    Description: The Jinshajiang suture zone, located in the eastern part of the Tethyan tectonic domain, is noticeable for a large-scale distribution of Late Jurassic to Triassic granitoids. These granitoids were genetically related to the evolution of the Paleo-Tethys Ocean. The Beiwu, Linong and Lunong granitoids occur in the middle zone of the Jinshajiang Suture Zone, and possess similar geochemical features, indicating they share a common magma source. SIMS zircon U-Pb dating reveals the Beiwu, Linong and Lunong granitic intrusions were emplaced at 233.9±1.4 Ma (2 sigma), 233.1 ±1.4 Ma (2 sigma) and 231.0±1.6 Ma (2 sigma), respectively. All of these granitoids are enriched in abundances of Si (SiO2 =65.2-73.5 wt.%), and large-ion-lithophile-elements (LILEs), but depleted in high-field-strength-elements contents (HFSEs, e.g., Nb, Ta, Ti). In addition, they have low P2O5 contents (0.06-0.11 wt.%), A/CNK values ([molecular Al2O3/(CaO+Na2O+K2O)], mostly〈1.1) and 10000Ga/Al ratios (1.7-2.2), consistent with the characteristics of I-type granites. In terms of isotopic compositions, these granitoids have high initial 87Sr/86Sr ratios (0.7078-0.7148), Pb isotopic compositions [(206Pb/204Pb)t=18.213-18.598, (207Pb/204Pb)t=15.637-15.730 and (208Pb/204Pb)t=38.323-38.791], zircon d18O values (7. per mil-9.3 per mil) and negative eNd(t) values (-5.1 to -6.7), suggesting they were predominantly derived from the continental crust. Their Nb/Ta ratios (average value=8.6) are consistent with those of the lower continental crust (LCC). However, variable ?Hf(t) values (-8.6 to +2.8) and the occurrences of mafic microgranular enclaves (MMEs) suggest that mantle-derived melts and lower crustal magmas were involved in the generation of these granitoids. Moreover, the high Pb isotopic ratios and elevated zircon d18O values of these rocks indicate a significant contribution of the upper crustal composition. We propose a model in which the Beiwu, Linong and Lunong granitoids were generated under a late collisional or post-collisional setting. It is possible that this collision was completed before Late Triassic. Decompression induced mantle-derived magmas underplated and provided the heat for the anatexis of the crust. Hybrid melts including mantle-derived and the lower crustal magmas were then generated. The hybrid melts thereafter ascended to a shallow depth and resulted in some degree of sedimentary rocks assimilation. Such three-component mixing magmas source and subsequent fractional crystallization could be responsible for the formation of the Beiwu, Linong and Lunong granitoids.

    Notes: The periplasmic Escherichia coli enzyme DsbA catalyses the efficient formation of disulphide linkages in numerous extracytoplasmic proteins. Enteropathogenic E. coli, a major cause of infantile diarrhoea worldwide, expresses a type IV fimbria known as the bundle-forming pilus that promotes adherence to tissue-culture cells. In this study, we report that transposon insertions in the dsbA locus abolish adherence and dramatically reduce the level of bundlin, the major structural subunit of the pilus encoded by the bfpA locus. Adherence and bundlin levels are restored by complementation with the cloned dsbA gene. DsbA has no effect on bfpA transcription as measured with bfpA–lacZ fusions. Replacement of either cysteine codon 129 or 179 of bfpA with a serine codon results in reduced levels of bundlin, similar to the effect of the dsbA mutation. As is the case with dsbA mutants, this decreased level of bundlin is not due to decreased transcription. The half-life of bundlin as detected by pulse-chase experiments is dramatically reduced in a dsbA mutant in comparison to the wild type. The effect of DsbA on bundlin oxidation is independent of signal-peptide processing. Thus, we demonstrate that the DsbA enzyme is critical for the biogenesis of a type IV fimbria because of the essential role of a disulphide bond in the stability of the major structural subunit. These data illuminate the early steps in the biogenesis of type IV fimbriae by demonstrating that newly synthesized prepilin is a transmembrane protein accessible to periplasmic and cytoplasmic processing enzymes.

    Notes: Enteropathogenic Escherichia coli (EPEC) adhere to epithelial cells in microcolonies, a pattern termed localized adherence (LA). LA is dependent upon the presence of 50–70MDa plasmids, termed EPEC adherence factor (EAF) plasmids. Expression of an EAF plasm id-encoded type IV fimbria, the bundle-forming pilus (BFP), is associated with the LA phenotype. TnphoA insertions in bfpA, the gene encoding the major structural subunit of the BFP, abolish LA. While bfpA::TnphoA mutants cannot be complemented for LA by plasmids carrying the bfpA gene alone in trans, this work shows that they can be complemented by plasmids carrying the bfpA gene, as well as approximately 10kb of downstream sequence, suggesting that such mutations have polar effects on downstream genes. The identification and characterization of a cluster of 13 genes immediately downstream of bfpA are described. The introduction into a laboratory Escherichia coli strain of a plasmid containing these 14 bfp gene cluster genes, along with pJPN14, a plasmid containing another fragment derived from the EAF plasmid, confers LA ability and BFP biogenesis. However, when a mutation is introduced into the last gene of the bfp cluster, neither LA nor BFP biogenesis is conferred. This work also provides evidence to show that the fragment cloned in pJPN14 encodes a factor(s) which results in increased levels of the pilin protein. Finally, it is shown that expression of the 14 genes in the bfp cluster from an IPTG-inducible promoter, in the absence of pJPN14, is sufficient to reconstitute BFP biogenesis in a laboratory E. cob strain, but is insufficient for LA. This is the first report demonstrating the reconstitution of a type IV pilus in a laboratory E. coli strain with a defined set of genes. The 8FP system should prove to be a useful model for studying the molecular mechanisms of type IV pilus biogenesis.

On the basis of analyzing disadvantages of traditionalmethods to identify distribution functions, which begin with overall fitting effect, this paper proposeda method by applying numerical simulation and fuzzy linear regression. It can be used to determinethe distribution function of crack propagation data. Through fuzzy linear regression, the result thatcrack propagation size under certain load cycles are in logarithmic normal distribution can beacquired. In the same way, the number of load cycles is also in logarithmic normal distribution whenthe crack propagates to a certain size

    Notes: A new model is proposed for the analysis of fatigue crack growth under random loading.The fatigue rule of crack length is transformed into the monotony function rule based on types of thecrack.     Notes: Grinding machining quality contains machining precision and grinding surface integrality. The factors influencing grinding machining quality have fuzzy characteristics. For example, the magnitude and measure methods of grinding surface roughness have fuzzy uncertainties. The grades of the machining quality are vague, and there is no definite boundary between “good” and “bad” machining quality. Analytical Hierarchy Process (AHP) combined with the fuzzycomprehensive evaluation technique is used to evaluate the grade condition of the quality in this paper. Considering the fuzziness of the factors, a two-stage fuzzy comprehensive evaluation model is proposed to evaluate the grinding machining quality. In light of different reliable degrees of different kinds of fuzzy operator models, the weighted average method is used. The membership degrees of the evaluation factors are determined by experts’ knowledge and experiences. The factor weights are calculated via the AHP method. Certain synthetic importance of each stage evaluation is presented through weighted sum of the relative important degree. Examples of conventional external grinding machining illustrate the effectiveness of the proposed model

    Notes: Thiolated polymers have been studied by many researchers because of the mucoadhesiveproperties of thiol group. Alginate is a natural and biocompatible polymer that has been widely usedin drug delivery. In this study, thiolated chitosan microspheres (TCMs) were prepared by ionicgelation process with tripolyphosphate and then, the bovine growth hormone (BGH) was loaded asa model drug. Finally, the BGH-loaded TCMs (BTCMs) were coated with alginate to improve thestability in gastrointestinal (GI) track. The alginate-coated BTCMs (ABTCMs) were observed asspherical shapes. The average particle sizes of ABTCMs were 6.97±0.55 -m and the sizedistributionwas shown uniformly. Release of BGH from ABTCMs was decreased by coating withalginate and increased rapidly with the change in medium pH from 1.2 to 7.4. Results indicate thatthe ABTCMs have a potential as a drug carrier for oral drug delivery
