    Notes: Abstract Obtaining an accurate, value for total ozone under a cloudy sky, especially when the sun is not high, is a major remaining problem associated with total ozone measurements. The Toronto spectrophotometer has been designed with this in mind. It has been fitted with a polarizing prism, and measures light at four wavelengths simultaneous which makes it possible to obtain two independent double ratios. Clouds produce two effects on ozone measurements; the first is purely an optical effect which causes an apparent increase in ozone, the second is most likely a real increase in ozone associated with large cumulus-type clouds. By considering the three following points it is possible to distinguish between these two cloud effects and probably measure the true total ozone for solar zenith angles less than 80°: 1. The multiply scattered component of polarized light is used to reduce optical cloud variance. This makes all skies appear like thick coulds. 2. A double difference similar to the AD method is used but the two ratios of the double difference are weighted inversely with

    Notes: Abstract The Toronto spectrophotometer was used to take an extensive series of ozone measurements for the period September 1971 to April 1972. As a result of these measurements it was found that short-lived variations of ozone occur which sometimes amount to more than 0.1 cm. These ‘ozone disturbances’ are advected because the same disturbance has been observed on the direct sun and the zenith sky at different times. Results which show the general nature of these disturbances will be presented. Also, a discussion will be given which stresses the need for an automated, mesoscale ozone-measuring network, capable of measuring in all weather conditions during all daytime hours.

    Notes: Summary A single intraperitoneal injection of ethane dimethanesulphonate (EDS) destroys all Leydig cells in the adult rat testis but 1–2 weeks later new foetal-type Leydig cells begin to regenerate within the interstitial tissue. A further EDS treatment at 4 weeks failed to kill the new population of foetal-type Leydig cells. Between 10–20 weeks, the new Leydig cells exhibited the characteristics of adult-type Leydig cells. These cells responded to another EDS treatment by exhibiting a second phase of complete degeneration followed by regeneration of a foetal-type and subsequently an adult-type cell population. The results indicate that the testis retains the ability to replenish its supply of Leydig cells despite successive phases of total degradation of Leydig cells.

    Notes: Summary The effects of FSH on the testicular interstitial tissue of immature hypophysectomized rats were studied by comparing morphological changes in Leydig cells with quantitative changes in interstitial tissue histology using morphometric analysis. Three groups of rats received subcutaneous injections of 0.5 ml saline vehicle or 10 μg rFSH or 20 ng oLH (equivalent to the amount of LH known to contaminate the FSH), twice daily for 7 days. Administration of FSH significantly increased testis weight and stimulated more advanced spermatogenesis compared to saline or LH. Morphometric analysis of testes of LH-treated rats showed a small but significant increase in total interstitial cell volume compared to saline treatment. FSH caused much greater increases in the total volume of interstitial tissue and interstitial cells than either saline or LH and significantly increased the total volume of interstitial fluid by comparison with the other groups. FSH but not saline or LH treatment resulted in a striking hypertrophy of Leydig cells, to produce cells ultrastructurally identical to Leydig cells from adults. Since the target tissue of FSH is the seminiferous epithelium, the observed effects on Leydig cells by FSH treatment suggest that the secretion of factors by the seminiferous tubules may mediate the maturation of Leydig cells.

    Notes: Summary The volume and surface area of lipid inclusions often present in the cytoplasm of rat Sertoli cells was measured directly from semi-thin sections of perfusion-fixed testicular tissues using an image analyser linked to a light microscope. Sertoli cell nuclei were used as a reference for comparing any variations in the measured parameters of lipid inclusions during the rat spermatogenic cycle. Volume density of Sertoli cell lipid inclusions was assessed by morphometric analysis of Sertoli cells photographically reconstructed from electron micrographs. Maximum lipid content in Sertoli cells occurred during stages IX–XIV of the spermatogenic cycle, then declined at stages I–III and remained low from stages IV–VIII. The persistence and increase in number of many large Sertoli cell lipid inclusions beyond the stage where spermatid residual bodies are phagocytosed within the Sertoli cells (stage IX) suggests that the synthesis and lipolysis of Sertoli cell lipid inclusions represents an intrinsic functional cycle of the Sertoli cells. Stage-dependent variations in the lipid content of rat Sertoli cells offers morphological evidence that the metabolic duties of the Sertoli cells are synchronised with the spermatogenic cycle to provide local coordination of the proliferation and maturation of the germ cells.
