    Notes: Abstract. Loss of heterozygosity (LOH) on chromosome 11q13 occurs in about 20% of sporadic adrenal neoplasms. Adrenal lesions, mostly benign, occur in up to 40% of patients from MEN I kindreds. The MEN I gene, positioned on 11q13, has been considered a primary candidate gene in these lesions. We studied a group of 15 patients with sporadic adrenal adenoma, and 1 patient with multinodular hyperplasia. Of the 16 patients, 4 had incidentally discovered masses, 5 had Conn's syndrome, 6 suffered from Cushing's syndrome, and 9 had high sex hormone production. Studies with the markers D11S480, PYGM, D11S449, and D11S987 in 13 patients (12 of whom were from our group of 16) revealed 4 losses of heterozygosity on D11S480 on 11q13, but the deletion did not affect the MEN I gene in any case. We present complete direct DNA sequencing data of the menin gene in 14 sporadic adrenal adenomas and one with adrenal hyperplasia. We identified one heterozygous missense mutation, T552S, in a hormonally inactive adrenal adenoma. One base exchange was identified close to the intron-exon boundary in intron 9 of a nodular adrenal hyperplasia. mRNA expression studies found that MEN I was transcribed in all 13 samples analyzed. In summary, our study identified the second patient with sporadic benign adrenal tumor presenting a menin gene mutation. Our complete direct sequencing approach adds evidence that menin gene mutations may account only for a minority of benign adrenal tumors if at all. Another tumor-suppressor gene inactivated in sporadic adrenal neoplasms may be located on chromosome 11q13.

    Notes: Summary This paper describes the first purification method for crystalline inclusions (cores) from plant peroxisomes and an ultrastructural characterization of these isolated cores. 5-day-old sunflower (Helianthus annuus L.) cotyledon fractions which were highly enriched in cores showed negligible activity of the matrix enzyme glycolate oxidase but high catalase activity. As proven by electron microscopy, crystalline particles were surrounded neither by matrix material nor by membranes. Their geometrical outlines and ultrastructure were identical to those of cores in tissue sections, as was their reactivity with three different polyclonal catalase antibodies in the immunogold technique. Three-dimensional reconstruction, based on the geometrical outlines and ultrastructure of sectioned isolated cores from sunflower, suggested that they were quadrangular blocks. Ultrastructural analysis revealed an even periodic arrangement of repeating units which are probably cubes with 20 nm long edges. Isolated peroxisomal cores from potato (Solanum tuberosum L.) tubers had outlines which suggested that they were even rhomboidal prisms. They showed a granular ultrastructure without any repeating units and contained catalase, demonstrated by immunogold labelling and enzyme activity measurement. The results presented here suggested the hypothesis that the structural elements in plant peroxisomal cores are made of enzymatically active catalase, although the substructure may vary from species to species.

    Description / Table of Contents: PVD-hardcoatings in Plastics Processing UnitsPVD-coatings for wear protection in plastics processing units were investigated. Therefore model wear tests were carried out in order to the tribological situation inside a screw/barrel system. The test methods covered both, the solid and the molten state. In addition the direct contact between the screw and barrel has been investigated. The results depict hard coatings to offer high wear resistance. The tribological interaction between moulding compounds and cavity surface is considerably influenced by TiN-coating systems. Demoulding experiments demonstrate an authoritative influence of the stoichiometry and the structure of the coatings on the adhesive force.
    Notes: Mit praxisgerechten Modellprüfapparaturen wurde die Einsatzmöglichkeit von PVD-Hartstoffschichten für verschleißbeanspruchte Bauteile in Schnecken-Plastifiziereinheiten überprüft. Simuliert wurden die Verhältnisse im Feststoff und Schmelzebereich sowie beim direkten Kontakt von Schneckensteg und Zylinderoberfläche. Es läßt sich zusammenfassend feststellen, daß Hartstoffschichten unter den vorgegebenen tribologischen Beanspruchungsbedingungen ein hohes Verschleißschutzpotential besitzen.Für die Anwendung der Hartstoffschichten in Spritzgießwerkzeugen wurden speziell die tribologischen Wechselwirkungen zwischen Kunststoff-Formmasse und Formnestoberfläche quantifiziert. Entformungsversuche zeigten, daß der Entformungsvorgang maßgeblich von der Schichtstöchiometrie und -struktur beeinflußt wird.

    Description / Table of Contents: Coatings in polymer processingCrxN coatings produced by PVD-magnetron sputtering (MSPVD) open unitsFor several years hard coatings have been used in plastic processing. The reduction of adhesion and contamination effects play a more and more important role besides the wear protection of injection moulds and extrusion tools. Applications of hard coatings in the field of highly stressed screws and barrels of plastification units are very seldom. In these cases the tribological effects are very intense so that the demanded wear resistance of hard coatings in the range 10μm is not sufficient. Different hard coatings based on titanium and chromium were tested according to their applicability for wear protection of screws in plastification units. Thickness, hardness, internal stresses and adhesion of the coatings were studied.Afterwards tribological tests in a model testing unit were investigated. Here the coatings were studied under tribological conditions, similar to those in a real plastification unit. The CrxN multilayer coatings showed extremely better wear resistance than the titanium based coatings. For this reason the CrxN coatings were optimized by help of the deposition parameters. These optimized coatings have been investigated according to their applicability for different polymers in comparison to other coating systems. First tests of CrCN/TiCN coated screw elements in practice showed very pleasing results. The test in practice with the optimized CrxN multilayer coating has yet to take place, but in the model tests the CrxN coatings showed excellent results.
    Notes: Hartstoffschichten werden bereits seit langem in der Kunststoffverarbeitung eingesetzt. Neben dem Verschleißschutz von Spritzguß-und Extrusionswerkzeugen stehen häufig auch die Verringerung von Klebneigungen und Formverschmutzungen im Vordergrund. Anwendungen im Bereich der hochbeanspruchten Schnecken und Zylinder von Plastifizieraggregaten sind noch selten. Die Verschleißeffekte sind hier so heftig, daß in der Praxis eine große Verschleißreserve gefordert wird, die Hartstoffschichten mit maximal 10 μm Schichtdicke naturgemäß nicht mitbringen. Verschiedene Hartstoffschichten auf Titan- und Chrombasis wurden hinsichtlich ihrer Eignung für den Verschleißschutz von Schnecken für Plastifizieraggregate untersucht. Schichtdicke, Härte, Eigenspannungen und Haftfestigkeit der Schichten wurden bestimmt.Anschließend wurden Verschleißversuche mit Hilfe einer Modellprüfapparatur, die die Schichten ähnlichen tribologischen Belastungen aussetzt, wie sie in einer Plastifiziereinheit auftreten, durchgeführt. Die CrxN-Mehrlagenschichten waren hierbei den auf Titan basierenden Schichten deutlich überlegen. Deshalb wurden für die CrxN-Schichten optimale Beschichtungsparameter bestimmt. Diese optimierten Schichten wurden mit verschiedenen anderen Schichtsystemen im Hinblick auf Ihre Eignung für unterschiedliche Kunststoffmaterialien verglichen. Erste Praxisversuche mit CrCN/TiCN-beschichteten Schneckenlementen sind sehr vielversprechend. Ein Praxistest der optimierten CrxN-Mehrlagenschichten steht zwar noch aus, jedoch konnten die guten Ergebnisse der CrxN-Schichten bei den Modellversuchen stets bestätigt werden.
