    Notes: The synthesis of a series of novel cyano-containing copolymers is described. Alternating copolymers of acrylonitrile with vinyl esters are obtained by increasing the electrophilic character of the nitrile monomers by complexation with zinc chloride. Copolymers of methyl and ethyl α-cyanoacrylates with vinyl esters are prepared using radical initiators in the presence of 7% acetic acid as inhibitor for anionic polymerization. The copolymers of methyl α-cyanoacrylate with the vinyl esters have Tg's above 140°C. Methyl vinylidene cyanide (MVCN) copolymerizes spontaneously with para-substituted styrenes to yield copolymers with high inherent viscosities and high Tg (160°C) and the copolymer of MVCN with vinyl acetate is also synthesized. The pyroelectric constants p for these polymers were measured and the values of p for the copolymers of vinyl acetate with methyl β,β-dicyanoacrylate, methyl α-cyanoacrylate, or MVCN were in the same range as the well-studied vinylidene cyanide/vinyl acetate copolymer. A higher concentration of dipoles generally results in higher Tg's and higher pyroelectric coefficients. © 1992 John Wiley & Sons, Inc.

    Description: Background: Mesenchymal stem cells (MSCs) have the potential to differentiate into specialized cell lineages such as osteoblasts and adipocytes in vitro. There exists a reciprocal relationship between osteogenic and adipogenic differentiation of MSCs that an osteogenic phenotype occurs at the expense of an adipogenic phenotype and vice versa, which in turn influence one another?s phenotype through negative feedback loops. Thus, it is important to understand what signaling molecules modulate the lineage commitment of MSCs. Protein kinase C (PKC) plays a central role in cellular signal transduction for mediating diverse biological functions, and dysregulation of PKC activity is involved in various metabolic diseases including cancer, diabetes, and heart disease. Although the role of individual PKC isoforms has been investigated in various fields, the potential role of PKC in bone metabolism is not completely understood. In this study, we investigated the potential role of PKC? in osteogenic lineage commitment of human bone marrow-derived mesenchymal stem cells (hBMSCs). Results: We observed that expression and phosphorylation of PKC? were increased during osteogenic differentiation of hBMSCs. Pharmacological inhibition and genetic ablation of PKC? in hBMSCs resulted in a significant attenuation of osteogenic differentiation as evidenced by reduced ALP activity and ECM mineralization, as well as down-regulation of the expression of osteoblast-specific genes. These effects were also accompanied by induction of adipogenic differentiation and up-regulation of the expression of adipocyte-specific genes involved in lipid synthesis in osteogenic induction of hBMSCs. Additionally, the activation of AMPK, which is a key cellular energy sensor, induced osteogenesis of hBMSCs. However, the inhibition of AMPK activity by compound C did not affect the activation of PKC? at all, indicating that there is no direct correlation between AMPK and PKC? in osteogenesis of hBMSCs. Conclusions: These results suggest that PKC? is a critical regulator for the balance between osteogenesis and adipogenesis of hBMSCs and thus has a potential novel therapeutic target for the treatment of metabolic bone diseases.

    Description: This study sought to adapt the existing value framework (VF) to produce a reliable and valid Korean oncology VF. Two VFs developed by The American Society of Clinical Oncology (ASCO) and the European Society for Medical Oncology (ESMO) were selected for examination in the present study. Forward and backward translations were conducted for six high-priced drugs indicated for non-small-cell lung cancer and multiple myeloma. Inter-rater reliability was measured based on the intraclass correlation coefficient (ICC) and variation was described using the coefficient of variation. The relative weights of factors critically considered by Korean oncologists were derived following the analytic hierarchy process (AHP), and focus group interviews (FGIs) were used to obtain qualitative data regarding the applications of these two VFs in the Korean setting. The ICCs of the Korean VFs were 0.895 (0.654–0.983) for ASCO and 0.726 (0–0.982) for ESMO translations, suggesting excellent reliability for ASCO and good reliability for ESMO. AHP demonstrated that clinical benefit has the highest priority, which is consistent with the ASCO VF. The FGIs suggested that the result for AHP is acceptable and that both ESMO and ASCO VFs should be used complementarily. Although further evaluation with a larger sample size is needed, the Korean versions of ESMO/ASCO VFs are valid and reliable tools and are acceptable to Korean stakeholders, yet they should be applied with caution.
