Regioselective cyclisation Reactions of Acyl-Substituted Epoxy-phosphonates with 2-Mercaptoazoles; Syntheses of Thiazolo[3,2-a]benzimidazole, Imidazo[2.1-b]thiazole, and Thiazolo [3,2-b] [1,2,4]Triazole DerivativesReaction of the acyl-substituted epoxyphosphonates 1 and 9 with the 2-mercaptoazoles 2, 13, and 16 initially yields α-substituted enones, arising from regioselective oxirane opening and subsequent dehydration. These were shown to exist mainly (as 3 and 17 from 1) or exclusively (as 10, 20, and 22 from 9) as cyclic tautomers. The subsequent cyclisation step is clearly depending on educts and reaction conditions: Generally high yields of the acyl-substituted 2,3-dihydrothiazolo[3,2-a]benzimidazoles 5 (from 1) and 11 (from 9), and of the analogous imidazo[2,1-b]thiazoles 18 and 24, respectively, are obtained, if the reactions with 2 and 16 are performed in boiling ethanol. - On treating (2,3-epoxyalkyl)phosphonates 1 (with small R3) with the mercaptoazoles 2 and 13 (but not with 16) under acetal-forming conditions, cyclisation with participation of the carbonyl group was observed, giving the ethoxy-substituted alkylidene derivatives 4 and 14. These were converted in situ into the isomeric [2-ethoxy-2-hetaryl)ethyl]phosphonates 8 and 15, respectively, due to a concomitant allylic rearrangement. The ethoxy derivatives 12 and 23, prepared from 9, were found to be stable under the same conditions. - The [(thiazolo[3,2-a]benzimidazolyl)vinyl]phosphonates 6 and the analogous thiazolo[3,2-b][1,2,4]triazoles 19 could be obtained either directly from the (2,3-epoxyalkyl)phosphonates 1 (by acid-catalyzed allylic rearrangement/dehydration of 3B), or via the ethoxy derivatives 4/8 and 14/15, respectively, or by base treatment of trimethylsilyloxy compounds like 7a. - The structures of all hetarylphosphonates were assigned from their 1H- and 13C-NMR spectra.

Die acylsubstituierten Epoxyphosphonate 1 und 9 reagieren mit den Mercaptoazolen 2, 13 und 16 unter regioselektiver Oxiran-Öffnung und Dehydratisierung primär zu α-substituierten Enonen, die überwiegend - wie 3 und 17 aus 1 - oder ausschließlich - wie 10, 20 und 22 aus 9 - als cyclische Tautomere vorliegen. Der nachfolgende Cyclisierungsschritt hängt von den Edukten und den Reaktionsbedingungen ab: In siedendem Ethanol führen die Umsetzungen mit 2 und 16 zu den acylsubstituierten 2,3-Dihydrothiazolo[3,2-a]benzimidazolen 5 und 11 bzw. den entsprechenden Imidazo[2,1-b]thiazolen 18 und 24. - Unter Ketalisierungsbedingungen können Epoxyphosphonate 1 mit kleinem R3 mit 2 und 13, jedoch nicht mit 16, unter Beteiligung des Carbonyls zu den Ethoxy-Derivaten 4 bzw. 14 cyclisiert werden, die schon während der Reaktion eine konkurrierende Allylumlagerung zu den [(2-Ethoxy-2-hetaryl)ethyl]phosphonaten 8 bzw. 15 erleiden. Die aus 9 zugänglichen Ethoxyverbindungen 12 und 23 lagern unter gleichen Bedingungen nicht um. - [(Thiazolo[3,2-a]benzimidazolyl)vinyl]phosphonate 6 und die analogen Thiazolo[3,2-b][1,2,4]triazole 19 können entweder direkt aus 1 (via sauer katalysierte Allylumlagerung/Dehydratisierung von 3B) oder über die isomeren Ethoxyverbindungen 4/8 bzw. 14/15, oder auch durch Basenbehandlung von (Trimethylsilyloxy)-Verbindungen wie 7a erhalten werden. - Die Strukturen der durchwegs neuen Hetarylphosphonsäureester wurden aus 1H- und 13C-NMR-Spektren abgeleitet.
