    Description: The green fluorescent protein (GFP) from the Pacific Northwest jellyfish Aequorea victoria has generated intense interest as a marker for gene expression and localization of gene products. The chromophore, resulting from the spontaneous cyclization and oxidation of the sequence -Ser65 (or Thr65)-Tyr66-Gly67-, requires the native protein fold for both formation and fluorescence emission. The structure of Thr65 GFP has been determined at 1.9 angstrom resolution. The protein fold consists of an 11-stranded beta barrel with a coaxial helix, with the chromophore forming from the central helix.     Description: Two-phase cryogenic propellant pumping techniques developed during the past 5 years are discussed. During this period the theory of two-phase pumping of hydrogen was developed into an operational technique that can result in significant benefits to space vehicle propulsion systems. Using two-phase pumping techniques, zero LH2 tank net positive suction head (NPSH) was demonstrated on turbopump and rocket engine firings, and limited experimental results show that two-phase pumping of liquid oxygen is feasible. Design techniques were developed for improving the two-phase pumping capability of inducers.

    Description: This paper presents the results of an advanced turbine blade test program aimed at improving turbine blade low cycle fatigue (LCF) life. A total of 21 blades were tested in a blade thermal tester. The blades were made of MAR-M-246(Hf)DS and PWA-1480SC in six different geometries. The test results show that the PWA-1480SC material improved life by a factor of 1.7 to 3.0 over the current MAR-M-246(Hf)DS. The geometry changes yielded life improvements as high as 20 times the baseline blade made of PWA-1480SC and 34 times the baseline MAR-M-246DS blade.

    Description: The Space Shuttle main engine (SSME) alternate high-pressure liquid oxygen pump experienced synchronous vibration and ball bearing life problems that were program threatening. The success of the program hinged on the ability to solve these development problems. The design and solutions to these problems are engirded in the lessons learned and experiences from prior programs, technology programs, and the ability to properly conduct failure or anomaly investigations. The failure investigation determines the problem cause and is the basis for recommending design solutions. For a complex problem, a comprehensive solution requires that formal investigation procedures be used, including fault trees, resolution logic, and action items worked through a concurrent engineering-multidiscipline team. The normal tendency to use an intuitive, cut-and-try approach will usually prove to be costly, both in money and time and will reach a less than optimum, poorly understood answer. The SSME alternate high-pressure oxidizer turbopump development has had two complex problems critical to program success: (1) high synchronous vibrations and (2) excessive ball bearing wear. This paper will use these two problems as examples of this formal failure investigation approach. The results of the team's investigation provides insight into the complexity of the turbomachinery technical discipline interacting/sensitivities and the fine balance of competing investigations required to solve problems and guarantee program success. It is very important to the solution process that maximum use be made of the resources that both the contractor and Government can bring to the problem in a supporting and noncompeting way. There is no place for the not-invented-here attitude. The resources include, but are not limited to: (1) specially skilled professionals; (2) supporting technologies; (3) computational codes and capabilities; and (4) test and manufacturing facilities.

    Description: The results of an experimental program to explore the feasibility of pumping two-phase oxygen (liquid and gas) at the pump inlet are reported. Twenty-one cavitation tests were run on a standard J-2 oxygen pump at the MSFC Components Test Laboratory. All tests were run with liquid oxygen 5 to 10 K above the normal boiling point temperature. During ten tests run at approximately at the pump inlet were noted before complete pump performance 50 percent of the nominal operating speed, two phase conditions were achieved. Vapor volumes of 40 to 50 percent at the pump inlet were noted before complete pump performance loss. The experimental results compared to predictions. Nine cavitation tests run at the nominal pump speed over a 5 K temperature range showed progressively lower net positive suction head (NPSH) requirements as temperature was increased. Two-phase operation was not achieved. The temperature varying NPSH data were used to calculate thermodynamic effects on NPSH, and the results were compared to existing data.
