Notes: Abstract Performance characteristics of polyethylene tube dispensers containing a mixture of (E,E)-8,10-dodecadien-1-ol (1), dodecan-1-ol (2), and tetradecan-1-ol (3) were evaluated for suitability as a mating disruptant for codling moth control. The rate of loss of pheromone component from a dispenser at any time was found to be described by the equation: −dP/dt=(k 1 k 2 +k d)P whereP is the amount of pheromone component in the dispenser well;t is time;k 1 is the ratio of the amount dissolved in the dispenser wall to the amount in the dispenser well;k 1 is the ratio of the evaporation rate to the amount dissolved in the dispenser wall;k d is the rate constant for chemical decomposition. The evaporation rate,E, of a pheromone component at any time was given by:E=k 1 k 2 P For all three components during the first three weeks,k 1 decreased from ca. 0.25 to ca. 0.10 and was approximately constant thereafter. The decrease ofk 1 with time may have been caused by weather-induced cross-linking of the polyethylene. Over timek 2 was constant and was 1.27 ± 0.26 × 10−3/hr for1, 1.96 ± 0.33 × 10su−3/hr for 2, and 0.31± 0.05 × 10−3/hr for 3. Thek 2 was zero for2 and3 and 6.96 × 10−4 for1. After 150 days in an orchard in 1991, 95% of1 was lost from the dispensers (61% of the loss was by chemical decomposition and 39% by evaporation). The heat summation units in a Yakima valley orchard during 1991 were 4.7% above the average for the 1980–1991 period, while during 1990 they were the highest for this period (26% above average). After the first three weeks of dispenser aging, the regression line half-lives for1 for 1990 and 1991 were 31.0 and 35.1 days, respectively. The difference in temperature between 1990 and 1991 did not affect the half-life of1 very much because so much of the loss was from photochemically induced decomposition. Based on an estimate of the required minimum evaporation rate for mating disruption of 2 mg/ha-hr and a half-life of 35 days for1, 2345 dispensers/ha would be required for one application per season; 944 dispensers/ha for two applications per season; and 734 dispensers/ha for three applications per season. If a different emission rate of1 is required for reliable mating disruption, then the number of dispensers required would be changed proportionately.

Notes: Abstract Mating disruption treatments for the tufted apple bud moth (TABM),Platynota idaeusalis (Walker), were tested in small plot trials in apple orchards in Pennsylvania. Treatments were evaluated by fruit injury and by capture of male TABM in traps baited with synthetic pheromone sources or virgin females. The TABM pheromone is a two component isomeric blend ofE-11-tetradecen-1-ol (E11-14:OH) andE-11-tetradecenyl acetate (E11-14:Ac). A 50∶50 ratio of these two components was used in standard monitoring septa and in mating disruption treatments released from either hollow fibers (‘fiber’) or PVC tubes (‘PVC’). Other pheromone blends tested included a 90∶10 ratio of E11-14: Ac and E11-14:OH (‘EAc’) and its reverse (‘EOH’), mixture ‘EAc’ with 30% of the Z-isomers (‘low AEc’), and a blend similar to the preceding with 2% Z9-12:Ac (‘generic’). These other blends were released from multi tube tape (‘tape’) or Shin-Etsu type rope (‘rope’) dispensers. Seasonal dispenser release rate in mg ha−1 h−1 was ca. 30 for the ‘rope’ dispensers, 14 for ‘PVC’ and 6 for ‘fiber’. ‘EAc-tape’ and ‘EOH-tape’ were equally effective in reducing catches of males in traps baited with synthetic lures and in traps baited with virgin females. Both treatments also reduced fruit injury. ‘EAc-rope’, ‘fiber’ and ‘PVC’ also were generally effective; whereas, the ‘low EAc’ and ‘generic’ treatments reduced trap capture less than 90% and did not reduce fruit injury. Dispenser density was positively correlated with reduction in trap capture for the ‘low EAc-rope’ and ‘genericrope’ treatments. Traps loaded with ‘fiber’ dispensers captured more male TABM than the other treatments in non-pheromone permeated environments. Trap capture of other tortricids was reduced in pheromone treatments. ‘EAc-rope’ and the ‘TABM’ treatments provided mean (s.e.) percent reduction in trap catch of 99.5 (0.4) and 42.9 (10.1), respectively, for the redbanded leafroller,Argyrotaenia velutinana (Walker), and 90.4 (6.8) and 90.4 (1.3), respectively, for the obliquebanded leafroller,Choristoneura rosaceana (Harris).

Description: Lysosomes are responsible for degradation and recycling of bulky cell material, including accumulated misfolded proteins and dysfunctional organelles. Increasing evidence implicates lysosomal dysfunction in several neurodegenerative disorders, including Parkinson's disease and related synucleinopathies, which are characterized by the accumulation of α-synuclein (α-syn) in Lewy bodies. Studies of lysosomal proteins linked to neurodegenerative disorders present an opportunity to uncover specific molecular mechanisms and pathways that contribute to neurodegeneration. Loss-of-function mutations in a lysosomal protein, ATP13A2 (PARK9), cause Kufor–Rakeb syndrome that is characterized by early-onset parkinsonism, pyramidal degeneration and dementia . While loss of ATP13A2 function plays a role in α-syn misfolding and toxicity, the normal function of ATP13A2 in the brain remains largely unknown. Here, we performed a screen to identify ATP13A2 interacting partners, as a first step toward elucidating its function. Utilizing a split-ubiquitin membrane yeast two-hybrid system that was developed to identify interacting partners of full-length integral membrane proteins, we identified 43 novel interactors that primarily implicate ATP13A2 in cellular processes such as endoplasmic reticulum (ER) translocation, ER-to-Golgi trafficking and vesicular transport and fusion. We showed that a subset of these interactors modified α-syn aggregation and α-syn-mediated degeneration of dopaminergic neurons in Caenorhabditis elegans , further suggesting that ATP13A2 and α-syn are functionally linked in neurodegeneration. These results implicate ATP13A2 in vesicular trafficking and provide a platform for further studies of ATP13A2 in neurodegeneration.
