    Description: The widespread reorganization of cellular architecture in mitosis is achieved through extensive protein phosphorylation, driven by the coordinated activation of a mitotic kinase network and repression of counteracting phosphatases. Phosphatase activity must subsequently be restored to promote mitotic exit. Although Cdc14 phosphatase drives this reversal in budding yeast, protein phosphatase 1 (PP1) and protein phosphatase 2A (PP2A) activities have each been independently linked to mitotic exit control in other eukaryotes. Here we describe a mitotic phosphatase relay in which PP1 reactivation is required for the reactivation of both PP2A-B55 and PP2A-B56 to coordinate mitotic progression and exit in fission yeast. The staged recruitment of PP1 (the Dis2 isoform) to the regulatory subunits of the PP2A-B55 and PP2A-B56 (B55 also known as Pab1; B56 also known as Par1) holoenzymes sequentially activates each phosphatase. The pathway is blocked in early mitosis because the Cdk1-cyclin B kinase (Cdk1 also known as Cdc2) inhibits PP1 activity, but declining cyclin B levels later in mitosis permit PP1 to auto-reactivate. PP1 first reactivates PP2A-B55; this enables PP2A-B55 in turn to promote the reactivation of PP2A-B56 by dephosphorylating a PP1-docking site in PP2A-B56, thereby promoting the recruitment of PP1.     Notes: Abstract— Observations have been made of the threshold fatigue properties of an Al-Mg-Si casting alloy. First, it has been shown that there exists a critical value of the stress ratio, R, below which the threshold stress intensity factor decreases with increasing R but above which it is constant. Second, measurements of the cyclic crack opening displacement have been used to examine crack closure effects and to deduce the effective cyclic stress intensity factor. The two sets of data are compared with existing models for threshold phenomena.

    Notes: Abstract Experiments have been carried out to investigate whether reducing the solution heat-treatment time of Al-7Si-0.6Mg castings from the currently recommended values adversely affects their fatigue properties. Fatigue endurance tests have been carried out in zero-tension (R=0) and measurements made of the casting defects that initiated the fatigue cracks. The work has been limited to stresses that produce a fatigue life of ~105 cycles and to two solution–treatment times (8 and 4 h). Two statistical techniques have been applied to the fatigue life data and no effect of solution heat-treatment time was detected at a confidence level of better than 95%. Similarly, no effect of cyclic test frequency could be detected for tests carried out at 1 and 60 Hz. The conclusions are confirmed by an analysis of the relation between fatigue life and the size of the casting defects that initiated fatigue failure. The scatter in fatigue lives is related to the scatter in the sizes of casting defects in the specimens. It is clear that there is a potential for considerable savings in heat-treatment costs for castings of the size and shape chosen for the study.

    Notes: Abstract— The fatigue properties of un-notched polished specimens of an aluminium casting alloy have been measured for various heat-treatment conditions and at various mean stresses. The relation between fatigue life and alternating stress is insensitive to heat-treatment and, apparently, to mean stress. It was observed that failure initiated at interdendritic shrinkage defects: evidence of classical crack initiation from persistent slip bands was also seen but such cracks, being less severe than the casting defects, never caused failure. A fracture mechanics analysis for the growth of fatigue cracks from the pores is described. It shows that the fatigue life can be quantitatively predicted from a knowledge of the size of casting defects: in particular it explains the lack of effect of heat-treatment and the apparent absence of a mean stress effect is shown to be caused by the variation in size of maximum defect present among the specimens tested. It is shown that reducing the size of shrinkage defects will increase the life, but only up to the stage at which initiation from persistent slip bands becomes operative.

    Notes: Abstract The intracellular pH of resting and stimulated muscle was monitored by two independent methods: measurement of pH iniodacetate-treated homogenates of freezeclamped tissue and the absorbance at 550–443 nm of intracellular neutral red dye in vivo. During tetanic stimulation, muscle of phosphorylase kinase-deficient mice shows a transient alkalinization whereas muscle in normal mice becomes more acid under similar conditions. The alkalinization appears to be caused by abnormally rapid AMP deamination associated with adaptation to phosphorylase kinase deficiency.
