    Notes: In order to investigate the extent and degree of heavy metal contamination in soils and sediments influenced by past mining activities, and to estimate the effects of indigenous bacteria on Cd speciation and bioremoval efficiency in sediments according to the various environmental conditions, tailings, soils, waters and sediments were collected from the Hwacheon Au-Ag-Pb-Zn mine in Korea. Elevated levels of metals, i.e., 29 As mg/kg, 30 Cd mg/kg, 261 Cu mg/kg, 601 Pb mg/kg and 3128 Zn mg/kg were found in sediments. High concentrations of As and other heavymetals in soils, waters and sediments were found near the tailings piles. From the results of sequential extraction analyses with tailings and soils, a high proportion of Cd is present as non-residual fractions. This indicates that Cd is more mobile and bioavailable than Cu, Pb and Zn. For seasonal variation, the concentrations of As and other heavy metals were much more elevated in soils collected before rainyseason than after rainy season. To estimate the bioremoval efficiency for Cd, bacteria which can adsorb Cd were isolated from sediments in this area. Microorganisms in sediments can alter the mobility of heavy metals and bacteria can leach heavy metals from sediments, adsorb them on their cell wall and accumulate them within an intracellular matrix. From the metal sorption tests with indigenous bacteria, the bioremoval efficiency of Cd in Cd single solution (10 ppm) was more than 90%. As well, the isolated bacterial strains exhibited a preferential metal sorption , i.e., Cd 〉 Pb 〉 Zn for single metal solutions and Pb 〉 Cd 〉 Zn for the mixed metal solutions. Therefore, the use of some microorganisms to remove heavy metals from contaminated sediments may represent an innovative purification process

    Notes: This study was made to investigate the extent and degree of As contamination in waters and sediments influenced by previous mining activity in the abandoned Dongil Au-Ag mine. Arsenic species was also identified in surface waters and groundwater. The chemical form of As in sediments was examined using sequential extraction analysis. The major contamination source of As in the mine area is suggested to be tailings with elevated levels of 8,718 As mg/kg. This was associated with the release of high level of As into the water system in the study area. The pH values of water samples ranged from 7.7 to 10.1, which was neutral to strong alkaline due tobuffering effect by high concentration of Ca and Mg dissolved from carbonate mineral and weathering of concrete. Stream waters contained high level of As within the range of 40.5 to 150.4 ㎍/L and most waters exceeded the permissible level (50 ㎍/L) of As for stream water in Korea. The concentration ratios of As(Ⅲ) to As (total), however, extended in the range of 5.8 to 75% and increased at low pH condition in stream waters. Arsenic concentration in sediments digested byaqua regia (HNO3+HCl) ranged from 162 to 2,077 mg/kg. The highest concentration of As may be due to the direct inflow of tailings. The relatively high percentage (17.1~35.3%) of As coprecipitated with amorphous Fe oxyhydroxides indicates that more severe contamination of As can occur by re-extracting due to changes of chemical environment such as reducing condition

    Notes: In order to investigate the levels of heavy metal contamination and to assess the risk of the adverse health effects on human exposure to toxic heavy metals influenced by past mining activities, environmental geochemical surveys were undertaken around abandoned metal mine sites (Okdong Cu-Pb-Zn, Dokok Au-Ag-Cu and Hwacheon Au-Ag-Pb-Zn mines). High concentrations of heavy metals were found in tailings from the Okdong (72 As mg/kg, 53.6 Cd mg/kg, 910 Cu mg/kg, 1,590Pb mg/kg, 5,720 Zn mg/kg), the Dokok (254 As mg/kg, 98.2 Cd mg/kg, 2,550 Cu mg/kg, 4,200 Pb mg/kg, 18,020 Zn mg/kg) and the Hwacheon (72 As mg/kg, 12.4 Cd mg/kg, 580 Pb mg/kg, 1,300 Zn mg/kg) mines. Elevated levels of As, Cd, Pb and Zn were also found in agricultural soils from these mine areas. Human risk effects are classified into a cancer effect and a toxic (noncancer) effect. Thehazard index representing a toxic risk for As was higher value than 1.0 in the Okdong and the Hwacheon mine sites, as was that for Cd in the Okdong and Dokok mine sites. Therefore, toxic risks for As and Cd exist via exposure (ingestion) to the contaminated soil, groundwater and rice grains in these mine areas. The cancer risk for As by the consumption of groundwater used as drinking water from the Okdong and the Hwacheon mine areas was 7E-4 and 1E-4, respectively. These risk levels exceed the acceptable risk (1 in 100,000) for regulatory purposes

    Notes: Abstract Purpose: Pre-clinical data have suggested that prolonged exposure to paclitaxel enhances its cytotoxicity, but various clinical trials utilizing long-term infusions of paclitaxel have been limited by unacceptable hematologic toxicity, most notably significant neutropenia. A phase I study of paclitaxel administered over 10 days, was performed to evaluate the hematologic and non-hematologic toxicities as well as to determine the maximum-tolerated dose for the 10-day infusion duration. Patients and methods: Twenty-nine solid tumor patients (predominantly non-small cell lung cancer and head and neck cancer) were treated with paclitaxel at doses ranging from 5 mg/m2/day to 25 mg/m2/day administered as a 10-day continuous infusion via a pump every 21 days. Dose escalation was permitted within individual patients. Dose-limiting toxicity (DLT) was defined as grade 3 or 4 non-hematologic toxicity, ANC ≤ 500 or platelet count ≤ 25,000 for ≥ 7 days or febrile neutropenia. The maximum tolerated dose (MTD) was defined as the highest dose level at which less than two out of six patients developed DLT. All of the patients had received prior chemotherapy; approximately two-thirds had received prior radiation as well. All patients received standard pre-medications for paclitaxel, including anti-histamines and corticosteroids. Prophylactic granulocyte colony-stimulating factor (G-CSF) was not used. Results: A total of 110 courses of paclitaxel were administered to 29 patients. The incidence of hematologic and non-hematologic toxicity was quite low among the patients treated at dose levels below 17 mg/m2/day. At higher doses, non-hematologic toxicities including arthralgias, myalgias, fatigue, nausea, stomatitis, and peripheral neuropathy were seen, although nearly all of the toxicities were less than grade 3 (NCI toxicity criteria). Hematologic toxicity mostly consisted of neutropenia and was more common at dose levels of 17 mg/m2/day or higher. Nevertheless, even at the highest dose levels (21 mg/m2/day and 25 mg/m2/day) grade 3 or 4 neutropenia occurred in only 50% of patients. Dose-limiting hematologic toxicity occurred in 2 of 4 patients treated at the 25 mg/m2/day dose level. Conclusion: Paclitaxel can be safely administered as a 10-day infusion. The MTD for this schedule is 210 mg/m2. Unlike the 96-hour paclitaxel infusions, dose-reduction for myelosuppression may not be necessary because the MTD of paclitaxel when administered over a 10-day infusion is similar to the MTD of paclitaxel when infused over 3 or 24 hours.

    Notes: Abstract The Okchon black shale in Korea provides an important example of natural geological materials containing toxic elements; the Chung-Joo, Duk-Pyung, Geum-Kwan, I-Won and Chu-Bu areas are underlain by these black shales and slates of the Guryongsan Formation. This formation is part of the Okchon Group which is found in the central part of Korea. Geochemical surveys were undertaken in these five study areas in the Okchon Zone in order to examine the level of enrichment and dispersion patterns of potentially toxic elements in rocks and soils. After appropriate preparation, samples were analysed by instrumental neutron activation analysis and inductively coupled plasma atomic emission spectrometry for a range of elements. Arsenic, Cu, Mo, V, U and Zn are highly enriched in the Okchon black shales and their mean concentrations are significantly higher than those in black slates. These elements are closely associated with one another from a geochemical viewpoint and may be enriched simultaneously. Mean concentrations of As, Mo and U in soils derived from black shales occurring in the Duk-Pyung and Chu-Bu areas are higher than the permissible level suggested by Kloke (1979), and the enrichment index decreases in the order of Duk- Pyung 〉 Chu-Bu 〉 Chung-Joo 〉 Geum-Kwan = I-Won areas. Uranium-bearing minerals such as uraninite and brannerite have been identified in black shales from the Chung-Joo area by electron probe micro-analysis. Uraninite grains are closely associated with monazite and pyrite with a grain size ranging from 2μm to 10μm whereas brannerite grains occur as a euhedral form 50μm in diameter.

    Notes: Upper aerodigestive tract tumorigenesis has been hypothesized to represent a field cancerization process with multistep events based on its association with known carcinogens, its frequent associated premalignant lesions, and its multifocal clinical manifestation. To further explore this working hypothesis, we have examined normal tissue and premalignant lesions in the field of tumors for evidence of genetic change. Paraffin sections of head and neck tumors harboring neighboring premalignant lesions were explored for the presence of chromosome polysomies using in situ hybridization and chromosome-specific centromeric probes. Cell exhibiting random polysomy were observed in the premalignant regions near the tumors. The frequency of polysomy in the tumor field increased as the tissue progressed from normal morphology (33%), to hyperplasia (67%), to dysplasia (95%), and to squamous cell carcinoma (96%). These results support the notions of field cancerization and multistep tumorigenesis in the aerodigestive tract. To determine whether the degree of accumulated genetic alterations might serve as a biomarker for risk of developing malignancy, a set of biopsies of oral premalignant lesions (leukoplakia, erythroplakia) were retrospectively chosen for polysomy analysis from two groups of individuals: one group who subsequently developed oral cancer and one group who did not develop oral cancer. Three of the five individuals who showed significant chromosome polysomies in their biopsies subsequently developed oral cancer, whereas only one of eight individuals with little evidence of polysomy subsequently progressed to oral cancer. These results suggest that evidence of generalized genetic change or instability might be useful as a genetic biomarker for risk assessment.

    Description: Among CO2-fixing metabolic pathways in nature, the linear Wood–Ljungdahl pathway (WLP) in phylogenetically diverse acetate-forming acetogens comprises the most energetically efficient pathway, requires the least number of reactions, and converts CO2 to formate and then into acetyl-CoA. Despite two genes encoding glycine synthase being well-conserved in WLP gene clusters, the functional role of glycine synthase under autotrophic growth conditions has remained uncertain. Here, using the reconstructed genome-scale metabolic model iSL771 based on the completed genome sequence, transcriptomics, 13C isotope-based metabolite-tracing experiments, biochemical assays, and heterologous expression of the pathway in another acetogen, we discovered that the WLP and the glycine synthase pathway are functionally interconnected to fix CO2, subsequently converting CO2 into acetyl-CoA, acetyl-phosphate, and serine. Moreover, the functional cooperation of the pathways enhances CO2 consumption and cellular growth rates via bypassing reducing power required reactions for cellular metabolism during autotrophic growth of acetogens.

    Description: In this study, the toxicities of two pharmaceuticals, verapamil and tramadol were evaluated in Daphnia magna using the conventional toxicity tests (acute and chronic test) and the expression patterns of five stress responsive genes. In the chronic toxicity test, several parameters, such as the survival percentage, the body length of D. magna , the time of first reproduction, and the number of offspring per female, were adversely affected during the exposure to 4.2 mg L −1 verapamil and 34 mg L −1 tramadol. During the 24-h short-term exposure, verapamil particularly caused a downregulated expression of the CYP4 and CYP314 genes, whereas tramadol upregulated the expression of the CYP314 gene. Neither pharmaceutical affected the expression of Dhb , Arnt , and Vtg . However, during the 21-day long-term exposure, both verapamil and tramadol significantly reduced the expression level of the Vtg gene, a biomarker of the reproduction ability in an oviparous animal, whereas neither affected the other genes. © 2011 Wiley Periodicals, Inc.
