    Description: The formation of branched lariat RNA is an evolutionarily conserved feature of splicing reactions for both group II and spliceosomal introns. The lariat is important for the fidelity of 5' splice-site selection and consists of a 2'-5' phosphodiester bond between a bulged adenosine and the 5' end of the intron. To gain insight into this ubiquitous intramolecular linkage, we determined the crystal structure of a eukaryotic group IIB intron in the lariat form at 3.7 A. This revealed that two tandem tetraloop-receptor interactions, eta-eta' and pi-pi', place domain VI in the core to position the lariat bond in the post-catalytic state. On the basis of structural and biochemical data, we propose that pi-pi' is a dynamic interaction that mediates the transition between the two steps of splicing, with eta-eta' serving an ancillary role. The structure also reveals a four-magnesium-ion cluster involved in both catalysis and positioning of the 5' end.     Description: Ancient components of the ribosome, inferred from a consensus of previous work, were constructed in silico , in vitro and in vivo . The resulting model of the ancestral ribosome presented here incorporates ~20% of the extant 23S rRNA and fragments of five ribosomal proteins. We test hypotheses that ancestral rRNA can: (i) assume canonical 23S rRNA-like secondary structure, (ii) assume canonical tertiary structure and (iii) form native complexes with ribosomal protein fragments. Footprinting experiments support formation of predicted secondary and tertiary structure. Gel shift, spectroscopic and yeast three-hybrid assays show specific interactions between ancestral rRNA and ribosomal protein fragments, independent of other, more recent, components of the ribosome. This robustness suggests that the catalytic core of the ribosome is an ancient construct that has survived billions of years of evolution without major changes in structure. Collectively, the data here support a model in which ancestors of the large and small subunits originated and evolved independently of each other, with autonomous functionalities.

    Description: Ship tracks are regarded as the most obvious manifestations of the effect of anthropogenic aerosol particles on clouds (indirect effect). However, it is not yet fully quantified whether there are climatically relevant effects on large scales beyond the narrow ship tracks visible in selected satellite images. A combination of satellite and reanalysis data is used here to analyze regions in which major shipping lanes cut through otherwise pristine marine environments in subtropical and tropical oceans. We expect the region downwind of a shipping lane is affected by the aerosol produced by shipping emissions but not the one upwind. Thus, differences in microphysical and macrophysical cloud properties are analyzed statistically. We investigate microphysical and macrophysical cloud properties as well as the aerosol optical depth and its fine-mode fraction for the years 2005–2007 as provided for by retrievals of the two Moderate Resolution Imaging Spectroradiometer instruments. Water-cloud properties include cloud optical depth, cloud droplet effective radius, cloud top temperature, and cloud top pressure. Large-scale meteorological parameters are taken from ERA-Interim reanalysis data and microwave remote sensing (sea surface temperature). We analyze the regions of interest in a Eulerian and Lagrangian sense, i.e., sampling along shipping lanes and sampling along wind trajectories, respectively. No statistically significant impacts of shipping emissions on large-scale cloud fields could be found in any of the selected regions close to major shipping lanes. In conclusion, the net indirect effects of aerosols from ship emissions are not large enough to be distinguishable from the natural dynamics controlling cloud presence and formation.

    Description: Representing the electrodynamics of relativistically drifting particle ensembles in discrete, co-propagating Galilean coordinates enables the derivation of a Particle-In-Cell algorithm that is intrinsically free of the numerical Cherenkov instability for plasmas flowing at a uniform velocity.
