    Notes: Abstract. The use of vaccines as a cost-effective means of controlling bacterial disease is now widely accepted in the trout and salmon farming industries. This paper quantifies the savings obtained from vaccination against enteric redmouth disease using published data for the performance of commercial vaccines under field conditions. Costings are made on the basis of prices current in January 1986. A model is derived which enables the value of vaccination to be determined at any particular site and some comparisons are made with antibiotic therapy.

    Notes: We determined the minimal size of the genomic region necessary for gas vesicle synthesis in halophilic archaebacteria by transformation experiments, comparative DNA sequence analysis and investigation of gas vesicle (Vac) mutants. The comparison of the three genomic regions encoding gas vesicles in Halobacterium halobium (p-vac- and c-vac-region) and Haloferax mediterranei (mc-vac-region) indicates high DNA sequence similarity throughout a contiguous sequence of 9kbp. In each case, this area encompassed at least 13 open reading frames (ORFs). Ten of these ORFs (gvpD to gvpM) were located 5′ to the vac gene encoding the major gas vesicle protein, but were transcribed from the opposite strand. At least two ORFs (gvpC, and gvpN) were located 3′ to each vac gene and transcribed from the same strand as the respective vac gene. In the p-vac-region present on plasmid pHH1 these ORFs were transcribed as at least three units, one transcript encompassing gvpD-gvpE, the second encompassing ORFs gvpF to gvpM, and the third unit comprising the ORFs located 3′ to the p-vac gene. In H. halobium Vac mutants copies of the insertion elements ISH2, ISH23, ISH26 or ISH27 were found to be integrated throughout the p-vac-region. The de novo synthesis of gas vesicles was tested by transformation of the Vac-negative species, Haloferax volcanii, with various subfragments of the mc-vac- or p-vac-region cloned into vector plasmids. In contrast to a fragment containing the entire 9 kbp region, none of the subfragments tested was sufficient to promote gas vesicle synthesis. However, gas vesicle synthesis could be restored in each Vac mutant containing an ISH element when the entire transcription unit encompassing the mutated gene on pHH1 was present in the wild-type form on the vector construct.

    Notes: The factors influencing the uptake of a 14C-labelled Vibrio anguillarum vaccine in direct immersion experiments with rainbow trout, Salmo gairdneri were studied. Immersion times of longer than 10 s did not increase vaccine uptake. A pre-immersion dip in a hyperosmotic solution had no effect on uptake, even at lower vaccine bath concentrations. It appeared that the head end of the fish was implicated in uptake. Vaccine uptake decreased at lower temperatures, whereas the use of an adjuvanted vaccine and a soluble vaccine preparation led to increased uptake. Larger fish took up more vaccine.

    Notes: The localization of a live, and a formalized vaccine preparation of Aeromonas salmonicida within the tissues of rainbow trout, Salmo gairdneri, was followed over a 5 day period. When presented by intraperitoneal injection, both the live bacteria and the vaccine localized in the spleen, liver, kidney and gut. When presented orally, the bacteria and the vaccine were confined almost exclusively to the gut region. Direct immersion resulted in low detectable levels within the tissues, with the spleen and kidney localizing the Aeromonas after its initial attachment to the outer surfaces of the fish.

    Notes: Several physical properties related to the surface characteristics of autoaggregating and non-autoaggregating strains of Aeromonas salmonicida have been investigated. Properties examined included resistance to the bactericidal action of trout serum, adhesion to fish leucocytes and fish cell monolayers in vitro, resistance to phagocytosis by fish leucocytes and the in vivo localization following intraperitoneal injection. For each strain the presence or absence of an extracellular protein A-layer was investigated and the pathogenicity for brook trout determined.Presumptive A-layer protein, in the form of a 49 kdal subunit, could be detected only in one of the strains examined. This strain autoaggregated and was the most resistant to serum bactericidal activity. Complement activation by the alternative pathway was thought to be responsible for this heat-labile bactericidal activity. Three strains that autoaggregated and three that did not had no detectable A-layer. Autoaggregating strains appeared more adhesive to both fish cell types but all strains were phagocytosed by fish leucocytes to a similar degree. An autoaggregating strain was localized in the spleen. The seven strains were only moderately pathogenic for brook trout, possibly as a result of the challenge system. In view of this, no property investigated could be correlated with greatly increased virulence.

    Notes: An outbreak of an acute oedematous disease characterised by orbital and abdominal swelling, and high mortality, in young turbot, was studied. The clinical and pathological picture was consistent with a circulating toxaemia principally affecting the heart, orbit and kidney. Vibrio anguillarum, virtually identical with type strain NCMB6, was isolated in pure culture from all organs of all fish. Therapy was only marginally successful using antibiotics but in a small group subjected to temperature reduction below 10°C, losses were reduced from 20 % to 5 % compared with the controls.

    Notes: One hundred kg of Rainbow trout, Salmo gairdneri, weighing 80–100g each were immersed in a formalized, bivalent vaccine of Vibrio anguiliárum (VA) and Vibrio ordalii (VO), at a1/10 dilution for 30s at 10°C. The protection levels achieved at 8 and 10 weeks post vaccination were assessed by intraperitoneal challenge with 105 live V. anguillarnm or V. ordalii. No decline in protection due to antigen depletion was found between batches throughout the procedure. The protection conferred by one component of the vaccine was slightly better than the other (80% against VO and 42 % against VA). A number of measures of immunity were monitored before and after challenge. These included serum and mucus antibody litres, bactericidal effects of serum and mucus and phagocytosis by peritoneal exudate cells. Serum antibody was present (peak litre of-log2 3.25 for VO, and 4.4 for VA) but had declined to background levels by the time of challenge, whereas bactericidal effects and phagocytosis rate of 40% were not increased by vaccination. The most likely immune mechanism responsible for the observed protection was discussed and suggested to be a cellular mechanism where serum antibody acts as an opsonin to increase phagocytosis.

    Notes: Abstract. Vibrio anguillarum was shown to adhere to excised sections of rainbow trout gut in numbers which were a function of the concentration of bacterial cells and the time for which the tissue was exposed. Using a standardized method for assaying adhered bacteria the course of infection with vibriosis in rainbow trout was studied. It is suggested that the gut may be an important site of bacterial multiplication in slowly developing infections of mature fish. Bacteria adhere to gut sections of fish protected by vaccination substantially less than to sections from non-vaccinated fish.
