Reactions with Organophosphorus Compounds, XLIII.1 - Structural Modifications of Nucleosides by Means of Triphenylphosphane/Diethyl AzodicarboxylateThe 3′-hydroxy group in nucleosides can be substituted easily by the nucleophiles azide and iodine with inversion of configuration using the system triphenylphosphane/diethyl azodicar-boxylate. In most cases neighboring group participation is not observed. Reaction of 5′-O-tritylthymidine (1) or 5′-O-trityldeoxyadenosine (3) with the reagent and HN3 leads to 1-(3′-azido-2′,3′-dideoxy-5′-O-trityl-β-D-threo-pentofuranosyl)thymine (2a) or 9-(3′-azido-2′,3′-didesoxy-5′-O-trityl-β-D-threo-pentofuranosyl)adenine (4a), respectively. In the same way 2′,5′-O-bistrityluridine (5) can be transformed into 1-(3′-azido-3′-desoxy-2′,5′-di-O-trityl-β-D-xylofuranosyl)uracil (6a), whereas 3′,5′-O-bistrityluridine (7) affords only the O2-2′-cyclouridine 8a under analogous conditions. Substitution of the 3′-hydroxy group of 5′-O-tritylthymidine (1) by iodine on reaction with the reagent and CH3I leads to 1-(3′-iodo-2′,3′-dideoxy-5′-O-trityl-β-D-threo-pentofuranosyl)thymine (2b) as well as 5′-O-trityl-O2-3′-cyclothymidine (2c).

Nucloeophile Substitutionen mit Hilfe des Systems Triphenylphosphan/Azodicarbonsäureester ermöglichen - wegen des weitgehenden Ausbleibens von Nachbargruppenbeteiligungen - den Austausch der sekundären 3′-Hydroxygruppe von Nucleosiden gegen Azid und Iod unter Inversion der Konfiguration. Aus 5′-O-Tritylthymidin (1) oder 5′-O-Trityldesoxyadenosin (3) entstehen mit dem genannten Reagenzsystem und N3H 1-(3′-Azido-2′,3′-didesoxy-5′-O-trityl-β-D-threo-pentofuranosyl)thymin (2a) bzw. 9-(3′-Azido-2′,3′-didesoxy-5′-O-trityl-β-D-threo-pentofuranosyl)adenin (4a). Analog bildet sich aus 2′,5′-O-Bistrityluridin (5) 1-(3′-Azido-3′-desoxy-2′,5′-di-O-trityl-β-D-xylofuranosyl)uracil (6a). 3′,5-O-Bistrityluridin (7) liefert dagegen unter den gleichen Bedingungen das O2-2′-Cyclonucleosid 8a. Bei der Substitution der 3′-Hydroxygruppe von 5′-O-Tritylthymidin (1) durch Iod mit Hilfe des genannten Reagenzsystems und CH3I entstehen sowohl 1-(3′-Iod-2′,3′-didesoxy-5′-O-trityl-β-D-threo-pento-furanosyl)thymin (2b) als auch 5′-O-Trityl-O2-3′-cyclothymidin (2c).
