    Description: The bacterial ubiD and ubiX or the homologous fungal fdc1 and pad1 genes have been implicated in the non-oxidative reversible decarboxylation of aromatic substrates, and play a pivotal role in bacterial ubiquinone (also known as coenzyme Q) biosynthesis or microbial biodegradation of aromatic compounds, respectively. Despite biochemical studies on individual gene products, the composition and cofactor requirement of the enzyme responsible for in vivo decarboxylase activity remained unclear. Here we show that Fdc1 is solely responsible for the reversible decarboxylase activity, and that it requires a new type of cofactor: a prenylated flavin synthesized by the associated UbiX/Pad1. Atomic resolution crystal structures reveal that two distinct isomers of the oxidized cofactor can be observed, an isoalloxazine N5-iminium adduct and a N5 secondary ketimine species with markedly altered ring structure, both having azomethine ylide character. Substrate binding positions the dipolarophile enoic acid group directly above the azomethine ylide group. The structure of a covalent inhibitor-cofactor adduct suggests that 1,3-dipolar cycloaddition chemistry supports reversible decarboxylation in these enzymes. Although 1,3-dipolar cycloaddition is commonly used in organic chemistry, we propose that this presents the first example, to our knowledge, of an enzymatic 1,3-dipolar cycloaddition reaction.     Description: This paper examines the economic experience of past energy booms and that of current unconventional shale gas and oil development. We focus on key economic characteristics of gas and oil production such as its employment potential, its geography, and its boom-bust nature. This background is used to discuss important economic policy issues arising with unconventional oil and gas development such as taxation, governmental use of those revenues, preemption, and equity in the distribution of costs and benefits. The paper concludes with economic policy recommendations for states and communities affected by such development including not viewing oil and gas development as an effective long-run economic development strategy, leveraging short-run financial gains from the development into permanent advantages, and strengthening the capacity for local governments to understand and manage this activity.

    Description: Ubiquinone (also known as coenzyme Q) is a ubiquitous lipid-soluble redox cofactor that is an essential component of electron transfer chains. Eleven genes have been implicated in bacterial ubiquinone biosynthesis, including ubiX and ubiD, which are responsible for decarboxylation of the 3-octaprenyl-4-hydroxybenzoate precursor. Despite structural and biochemical characterization of UbiX as a flavin mononucleotide (FMN)-binding protein, no decarboxylase activity has been detected. Here we report that UbiX produces a novel flavin-derived cofactor required for the decarboxylase activity of UbiD. UbiX acts as a flavin prenyltransferase, linking a dimethylallyl moiety to the flavin N5 and C6 atoms. This adds a fourth non-aromatic ring to the flavin isoalloxazine group. In contrast to other prenyltransferases, UbiX is metal-independent and requires dimethylallyl-monophosphate as substrate. Kinetic crystallography reveals that the prenyltransferase mechanism of UbiX resembles that of the terpene synthases. The active site environment is dominated by pi systems, which assist phosphate-C1' bond breakage following FMN reduction, leading to formation of the N5-C1' bond. UbiX then acts as a chaperone for adduct reorientation, via transient carbocation species, leading ultimately to formation of the dimethylallyl C3'-C6 bond.     Notes: An acid-tolerant fungus, Scytalidium acidophilum, was cultivated in peat extract for potential use as a single-cell protein (SCP) source. In shake flask fermentations with a 0.3% yeast extract diluted peat extract medium, it was found that the best conditions for growth occurred at pH 2.0 and 25°C. The maximum biomass concentration was produced after 10 days, with no significant differences for agitation rates between 100 and 200 rpm. Microbial contamination was not observed in nonaseptic operations. The protein content and amino acid composition of the mycelium produced compares with those reported for S. acidophilum grown in other fermentation media. The advantages of growing S. acidophilum in acid peat extract are presented.

    Notes: An efficient method to grow Escherichia coli W to high cell concentrations on the pilot scale is described and discussed. The method involves growth linked introduction of glucose; and ammonia to the culture, sparing with oxygen, and maintenance of aerobic conditions by gradually decreasing the temperature in the culture in order to keep the oxygen demand within the limits of the capacity of supply. Under these conditions the linear rate of cell mass production is actually the result of exponential growth with a gradually decreasing growth-rate constant.About 10 kg packed cells were produced in a 50 liter working-volume fermentor in one run of 13 hr. The concentration of the cells at the end of the growth was about 47 g dry cells/liter. The expenditure for nutrients was minimal and the controls were of simple automatic nature. From the determined yield constants for glucose, nitrogen, phosphorus, and oxygen it may be inferred that the cells grown by this method are similar to those grown exponentially at constant temperature.

    Description: Numerical geochemical modeling was used to study the effects on pore-water composition and mineralogy from carbon dioxide (CO 2 ) injection into the Pennsylvanian Morrow B Sandstone in the Farnsworth Unit in northern Texas to evaluate its potential for long-term CO 2 sequestration. Speciation modeling showed the present Morrow B formation water to be supersaturated with respect to an assemblage of zeolite, clay, carbonate, mica, and aluminum hydroxide minerals and quartz. The principal accessory minerals in the Morrow B, feldspars and chlorite, were predicted to dissolve. A reaction-path model in which CO 2 was progressively added up to its solubility limit into the Morrow B formation water showed a decrease in pH from its initial value of 7 to approximately 4.1 to 4.2, accompanied by the precipitation of small amounts of quartz, diaspore, and witherite. As the resultant CO 2 -charged fluid reacted with more of the Morrow B mineral matrix, the model predicted a rise in pH, reaching a maximum of 5.1 to 5.2 at a water–rock ratio of 10:1. At a higher water–rock ratio of 100:1, the pH rose to only 4.6 to 4.7. Diaspore, quartz, and nontronite precipitated consistently regardless of the water–rock ratio, but the carbonate minerals siderite, witherite, dolomite, and calcite precipitated at higher pH values only. As a result, CO 2 sequestration by mineral trapping was predicted to be important only at low water–rock ratios, accounting for a maximum of 2% of the added CO 2 at the lowest water–rock ratio investigated of 10:1, which corresponds to a small porosity increase of approximately 0.14% to 0.15%.
