    Notes: Summary Recombinant DNA technology was used to insert a fetal liver genomic library ApaI fragment encoding for human erythropoietin (Epo) into Bowes melanoma cells. The cells expressed the erythropoietin gene, and Epo was secreted into the culture medium together with the normally-secreted tissue plasminogen activator. Attempts to grow the cells in glass spinners in Dulbecco's medium supplemented with fetal bovine serum produced cell aggregates growing in suspension. When calcium-free suspension culture media (Joklik, DME-S, McCoy 5A-S) were used, single cell suspension cultures were obtained and high Epo production observed. When attempts were made to scale up the small glass spinners, poor growth or Epo production occurred unless the vessels were aerated. This was shown to be because of the drop in pH, possibly due to CO2 accumulation, rather than due to oxygen depletion. It was shown that a semi-continuous operation could be achieved in aerated 8-1 spinners fitted with either a conventional stirrer or a vibromixer agitator. The system was scaled up to a 100-1 stainless steel vessel fitted with a vibromixer agitator. This system was operated for over 4 months with weekly harvests producing over 100 million units of Epo in about 1000 1 of culture fluid. Interference by the serum proteins with downstream purification of the hormone from the culture fluid made the use of serum-free media highly desirable. Studies showed that the Epo was produced in serum-free systems containing peptones.

    Notes: Abstract In chemical separation processes such as supercritical extraction the use of an entrainer cosolvent can dramatically improve selectivity and yield. Ideally, in an extraction process, an entrainer cosolvent should complex with only the desired solute, pulling it from the feed. But not all cosolvents are entrainers, and a cosolvent that is effective in one application may not be effective in others. Often, competing hydrogen bonding interactions limit the effectiveness of an entrainer cosolvent. In this paper FTIR spectroscopy is used to study hydrogen bonding competition in solute/solvent/entrainer cosolvent mixtures. The extent of hydrogen bonding is determined from analysis of hydrogen-bonded and non-hydrogen-bonded infrared absorption peaks. Since these peaks overlap, curvefitting and Fourier self-deconvolution techniques are used to resolve them. Concentrations of monomeric and hydrogen-bonded species are modeled using the associated perturbed anisotropic chain theory (APACT). Using APACT it is shown that the equilibrium constant, derived from activities, can be written as the product of a temperature-dependent term and the ratio of concentrations: K=(RT) vIIC i vi . This gives a statistical mechanical basis for the empirical observation that for hydrogen-bonding equilibria, the ratio of concentrations is approximately equal to the ratio of activities.

    Notes: The prediction of polymer/polymer miscibility is addressed using analog calorimetry and molecular modeling. For each polymer, an analog compound representing one or two repeat units was chosen. Heat-of-mixing was measured for liquid mixtures of analog compounds and then used in a binary interaction model to predict polymer miscibility. Specifically, we have measured exothermic heats-of-mixing for 4-ethyl phenol, an analog of poly(vinly phenol), with several analogs containing ether, ester, or ketone functional groups. The exothermic heat-of-mixing results are consistent with the observed miscibility of poly(vinyl phenol) with polymers containing these functional groups. Using interaction parametes derived from the analog calorimetry in the binary interaction model or using premixes of 4-ethyl phenol in ethyl benzene, we correctly predict the magnitude and relative order of the fraction of vinyl phenol units in copolymers with styrene required for miscibility with poly(methyl methacrylate), polyacetal, and a polyketone. the miscibility trends for poly(vinyl phenol) blends predicted from analog calorimetry and the binary interaction model are in reasonable agreement with those predicted from the association model of Painter and Coleman, despite the different bases of the two approaches. We have used molecular modeling to complement the analog calorimetry and to assess steric effects on hydrogen-bonding ability for models of poly(n-butyl acrylate) and poly(t-butyl acrylate) with phenol. The modeling results suggest that, in some cases, steric effects and the three-dimensional structure of the polymer can significantly influence the hydrogen-bonding ability of polymers relative to their analogs.

    Description: Seven years of measurements from the Polar spacecraft are surveyed to monitor the variations of plasma density within the magnetospheric cusps. The spacecraft's orbital precession from 1998 through 2005 allows for coverage of both the northern and southern cusps from low altitude out to the magnetopause. In the mid- and high- altitude cusp, plasma density scales well with the solar wind density ( n c u s p / n s w ∼0.8). This trend is fairly steady for radial distances greater then 4 R E . At low altitudes ( r 〈4 R E ) the density increases with decreasing altitude and even exceeds the solar wind density due to contributions from the ionosphere. The density of high charge-state Oxygen (O 〉+2 ) also displays a positive trend with solar wind density within the cusp. A multifluid simulation with the BATSRUS MHD model was run to monitor the relative contributions of the ionosphere and solar wind plasma within the cusp. The simulation provides similar results to the statistical measurements from Polar and confirms the presence of ionospheric plasma at low altitudes.

    Description: The reward generated by social interactions is critical for promoting prosocial behaviors. Here we present evidence that oxytocin (OXT) release in the ventral tegmental area (VTA), a key node of the brain’s reward circuitry, is necessary to elicit social reward. During social interactions, activity in paraventricular nucleus (PVN) OXT neurons increased. Direct activation of these neurons in the PVN or their terminals in the VTA enhanced prosocial behaviors. Conversely, inhibition of PVN OXT axon terminals in the VTA decreased social interactions. OXT increased excitatory drive onto reward-specific VTA dopamine (DA) neurons. These results demonstrate that OXT promotes prosocial behavior through direct effects on VTA DA neurons, thus providing mechanistic insight into how social interactions can generate rewarding experiences.
