    Notes: Marine fishes, red drum Sciunops ocellatus. oblong rock fish Sebastes oblongus and Japanese flounder Purulichthys olivaceus showing clinical signs of nodavirus infection and mortality were subjected to histopathological and reverse-transcription polymerase chain reaction (RT-PCR) detection assay. Histologically, the diseased fish showed prominent vacuolisation in the brain, spinal cord, and retina of eyes. Fish nodavirus was detected by RT-PCR amplification in these fish species. Sequence analysis of the RT-PCR products were identical in comparison among T4 regions of Japanese flounder, red drum, and oblong rock fish isolates. From the sequence analysis, three isolates in this study could be included in the red-spotted grouper nervous necrosis virus (RGNNV), which is one of the clusters among four major clusters of NNV.

    Notes: Abstract: This study aims to determine the seasonal occurrence of marine birnavirus (MABV) at a coastal site in the Uwa Sea, Japan, in 1997 and 1998. To detect MABV from seawater, a simple method was developed for concentrating MABV by dialysis and ethanol precipitation. The concentrated virus was used for polymerase chain reaction (PCR), enzyme-linked immunosorbent assay, and virus isolation. Viral genome was detected through the experimental period. The amount of PCR product varied; it was small in summer, but increased from fall to winter. Viral protein was also detected, and the amount in the January sample was equivalent to approximately 102 TCID50 (50% tissue culture infectious dose) of the virus. However, infectious viruses were not isolated. This suggested that MABV was released from hosts to environmental seawater in winter and possibly degraded after release.

    Description: We have investigated human-stress monitoring by making use of salivary nitrate, which can be a candidate for stress markers, with ion-selective field-effect transistors (ISFETs). ISFETs are suitable for on-site single-drop analysis of salivary nitrate within 10 s. However, when ISFETs are used for salivary nitrate, ISFETs have a problem that is called the initial drift. The initial drift makes accurate nitrate monitoring difficult. Thus, the purpose of this study is to prevent the initial drift and to search for a new, simple polymer to possess a better performance of sensor responses than conventional matrix membranes, such as PVC. In this research, we investigated ISFETs using specific matrix membranes, for example KP-13, Pellethane®­­, and P7281-PU. The initial drift was evaluated from the fluctuations of the response values generated by the ISFETs when immersed in saliva or aqueous solution. As a result, P7281-PU showed a prevention effect on the initial drift, both in the whole saliva and in various solutions. Furthermore, the cause of drift may be H+ diffusion, and the drift prevention effect of P7281-PU may be affected by urethane bond capturing H+ in the ion-selective membrane. This result suggests that a continuous nitrate monitoring is feasible and may be applied to wearable sensors.
