    Notes: History-dependent metastable states with different bulk properties are formed in the vortex state of the type-II superconductor 2H-NbSe2. Magnetic measurements demonstrate the difference between the shielding responses of a field- and a zero-field-cooled state, and provide a procedure for switching the system from one state to the other. © 1999 American Institute of Physics.

    Notes: Magnetization and 57Fe Mössbauer studies have been made in Fe3−xRuxSi for 0≤x≤2. Up to x=1.35, the alloys behave like collinear ferromagnets and μ/f.u. and HF (Fe) values do not show any appreciable change, thus suggesting that Ru replacing magnetic Fe (A,C) atoms also possess a large moment of ∼1 μB. For x〉1.35, the approach of magnetization to saturation becomes progressively slower and the system acquires an inhomogeneous magnetic structure. In the region 1.45 ≤x〈1.8, Tm and Thf, the HF (Fe) evolution temperature, fall rapidly, with Thf(very-much-less-than)Tm resulting in a behavior a la RuFeSi. For x〉1.8, majority of the Fe atoms are on B site and they presumably undergo AF/random freezing. Even though Tm and magnetization behavior show considerable variation across the region 1≤x≤2, the character of Mössbauer spectra does not change and 〈HF (Fe)〉av measured at 4.2 K remain ∼250 kOe.

    Notes: Abstract Heart tissue contains large amounts of the protein encoded by the Ca2+ pump gene SERCA2. The SERCA2 RNA can be spliced alternatively to produce mRNA encoding the proteins SERCA2a and SERCA2b which differ in their C-terminal sequences. In this study we report the tissue distribution of SERCA2a and SERCA2b isoforms byin situ hybridization to rabbit heart and stomach. The expression of SERCA2 mRNA was high in myocardial cells, being the highest in the atrial region. In contrast, there was more SERCA2 protein in Western blots in ventricles than in atria. Myocardial cells expressed predominantly the mRNA for the isoform SERCA2a. Whereas the stomach smooth muscle and the neuronal plexus expressed SERCA2 at levels much lower than myocardial cells, the expression was very high in the stomach mucosa. Mucosa contained mainly the mRNA for SERCA2b. From immunocytochemistry it was concluded that the anti-heart SR Ca2+ pump antibody IID8 reacted much better with heart and surface mucosal cells in the stomach than with the stomach smooth muscle, and that IID8 reactivity was intracellular. In contrast PM4A2B, an antibody against the plasma membrane Ca2+ pump, reacted well with heart and stomach smooth muscle, plexus and mucosa, and its localization appeared to be in the plasma membrane. Thus, stomach smooth muscle expressed SERCA2b mRNA and protein at low levels, mucosa expressed SERCA2b mRNA and protein at high levels, atria and ventricle expressed SERCA2a mRNA and protein at high levels, mRNA being more in atria, but protein being more in ventricles.

    Notes: Abstract The sarcoplasmic reticulum (SR) membranes isolated from rabbit heart were preincubated at pH 6.8 or 7.8 and their Ca2+ pump properties were compared at pH 6.8. The ATP-dependent azide insensitive oxalate-stimulated Ca2+ uptake was reduced more rapidly from the membranes preincubated at 37°C at pH 7.8 than from those preincubated at pH 6.8. The Ca2+−Mg2+-ATPase, and the Ca2+-dependent formation of 110 kDa acylphosphate were also inhibited by the preincubation at the higher pH. Including 1 mM DTT in the preincubation medium reduced the inactivation. The preincubation at 37°C in the presence or absence of DTT caused membranes to become more leaky as the loss of Ca2+ uptake was more rapid than that of ATPase or the acylphosphate formation. The loss of these activities was not accompanied by a breakdown of the protein as monitored in Western blots. It is hypothesized that the SR Ca2+ pump inactivation involves a key-SH group and that the lower pH provides a compensatory protective mechanism for the SR during acidosis.

    Notes: Abstract Densities of sarcoplasmic reticulum (SR) Ca2+-pump were compared in proximal and distal segments of pig left coronary artery using two biochemical methods: acylphosphate formation and immunoreactivity in Western blots, and a functional assay based on contraction to SR Ca2+-pump inhibitors. In the microsomes prepared from smooth muscle, the level of the 115 kDa SR Ca2+-pump acylphosphate was 7.1 ± 0.3 -fold greater in distal than in proximal segments. Similarly in Western blots using these microsomes, the reactivity of the 115 kDa band to an anti-SR Ca2+-pump antibody was 5.3 ± 0.8 -fold greater in distal than in proximal segments. Endothelium free coronary artery rings contracted to the SR Ca2+-pump inhibitors Cyclopiazonic acid (CPA, EC50 = 0.19 ± 0.06 μM) and thapsigargin (EC50 = 0.0095 ± 0.0035 μM). With 10 μM CPA, the force of contraction per tissue wet weight was 4.2 ± 0.5-fold greater in distal than in proximal rings, and with 1 μM thapsigargin it was 4.0 ± 1.0 -fold greater. The contractions produced by 60 mM KCl were used as a control. In contrast to the CPA and thapsigargin, the force per mg tissue weight produced by 60 mM KCl did not differ significantly between the proximal and distal segments. Thus, the results from the two biochemical methods and those from the contractility data were all consistent with the smooth muscle in the distal segments of the coronary artery containing a higher density of the SR Ca2+-pump than the proximal segments.

    Notes: Abstract Pig coronary artery rings denuded of endothelium contract to the vasoactive hormone angiotensin II (Ang II). The nature of Ang II receptors and their Ca2+-pool utilization were examined for contraction of the artery rings and for increase in ultracellular [Ca2+] ([Ca2+]i) in smooth muscle cells cultured from them. Ang II contracted the arteries (EC50=7±4 nM) but with a lower maximal force (1.4±0.25 N/g tissue) than the contraction with 60 mM K+ (6.11±0.63 N/g tissue). In the cultured cells it caused a transient increase in [Ca2+]i with an EC50 value of 11±4 nM. The cells bound Ang II with a dissociation constant (Kd) of 7±2 nM. Based on the effects of the Ang II antagonists saralasin, DuPont 753, dithiothreitol and PD123319, the Ang II receptors responsible for contraction, increase in [Ca2+]i and Ang II binding to coronary artery smooth muscle were of type AT1. The contraction to Ang II was abolished by EGTA but not by nitrendipine. The sarcoplasmic Ca2+ pump inhibitors cyclopiazonic acid (10 μM CPA) and thapsigargin (1 μM) produced contractions of 4.35±0.73 and 2.07±0.54 N/g, respectively. Ang II contractions in the control arteries were nearly abolished upon pretreatment with CPA and thapsigargin. CPA and thapsigargin induced contractions were abolished by exposure to EGTA for 1 h but short exposure of the cells to EGTA only modulated the CPA or thapsigargin induced increase in [Ca2+]i; Ang II induced increase in [Ca2+]i was not inhibited by 1 μM nitrendipine but was reduced significantly by a 30–60 sec exposure to EGTA. CPA and thapsigargin caused an increase in [Ca2+]i even after 30–200 sec exposure to EGTA. Ang II when added after CPA or thapsigargin did not cause a further increase in [Ca2+]i but when added before them it caused an increase in [Ca2+]i and reduced the increase caused by subsequent addition of CPA or thapsigargin. These data are consistent with the concept that Ang II utilizes an intracellular Ca2+-pool which is a small component of the CPA or thapsigargin sensitive Ca2+-pool.

    Notes: Abstract Pig coronary artery cultured smooth muscle cells were skinned using saponin. In the presence of an ATP-regenerating system and oxalate, the skinned cells showed an ATP-dependent azide insensitive Ca2+-uptake which increased linearly with time for 〉1 h. The Ca2+-uptake occurred with Km values of 0.20±0.03 μM for Ca2+ and 400±34 μM for MgATP2−. Thapsigargin and cyclopiazonic acid inhibited this uptake with IC50 values of 0.13±0.02 and 0.56±0.04 μM, respectively. These properties of SR Ca2+-pump are similar to those reported for membrane fractions isolated from fresh smooth muscle of coronary artery and other arteries. However, optimum pH of the uptake in the skinned cells (6.2) was lower than that reported previously using isolated membranes (6.4–6.8).

    Notes: Abstract 151Eu Mössbauer studies have been made across the respective superconductive transition temperatures in two compounds, La1.7Eu0.1Sr0.2CuO4−y and EuBa2Cu3O7, having zero resistance state at 25K and 85K respectively. The151Eu isomer shift data establish that Eu ions are in trivalent state at all temperatures. The Mössbauer f-factor does not show any significant anomaly across the TC.
