    Notes: Abstract: The influence of various carbon and nitrogen sources on fusarin C synthesis was examined in submerged cultures of Fusarium moniliforme NRRL 13616. Using a zinc-deficient, synthetic medium, highest levels of fusarin C were produced by cultures grown with urea or ammonium sulfate as the nitrogen source and fructose, sucrose, or glucose as the carbon source. In media supplemented with various concentrations of glucose and ammonium sulfate, glucose concentrations which provided excess carbohydrate significantly increased fusarin C synthesis, regardless of the ammonium sulfate concentration.

    Notes: Abstract A fusarin C-producing Fusarium moniliforme strain was grown in submerged culture with defined media containing differing amounts of glucose (30 g/l or 90 g/l) and zinc (5 ppb or 3200 ppb). The influence of zinc on fusarin C synthesis and lipid composition was dependent on the initial glucose concentration. In cultures supplied with 30 g/l glucose, zinc inhibited fusarin C and lipid synthesis by diverting common substrates to ethanol synthesis. Zinc-supplemented cultures with 90 g/l glucose had ample carbon substrate to produce both ethanol and fusarin C. More total lipid with a higher unsaturated fatty acid content (more oleic acid and less stearic acid) was found in F. moniliforme biomass produced in zinc-deficient media.

    Notes: Abstract The entomopathogenic Hyphomycete Paecilomyces fumosoroseus was grown in five different liquid media, which have been developed for mass production of Beauveria spp. or P. fumosoroseus. Production was followed for 96 h by measuring both biomass and concentration of propagules. Maximal biomass was obtained with two media, Jackson and Catroux media (40–60 mg ml-1 suspension produced after 42 h incubation), where the exponential phase of growth began earlier than in the other media. While high concentrations of propagules (1.4–5.5 × 108 propagules ml-1) were produced in three media (Jackson, Paris, and Catroux media) after 48–72 h incubation, production of propagules was lower in the two other media, containing maltose as carbon source (Goral and Kondryatiev media) with 0.4–3.7 × 107 propagules ml-1 after 96 h incubation. P. fumosoroseus produced oblong blastospores in the three most productive media and conidiospore-like (ovoid to subspherical) propagules in the two other media. Infection potential of produced propagules was tested on the silverleaf whitefly ( Bemisia argentifolii). Whiteflies were sprayed as 2nd instars with P. fumosoroseus propagules produced in the five liquid media (1.9 × 103 and 1.9 × 104 propagules cm-2). All the media produced propagules that were infectious for larvae. With the lower dosage, mortality rates were significantly lower with propagules produced in one of the two least productive media (57%) (in the Kondryatiev medium) compared to those obtained with the three most productive media (〉90%). However, when whiteflies were treated with the higher dosage, mortality rates (91–99%) between media were not significantly different.

    Notes: Summary Pseudomonas fluorescens strain 2-79 (NRRL B-15 132) is a biocontrol agent of take-all of wheat caused by the fungus Gaeumannomyces graminis var. tritici. Strain 2-79 produces the antibiotic phenazine 1-carboxylic acid, which acts as the primary mechanism of disease suppression. As a first step toward designing efficient methods of mass producing and formulating this biocontrol agent, the regulation of growth and antibiotic production by growth factors (including purines, pyrimidines, vitamins) and minerals (supplying B3+, Ca2+, Co2+, Cu2+, Fe2+, Mg2+, Mn2+, Mo6+, Zn2+) was examined in defined liquid culture. Additions of boric acid and sulfates of iron and magnesium enhanced both cell and associated antibiotic accumulation. However, accumulation of the antibiotic alone improved with additions of zinc sulfate, ammonium molybdate, and cytosine, but worsened with addition of adenine. Interactive effects involving the sulfates of iron, magnesium, and zinc were observed, and optimal iron-magnesium and iron-zinc combinations were demonstrated with respect to biomass and antibiotic accumulation, respectively.

    Notes: Abstract Optimization of O2 delivery was the key to successful conidiation of Colletotrichum truncatum in submerged fermentor cultures supplied with 20 g carbon/l and C:N at the optimal 10:1 mass ratio for spore efficacy. Minimal mycelial fragmentation and maximal biomass and spore yields were provided by an O2 transfer program that called for gradual increases in stirring rate to compensate for rising cell concentration and viscosity. The utility of an event-based O2 transfer program was further supported by our observation of different O2 requirements for each phase of the life cycle. Spore germination did not occur in cultures sparged with N2. However, even low levels of O2 [10% dissolved O2 tension (DOT)] allowed 100% germination. The specific growth rate of the mycelia was a Monod-like function of DOT. The maximal growth rate was achieved when ≽15% DOT was provided via O2 transfer at a specific rate of 5.4 × 10−3 mol/g per hour. Sporulation had a strict O2 requirement, and its rate and yield were optimized by providing 55% DOT following the cessation of growth. The specific O2 demand of optimally sporulating mycelia was 4.9 × 10−4 mol/g per hour, an order of magnitude less than that associated with growing mycelia. Behaving as a pseudoplastic fluid, the fermentation broth reached a maximum apparent viscosity of 70 P at the onset of sporulation when the O2 demand was low. However, the maximum power requirement approx. 7.9 W/l occurred during the last 36 h of growth when the O2 demand was highest.

    Notes: Summary Submerged culture experiments were conducted to determine the optimal nitrogen source for rapidly producing conidia of the bioherbicide,Colletotrichum truncatum. Germination ofC. truncatum conidial inocula in submerged culture occurred most rapidly (〉95% in 6 h) in media provided with a complete complement of amino acids. When (NH4)2SO4, urea, or individual amino acids were provided as the sole nitrogen source, conidial germination was less than 20% after 6 h incubation. Conidia production was delayed inC. truncatum cultures grown in media with urea or individual amino acids as nitrogen sources compared to cultures supplied with Casamino acids or complete synthetic amino acid nitrogen sources. The use of methionine, lysine, tryptophan, isoleucine, leucine or cysteine as a sole nitrogen source severely inhibitedC. truncatum conidia production. Media with synthetic amino acid mixtures less these inhibitory amino acids produced significantly higher conidia yields compared to media with amino acid mixtures containing these amino acids. When various amounts of each individual inhibitory amino acid were added to media which contained amino acid mixtures, cysteine and methionine were shown to be most effective in reducing conidiation. An optimal nitrogen source forC. truncatum conidiation in submerged culture should contain a complete mixture of amino acids with low levels of cysteine, methionine, leucine, isoleucine, lysine and tryptophan for rapid conidiation and optimal conidia yield.
