    Description: The repair of toxic double-strand breaks (DSB) is critical for the maintenance of genome integrity. The major mechanisms that cope with DSB are: homologous recombination (HR) and classical or alternative nonhomologous end joining (C-NHEJ versus A-EJ). Because these pathways compete for the repair of DSB, the choice of the appropriate repair pathway is pivotal. Among the mechanisms that influence this choice, deoxyribonucleic acid (DNA) end resection plays a critical role by driving cells to HR, while accurate C-NHEJ is suppressed. Furthermore, end resection promotes error-prone A-EJ. Increasing evidence define Poly(ADP-ribose) polymerase 3 (PARP3, also known as ARTD3) as an important player in cellular response to DSB. In this work, we reveal a specific feature of PARP3 that together with Ku80 limits DNA end resection and thereby helps in making the choice between HR and NHEJ pathways. PARP3 interacts with and PARylates Ku70/Ku80. The depletion of PARP3 impairs the recruitment of YFP-Ku80 to laser-induced DNA damage sites and induces an imbalance between BRCA1 and 53BP1. Both events result in compromised accurate C-NHEJ and a concomitant increase in DNA end resection. Nevertheless, HR is significantly reduced upon PARP3 silencing while the enhanced end resection causes mutagenic deletions during A-EJ. As a result, the absence of PARP3 confers hypersensitivity to anti-tumoral drugs generating DSB.

    Description: Organisms must process information encoded via developmental and environmental signals to survive and reproduce. Researchers have also engineered synthetic genetic logic to realize simpler, independent control of biological processes. We developed a three-terminal device architecture, termed the transcriptor, that uses bacteriophage serine integrases to control the flow of RNA polymerase along DNA. Integrase-mediated inversion or deletion of DNA encoding transcription terminators or a promoter modulates transcription rates. We realized permanent amplifying AND, NAND, OR, XOR, NOR, and XNOR gates actuated across common control signal ranges and sequential logic supporting autonomous cell-cell communication of DNA encoding distinct logic-gate states.     Description: 〈p〉Communication between the endoplasmic reticulum (ER) and mitochondria plays a pivotal role in Ca〈sup〉2+〈/sup〉 signaling, energy metabolism, and cell survival. Dysfunction in this cross-talk leads to metabolic and neurodegenerative diseases. Wolfram syndrome is a fatal neurodegenerative disease caused by mutations in the ER-resident protein WFS1. Here, we showed that WFS1 formed a complex with neuronal calcium sensor 1 (NCS1) and inositol 1,4,5-trisphosphate receptor (IP〈sub〉3〈/sub〉R) to promote Ca〈sup〉2+〈/sup〉 transfer between the ER and mitochondria. In addition, we found that NCS1 abundance was reduced in WFS1-null patient fibroblasts, which showed reduced ER-mitochondria interactions and Ca〈sup〉2+〈/sup〉 exchange. Moreover, in WFS1-deficient cells, NCS1 overexpression not only restored ER-mitochondria interactions and Ca〈sup〉2+〈/sup〉 transfer but also rescued mitochondrial dysfunction. Our results describe a key role of NCS1 in ER-mitochondria cross-talk, uncover a pathogenic mechanism for Wolfram syndrome, and potentially reveal insights into the pathogenesis of other neurodegenerative diseases.〈/p〉

    Description: Dinoflagellate cyst (dinocyst) records from the southern Nordic Seas were compiled in order to evaluate the evolution of upper ocean conditions, on a millennial timescale and supported by a highly resolved record from the Vøring Plateau. After the transitional phase from the last deglaciation, three main phases define the Holocene. The early Holocene (〉7.5 ka BP) features important numbers of cool-temperate species dominated by Nematosphaeropsis labyrinthus and Impagidinium pallidum in the west. The assemblage composition changes drastically at the transition from the early to the mid-Holocene, from when on Operculodinium centrocarpum dominates. The changeover is dated between ~6.1 and 7.5 ka BP, perhaps earlier closer to the Iceland–Scotland Ridge, and appears to be linked to the onset of a modern type of surface circulation. ‘Warmest’ assemblages occur at the Vøring Plateau shortly after the transition, when Atlantic waters also appear to have spread farthest westward. The recurrence of colder elements can be linked to cooling from ~2.4 ka BP at the Vøring Plateau and presumably earlier in the west but is difficult to date there because of the low sedimentation rates. This is a general issue in many areas of the Nordic Seas and appears to have an important effect on cyst concentrations and assemblage composition, with the possible loss of oxygenation-sensitive cysts in the older parts of the cores. Comparing dinocyst-based quantitative reconstructions with those retrieved from other plankton reveals a significantly different trend between proxies, linked to a differing autecological response to seasonal changes at their respective depth habitats.

    Notes: Highly densified 0.99 SnO2-0.01 CuO-based ceramics with antimony, tantalum, or niobium oxides as additives have been prepared. Samples have been characterized by dc conductivity and complex impedance spectroscopy as a function of temperature and by current density as a function of electric field. Microstructures have been analyzed by scanning electron microscopy, electron microprobe, and Auger spectroscopy. The electrical properties of Ta- and Nb-doped ceramics are grain boundary controlled at least at low temperature. Sb-doped materials show a different electrical behavior due to the presence of a segregation layer of antimony which inhibits grain boundary effects.

    Notes: We have tested at Super ACO (LURE) a new angular resolved photoemission device for the 12–30 eV energy range, using multidetection of photoelectrons: all electrons emitted in a plane are collected and focused onto a fluorescent screen in a coordinate grid directly related to (Ei, k(parallel)). This multidetection provides a substantial diminution of the acquisition time. Ten seconds are needed to obtain a complete off-normal set of energy distribution curves (EDC). Real time visualization of the valence-band structure for one azimuthal plane is even possible. Digital image treatment is performed after the experiments, permitting an easier extraction of the characteristics of the bands and the emission structures.

    Notes: A stationary plasma thruster is experimentally studied using different optical spectroscopies of xenon ions. Doppler shift in laser induced fluorescence is used for velocity determination while the ion density is determined by emission spectroscopy. These experiments show unambiguously that the ionization and the acceleration zones are spatially distinct inside the thruster channel. Moreover, it is shown that these results can be easily taken into account with a very simple quasineutral stationary one-dimensional model. © 2002 American Institute of Physics.

    Notes: We performed a correlated study of sulfide-passivated GaAs(001) surfaces, using x-ray photoelectron spectroscopy and reflection anisotropy spectroscopy. The reflection anisotropy spectra reveal after desorption of the sulfide overlayer the presence of As and Ga dimers analogous to ones observed on As-decapped surfaces. We identify in the 3d core-level spectra the surface components due to As and Ga dimers: their chemical shifts are −0.28 and −0.35 eV, respectively. We propose an interpretation of the main surface components in the framework of a simple model, based on charge neutrality, electron counting, and electronegativity concepts. © 1996 American Institute of Physics.
