inhibitors. Thus, it could be concluded that (1) the effect of FC on cytoplasmic pH probably depends on the phosphorylation state of plasma membrane proteins and may have either sign; (2) the activation of H+-ATPase by FC most likely proceeds directly through conformational receptor-enzyme interaction.

    Notes: The osmotic water permeability (Pos) of cell membranes isolated from leaves of 40-, 50- and 60-day-old Mesembryanthemum crystallinum plants was estimated by measuring light-scattering kinetics using stopped-flow spectrophotometry. The measurements were performed on the plasma membrane (PM), purified tonoplast (TP), and TP-enriched vesicles. The PM and TP-enriched vesicles were obtained by partitioning the microsomal fraction in an aqueous polymer two-phase system, whereas the purified TP vesicles were prepared by microsomal vesicle flotation on a sucrose cushion. The Pos of isolated membranes declined with plant age. The kinetic experiments showed that there was no difference between the Pos of the PM and TP isolated from plants of all ages. A 24-h exposure of plants to 400 mM NaCl caused a decline in the Pos as well. These findings suggest that, during M. crystallinum transition to CAM, which was induced by plant ageing or salinity, plant osmoregulatory responses included changes in the Pos of the leaf-cell membranes. These variations in the Pos are discussed in the context of adaptive mechanisms responsible for the maintenance of the water balance in the common ice plant.

    Notes: Exposure of plants to elevated temperatures induces a complex set of changes that enable plants to adapt following heat stress. In order to test the effect of Ca2+ on heat shock-induced changes in cell protein synthesis the incorporation of [S]methionine into protein was studied in cultured sugar beet (Beta vulgaris L.) cells incubated in media containing different calcium concentrations. Heat shock inhibited the synthesis of non-heat shock proteins (non-HSPs) and promoted the synthesis of a set of HSPs, typical of plants. The synthesis of non-HSPs was greatly inhibited by external Ca2+ removal by treatment of the cells with ethylene glycol-bis(β-aminoethylether)-N,N,N′,N′-tetraacetic acid. In contrast, extracellular Ca2+ appeared not to be strictly required for the de novo production of HSPs, but this cation exerted different effects on the synthesis of individual HSPs. Cell injury increased if the cells were exposed simultaneously to high temperature and Ca2+-deficient medium. Recovery of HSP synthesis and reduced cell injury were observed after addition of exogenous calcium to Ca2+-depleted cells. These findings are consistent with a Ca2+ requirement for the survival of the cells under heat shock, and likely for the development of cell thermotolerance.

    Notes: Abstract.  Taking the binding of fusicoccin to plasma membranes as an indicator of complex formation between the 14-3-3 dimer and H+-ATPase, we assessed the effect of osmotic stress on the interaction of these proteins in suspension-cultured cells of sugar beet (Beta vulgaris L.). An increase in osmolarity of the cell incubation medium, accompanied by a decrease in turgor, was found to activate the H+ efflux 5-fold. The same increment was observed in the number of high-affinity fusicoccin-binding sites in isolated plasma membranes; the 14-3-3 content in the membranes increased 2- to 3-fold, while the H+-ATPase activity changed only slightly. The data obtained indicate that osmotic regulation of H+-ATPase in the plant plasma membrane is achieved via modulation of the coupling between H+ transport and ATP hydrolysis, and that such regulation involves 14-3-3 proteins.

    Notes: Abstract DNA translation to the protein sequences determines the common usage of gene name as the enzyme identifier. The previously constructed single-family-member phylogenetic trees are produced by the pair alignment. The alignments strictly depend upon the user-defined parameters and algorithmic peculiarities, such as but not limited to: homology matrix, initial gap penalty value and gap elongation function. This rises the necessity to create complete clusterization which reflects the protein primary structure relationships. This protein-based clusterization should be made using the objective pair alignment. The standard dynamic alignment procedure is modified in order to discriminate between the suboptimal resulting scores. The special function treats the presence of continuous matching n-tuples as a good property of alignment. Pair alignment is objectified by finding the optimal gap penalty, that allows to get the maximal difference in identity between random and relative sequences. The method is applied to the cytochrome P450 superfamily. Our sample also contained 15 nitric oxide synthases and 30 random sequences. The similarity matrix, obtained by objective pair alignment, is worked up by standard UPGMA method.

    Notes: Summary Meiotic studies have been carried out in a series of 1100 infertile and sterile males. Of these, 599 cases have been studies in testicular biopsy, and 501, in semen samples. This is the largest meiotic series published so far. The incidence of meiotic anomalies was 4.3%. The most frequent chromosome abnormality was desynapsis (3.7%). However, the number of cases with a meiotic arest, usually due (73.9%) to synaptic anomalies in prophase I, was much higher (18.4%). An attempt is made to correlate the incidence of meiotic anomalies with the results of semen analysis. We discuss the prognosis of desynapsis, based on 41 cases studied, and reevaluate the results obtained in semen samples as compared with our previous results.

    Notes: Summary We describe the results of meiotic and synaptonemal complex (SC) studies in a selected series of 45 subfertile males with different meiotic and seminal alterations. SC anomalies (pairing anomalies, fragmented SCs, or presynaptic arrest) were observed in 32 cases (71.1%). In 31% of the abnormal cases, meiotic anomalies could only be detected through the study of SCs. The origin of synaptic anomalies may be related to the assembly of myosin molecules along the chromosomes. SC analysis should become routine in the study of subfertile males.

    Notes: Summary We describe in this paper the human male synaptic cycle using light and electron microscopy and the distribution of cells in the different stages of prophase I. The pattern of chromosome pairing and synapsis is an important tool to determine accurately whether a given synaptic behavior in infertile or sterile men is really abnormal or not. The relationship of prepachytene to pachytene cells is also important for the diagnosis of the different types of meiotic arrest at the primar spermatocyte level.
