    Notes: Interspecific hybrids and backcross generations between the wild perennial species Helianthus resinosus, Helianthus paucifiorus, Helianthus laevigatus, Helianthus nuttallii ssp. nuttallii T. & G. and Helianthus giganteus, resistant to broomrape (Orobanche cernua) and susceptible inbred lines were obtained to study crossability to cultivated sunflower and the transmission and expression of resistance to this parasitic weed. Conventional crosses with all the species tested were successful except for the crosses with diploid H. giganteus, for which embryo rescue techniques were needed to overcome hybrid incompatibility. Pollen viability and seed set were highest for F1 hybrids with hexaploid species and lowest for those with the diploid H. giganteus. We evaluated F1, BC1F1, some BC2F1 plants and the wild and cultivated parents. The wild species and interspecific hybrids were resistant to broomrape infection except for H. nuttallii, which showed segregation, indicating that the resistance is dominant. The crossability and resistance of F1, and back-cross generations of species with different ploidy levels indicate that the transfer of broomrape resistance to cultivated sunflower is feasible.

    Notes: Resistance to the prevailing races of sunflower rust, Puccinia hehanthi Schw., is lacking in the commercial hybrids (Helianthus annuus L.). The objective of this study was to identify new sources of resistance to the four North American rust races in wild Helianthus species, and to determine their mode of inheritance. Seventy-eight accessions of H. annuus L., H. argophyllus Torrey and Gray, and H. petiolans Nutt. were evaluated in the greenhouse. Resistance to races 1, 2, 3, and 4 was observed in 25, 28, 15, and 26% of the plants, respectively, and 10% of the plants were resistant to all four races. Seven accessions that had a high percentage of resistant plants to all the four races were selected and one resistant plant from each accession was crossed with susceptible inbred line HA89. Three to four F1 plants resistant to all four races from each cross were backcrossed with HA89. F1 plants from PI-413118 × HA89 and PI 413175 × HA89 were resistant to all four races. The PI 413023 × HA89 F1 plants were 100 % resistant to races 3 and 4 and segregated in a 3: 1 resistant (R) to susceptible (S) ratio to races 1 and 2. The other four F1 combinations segregated 3R: IS ratios to all four races. Bc1F1 progenies revealed that plants from PI 413048, PI 413037, PI 413038, and PI 413171 used in the crosses possessed two dominant genes in heterozygous condition for resistance to each of the four races, whereas plants from PI 413023 possessed two dominant genes in heterozygous condition for resistance to each of races 1 and 2, and one dominant resistance gene in homozygous condition for each of races 3 and 4. Plants from PI 413118 and PI 413175 carried a single dominant gene in homozygous condition for resistance against each of the four races.

    Notes: Sunflower downy mildew (SDM) caused by Plasmopara halstedii, is a major disease of sunflower. Eleven resistance genes have been identified, but allelic relationships among these genes are not clear. This study examined the inheritance and allelic relationships of genes conferring resistance to SDM races 1, 2 and 3 (virulence phenotypes 100, 300 and 700, respectively) and confirmed a twelfth resistance gene. Three USDA Plant Introductions, AMES 3235, PI 497250, and PI 497938, and three released lines, RHA 266, RHA 274 and DM-2 were studied. RHA 266 has only the Pl1 gene for race 1 resistance. Digenic inheritance of resistance was found in AMES 3235, PI 497250, and RHA 274. These lines have the Pl1 and Pl12 genes, conferring resistance to race 1, and the Pl2 and Pl11 genes, conferring resistance to race 2. DM-2 and PI 497938 have Pl12 (but not Pl1 for resistance to race 1, the Pl12 gene (but not the Pl2) for resistance to race 2, and Pl5 for resistance to race 3. These resistance genes will serve as a foundation for future gene designations and genetic diversity studies of resistance to SDM.

    Notes: Chinese leymus [Leymus chinensis (Trin.) Tzvel.] is an important forage distributed in East Asia. The seed-set rates and the pollen–stigma compatibility in six populations were investigated in 2001. Proportionately seed-set ranged from 0·065 to 0·567 under open pollination and 0·0056 to 0·0426 under self-pollination. The former is significantly higher than the latter in each population. Microscopic observations showed that proportionately only 0·0551 to 0·1167 of self-pollinated pollen grains were compatible but most cross-pollinated pollen grains were compatible. The tubes of most incompatible pollen grains aborted upon entering into the stigmas. Among the six populations, there was a significant correlation between seed-set under open pollination and the compatible pollen rates under cross-pollination. These results suggest that Chinese leymus is a self-incompatible species, and the compatibility of pollen and stigma might be one of the factors influencing seed-set in natural conditions. This information will be useful for future breeding efforts.

    Notes: Abstract  An RFLP linkage map was constructed for cultivated sunflower Helianthus annuus L., based on 271 loci detected by 232 cDNA probes. Ninety-three F2 plants of a cross between inbred lines RHA 271 and HA 234 were used as the mapping population. These genetic markers plus a fertility restoration gene, Rf 1, defined 20 linkage groups, covering 1164 cM of the sunflower genome. Of the 71 loci 202 had codominant genotypic segregation, with the rest showing dominant segregation. Thirty-two of the 232 probes gave multiple locus segregation. There were 39 clusters of tightly linked markers with 0 cM distance among loci. This map has an average marker-to-marker distance of 4.6 cM, with 11 markerless regions exceeding 20 cM.

    Notes: Summary The zymogram phenotypes of glucose-phosphate isomerase (GPI), alcohol dehydrogenase-1 (ADH-1), glutamate oxaloacetate transaminase (GOT), superoxide dismutase (SOD), lipoxygenase (LPX), esterase (EST) and the banding patterns of gliadin and glutenin seed storage proteins were determined for Triticum aestivum cv. ‘Chinese Spring’ (CS), Dasypyrum villosum, the octoploid amphiploid T. aestivum cv. ‘Chinese Spring’ D. villosum (CS × v) (2n=8x=56; AABBDDVV), and for five CS-D. villosum disomic addition lines. The genes Gpi-V1, Adh-V1, Got-V2, and Sod-V2 coding for GPI-1, ADH-1, GOT-2, and SOD-2 isozymes were located in D. villosum on chromosome 1V, 4V, 6V, and 7V, respectively. Genes coding for gliadin- and glutenin-like subunits are located in D. villosum chromosomes 1V. There are no direct evidence for chromosomal location of genes coding for GOT-3, EST-1 and LPX-2 isozymes. The linkage between genes coding for glutenin-like proteins and GPI-1 isozymes in chromosome 1V is evidence of homoeology between chromosome 1V and the chromosomes of homoeologous group 1 in wheat.

    Notes: Summary The biochemical complexity and its consequence has been investigated in the amphiploids M x v and CS x v derived from crossing the tetraploid wheat Triticum turgidum var durum cv ‘Modoc’ and the hexaploid wheat T. aestivum cv ‘Chinese Spring’, respectively, with Dasypyrum villosum. Electrophoretic analysis of variation in six enzyme systems (GOT, ADH, GPI, SOD, EST, and LPX) and in high molecular weight glutenin seed storage proteins indicated that in the amphiploids these proteins were specified by a minimum of seven sets of homologous genes on wheat and D. villosum chromosomes and that in each set there were allelic differences. The enzymes detected in each amphiploid were fully accounted for by simple additivity of protomers specified by the homologous genes inherited from their parents. The amphiploids also expressed novel oligomeric enzymes not produced in either one of their parents. The ascertained expression for all the alleles inherited by both parents and the resulting biochemical complexity suggested that some peculiar feature of the amphiploids such as high nitrogen content in the plant and in the kernels and their immunity to the powdery mildew disease caused by both Erysiphe graminis f.sp. tritici and E. graminis f. sp. haynaldiae may be the consequence of the indicated complexity but specified by other sets of genes. The biochemical complexity of the M x v amphiploid may be the basis for its versatility as new crop species.
