    Notes: A simulation approach to measuring supply chain performance is evaluated which incorporates order release theory. Within manufacturing a number of order release mechanisms have been developed. The importance of order release is first examined and its applicability to monitoring the performance of the supply chain is proposed. A simulation model of a typical, single channel logistics network was developed. Using the simulation model, each of the order release mechanisms was assessed and close agreement was obtained with the work of previous researchers. A new order release approach is proposed which is found to be superior to those analysed previously and should lead to improved supply chain performance.

    Notes: Presents the results of a study on the value of producing a standard type quality manual using the definition mode zero (IDEF0) format. IDEF0 was developed for the integrated computer-aided manufacturing (ICAM) program of the US Air Force. ICAM was established with the objective of improving the productivity of aerospace contractors, and IDEF0 is a graphic modelling tool for generating instructions that are clearer and easier to understand. A pilot test and a survey were conducted to gauge the response of local workers in terms of how readily they were able to understand the contents of a quality manual written in both a conventional language and the IDEF0 format. The results of the pilot test and survey are very encouraging, with around 70 per cent of the subjects reporting high efficiency, clarity and legibility of the quality manual writing using the IDEF0 format.

    Notes: ISO 9000 certification is one of the most prevalent quality assurance systems in the world. Despite its popularity, there is considerable confusion and frustration surrounding its implementation in small to medium-sized enterprises. Major obstacles to ISO 9000 registration are identified and include implementation costs, inadequate resources, and insufficient external assistance. Thus, SMEs require a more cost-effective and well-planned ISO implementation process. Based on case study material from ten companies, located in Hong Kong, the activities involved in obtaining ISO 9000 registration were identified. Project management techniques were then applied to develop a generic project network and resource loading profile for ISO 9000 implementation. Using this benchmarking approach SMEs should be able to plan the adoption of ISO 9000 more effectively with the limited resources available to them.

    Notes: Summary Spermatogenesis in the Budgerigar can be arrested by reducing the birds' photoperiod to 8 hours of daylight or less. When this occurs, Sertoli cell cytoplasm shows a great increase in the size and number of residual bodies, while the smooth endoplasmic reticulum is reduced. If the bird is kept at 8 hours of daylight for some weeks large lipid droplets are seen in Sertoli cytoplasm, and degenerated spermatids are apparently phagocytosed. The interstitium shows fewer active Leydig cells, a paucity of lipids and occasional ovoid mitochondria. The basal lamellae of the tubule which are thick and convoluted before and during spermatogenesis become thinner and straighter. It is thought that these morphological changes reflect changes in metabolic activity.

    Notes: Summary The development of the acrosome of the budgerigar spermatid was studied under the electronmicroscope. The acrosome arises from a granule derived from the Golgi apparatus which interacts with the nucleus to form a cone shaped acrosomal cap and a perforatorium. A quantity of cytoplasm and plasma membrane is captured from the cell periphery and incorporated into the substance of the acrosome. The possible significance of the developmental phases and comparative avian spermatology is discussed.

    Description: CO 2 and CH 4 exchange is strongly affected by hydrology in landscapes underlain by permafrost. Hypotheses for these effects in the model ecosys were tested by comparing modelled CO 2 and CH 4 exchange with CO 2 fluxes measured by eddy covariance from 2006 to 2009, and with CH 4 fluxes measured with surface chambers in 2008, along a topographic gradient at Daring Lake, NWT. In an upland tundra, rises in net CO 2 uptake in warmer years were constrained by declines in CO 2 influxes when vapor pressure deficits ( D ) exceeded 1.5 kPa, and by rises in CO 2 effluxes with greater active layer depth (ALD). Consequently net CO 2 uptake rose little with warming. In a lowland fen, CO 2 influxes declined less with D and CO 2 effluxes rose less with warming, so that rises in net CO 2 uptake were greater than those in the tundra. Greater declines in CO 2 influxes with warming in the tundra were modelled from greater soil-plant-atmosphere water potential gradients that developed under higher D in drained upland soil, and smaller rises in CO 2 effluxes with warming in the fen were modelled from O 2 constraints to heterotrophic and below-ground autotrophic respiration from a shallow water table in poorly drained lowland soil. CH 4 exchange modelled during July and August indicated very small influxes in the tundra, and larger effluxes characterized by afternoon emission events caused by degassing of warming soil in the fen. Emissions of CH 4 modelled from degassing during soil freezing in October – November contributed about one-third of the annual total.

    Description: Motivation: Mouse embryonic stem cells (mESCs) have developed into a prime system to study the regulation of pluripotency in stable cell lines. It is well recognized that different, established protocols for the maintenance of mESC pluripotency support morphologically and functionally different cell cultures. However, it is unclear how characteristic properties of cell colonies develop over time and how they are re-established after cell passage depending on the culture conditions. Furthermore, it appears that cell colonies have an internal structure with respect to cell size, marker expression or biomechanical properties, which is not sufficiently understood. The analysis of these phenotypic properties is essential for a comprehensive understanding of mESC development and ultimately requires a bioinformatics approach to guarantee reproducibility and high-throughput data analysis. Results: We developed an automated image analysis and colony tracking framework to obtain an objective and reproducible quantification of structural properties of cell colonies as they evolve in space and time. In particular, we established a method that quantifies changes in colony shape and (internal) motion using fluid image registration and image segmentation. The methodology also allows to robustly track motion, splitting and merging of colonies over a sequence of images. Our results provide a first quantitative assessment of temporal mESC colony formation and estimates of structural differences between colony growth under different culture conditions. Furthermore, we provide a stream-based visualization of structural features of individual colonies over time for the whole experiment, facilitating visual comprehension of differences between experimental conditions. Thus, the presented method establishes the basis for the model-based analysis of mESC colony development. It can be easily extended to integrate further functional information using fluorescence signals and differentiation markers.     Description: Changes in arctic C stocks with climate are thought to be caused by rising net primary productivity (NPP) during longer and warmer growing seasons, offset by rising heterotrophic respiration (Rh) in warmer and deeper soil active layers. In this study, we used the process model ecosys to test hypotheses for these changes with CO2 and energy fluxes measured by eddy covariance over a mesic shrub tundra at Daring Lake, Canada, under varying growing seasons. These tests corroborated substantial rises in NPP, smaller rises in Rh, and, hence, rises in net ecosystem productivity (NEP) from 17 to 45 g C m−2 yr−1 (net C sink), modeled with higher Ta and longer growing seasons. However, NEP was found to decline briefly during midsummer warming events (Ta 〉 20°C). A model run under climate change predicted for Daring Lake indicated that rises in NPP would exceed those in Rh during the first 100 years, causing NEP to rise. Rises in NPP were driven by more rapid net N mineralization from more rapid Rh in warming soils. However, greater declines in NEP were modeled during more frequent and intense midsummer warming events as climate change progressed. Consequently, average annual NEP (± interannual variability) rose from 30 (±13) g C m−2 yr−1 under current climate to 57 (±40) g C m−2 yr−1 after 90 years but declined to 44 (±51) g C m−2 yr−1 after 150 years, indicating that gains in tundra NEP under climate change may not be indefinite.
