    Notes: The gross and microscopic anatomy of the venom producing parotoid glands of Bufo alvarius has been studied by light and electron microscopy. Histochemical reactions for the presence of venom constituents and of components in biochemical pathways in the synthesis and release of venom were performed. The gland is composed of numerous lobules. Each lobule is an individual unit with a lumen surrounded by a double cell layer. Microvilli of the outer layer interdigitate with microvilli of the inner layer. Cells of the outer layer resemble smooth muscle cells, are rich in adenosine triphosphatase and glucose6-phosphatase, and contain numerous pinocytotic vesicles, glycogen granules and various organelles, These organelles include “crystalloids” of what seem to be highly organized agranular reticulum. These outer layer cells probably function in some aspects of venom synthesis, active cellular transport and contraction in the discharge of the secretory product. The inner cell layer demonstrates a positive chromaffin reaction, contains steroid material, various organelles, some pinocytotic vesicles and glycogen granules, and appears devoid of a plasmalemma on its inner surface. This layer is probably involved in venom formation and release via an apocrine type of secretion.Bufo alvarius parotoid gland shows significant morphological and histochemical differences from that of B. marinus and more nearly resembles a typical steroid producing organ.

    Notes: Ultrastructural studies on blood leukocytes of the channel catfish, Ictalurus punctatus, show the presence of heterophils (neutrophils), small lymphocytes, monocytes, and thrombocytes. Monocytes cannot always be distinguished from large lymphocytes. Cells resembling macrophages or transitional forms between monocytes and macrophages are occasionally seen. Blood eosinophils and basophils are not found. Thrombocytes and small lymphocytes are the most abundant leukocytes, while monocytes are the least frequently encountered leukocyte. Glycogen, present in all leukocytes, is most abundant in heterophils and least abundant in monocytes. Although monocytes are similar to heterophils in size and shape, a greater amount of rough endoplasmic reticulum, free ribosomes, and fewer granules are observed in monocytes. Heterophils possess oval or elongate granules, which often contain a crystalline or striated structure; small tubules which resemble smooth endoplasmic reticulum, and cristae which traverse the long axes of the mitochondria are frequently seen. Small lymphocytes are characterized by the presence of pseudopodia, many free ribosomes, numerous large mitochondria, dictyosomes (Golgi), and long profiles of rough endoplasmic reticulum. The dictyosomes are often associated with a large zone of exclusion. Bundles of microtubules are observed near the elongated ends of thrombocytes. Deep indentations of the plasmalemma, which give the appearance of vacuoles, are also seen in thrombocytes.

    Notes: Summary Using histochemical techniques, the reactivities of selected enzymes and other metabolic components were examined in the myocardium, coronary arteries, and coronary arterioles of normal, two-week-sympathectomized, and sham-operated canine hearts. There were no differences in the histochemistry of coronary arteries in any of the hearts, but important differences were noted in the myocardium and especially in the arterioles. The reactivities of the enzyme glucose-6-phosphate dehydrogenase and the nucleic acids were increased in arterioles of the sympathectomized heart, possibly indicating an increased protein synthesis. The reactivities of succinate dehydrogenase, NAD-isocitrate dehydrogenase, and cytochrome oxidase were reduced in myocardium and arterioles of sympathectomized hearts as well as in arterioles of sham-operated hearts; the changes were greater in the sympathectomized arterioles where there was also observed an increase in reactivity of lactate dehydrogenase. These findings suggest a depression in aerobic metabolic capacity and, in the case of the sympathectomized arteriole, imply a possible shift in adaptation from aerobic to anaerobic metabolism.

    Notes: Summary Articular—epiphyseal cartilage from the femur of New Zealand rabbits was subjected to histochemistry for determination of the presence of metabolic enzymes along its zonal stratification. Glycolytic enzymes were strongly reactive in all of the zones. Krebs cycle enzymes, enzymes of the hexose monophosphate shunt and the respiratory chain enzymes showed a progressive increase in reactivity from the tangential zone through the top half of the epiphyseal zone. Indicators of lipid metabolism were fairly high in all regions of the cartilage.

The gland is composed of numerous individual lobules, each lobule surrounded by a double cell layer. The interior surface of the outer layer is thrown into small cytoplasmic projections which traverse an intercellular space and interdigitate with microvilli formed by the outer plasmalemma of the inner layer. The outer layer resembles smooth muscle-like cells, is rich in adenosine triphosphatase, contains many pinocytotic vesicles and various organelles and may function in some aspect of venom synthesis, active cellular transport and contraction in the discharge of the secretory product. The inner layer shows a positive chromaffin reaction, contains various organelles, appears devoid of a plasmalemma on its inner surface and is involved in venom formation and release via an apocrine type of secretion. The intercellular space is rich in PAS positive materials, while the secretory product, itself, demonstrates a positive chromaffin reaction. The significance of these findings is discussed.
