    Notes: Three types of tobacco (Nicotiana tabacum cv. Havana 38) callus: 1) healthy stem callus, 2) TMV-infected stem callus, 3) TMV-infected leaf callus; and leaves differentiated from healthy stem callus, and from TMV-infected leaf callus were compared for fine structure. In addition, the fine structure was observed of plastids in cells of leaves differentiated from callus isolated from stem sections of TMV-infected hybrid tobacco plants (N. tabacum cv. Havana 38 ×N. glutinosa) grown under high temperature. The cytoplasmic organelles in tissue cultured cells were similar to those in cells of greenhouse-grown tobacco plants. Except for plastids, TMV infection did not noticeably affect morphologically other cellular organelles in tissue culture cells. In TMV-infected leaf callus, numerous small bodies were seen in plastid-like bodies, while vesicle-like structures were observed in the stroma of plastids in leaves differentiated from callus of hybrid tobacco inoculated with TMV. Morphological variations of mitochondria, such as swelling and vacuolization of the inner matrix, occurred frequently in TMV-infected leaf callus. Needle-like crystalline inclusions or looped inclusions composed of many fine, long filaments were considered TMV particles orientated parallel to each other. The TMV particles were detected in the cytoplasm of tissue culture cells.

    Notes: Summary Single cells of hybrid tobacco callus, isolated from germinating seeds, were grown individually in microchambers in the absence of any other cells, in fresh, liquid, coconut milk-medium. These produced a conlony of 50–75 cells in 10–15 days. In all instances the plane of the first cell division, and in most cases also that of the second division, were at right angles to the long axis of the cell. There was more variation in the size, shape and pattern of the second and subsequent divisions in single cells isolated from fresh stem pith and seed callus than those from old stem callus. Upon transfer from the microchambers to agar medium the colony of cells gave rise to a huge mass of callus tissue in about 3 months. In no instance did the cell colonies or any stages preceding them resemble or simulate any stages of normal embryogeny of tabacco. Single-cell clones obtained by this method from seed callus or fresh pith callus produced shoots with leaves and roots in synthetic medium with various indoleacetic acid-kinetin combinations. Normal plants established from the above cultures in the greenhouse flowered in due course of time. This method offers the possibility of producing a very large number of clones or identical plants in species where vegetative propagation is not otherwise possible, apart from its use in studies on the genetics and morphogenesis in higher plants.

    Notes: Summary Growth, nutrition and chlorophyll development were studied in chlorophyllous callus tissues isolated from the following edible angiospermous plants: carrot root, crown gall of tomato, endive embryo, leaf petiole and stem of lettuce, leaf petiole of parsley, pea stem and rose stem. Growth patterns of these tissues in vitro were sigmoid. Synthetic media produced less growth, in terms of fresh weight increase, than media containing coconut milk, a highly complex and little understood natural substance. Murashige and Skoog's synthetic medium proved useful for satisfactory growth and chlorophyll production in a number of tissues. Its usefulness was further increased by additional amounts of copper sulphate, potassium nitrate and monobasic ammonium phosphate. Increased levels of iron and magnesium inhibited growth. Incorporation of yeast extract in the tobacco-high-salts-medium produced the highest amount of growth and chlorophyll formation in endive tissue. Presence of exogenous sucrose was essential for the continued good growth of the above callus tissues in vitro. Highest amount of growth took place either in white light or in the dark. Different tissues had different responses to high or low intensities of light. Endive and carrot tissues produced in vitro were palatable to human taste. Endive tissue was particularly good as it also differentiated many small rosettes of leaves, shoots and had a mild aromatic flavor typical of the endive plants grown in nature.
