    Notes: Summary The distribution of endothelin mRNA and immunoreactivity in the human brain was investigated using the technique of in situ hybridization and immunocytochemistry. Cryostat sections from 22 cases of neurologically normal adult human brain, collected 3–7 h post-mortem were hybridized with 35S-labelled complementary (c)RNA probes prepared from the 3′ non-coding region of endothelin-1 cDNA, and the chromosomal genes encoding endothelin-2 and -3. In situ hybridization with all three cRNA probes revealed labelled neuronal cell bodies in laminae III–VI of the parietal, temporal and frontal cortices. Labelled cells were also seen, scattered throughout the para- and periventricular; supraoptic and lateral hypothalamic nuclei, the caudate nucleus, amygdala, hippocampus, basal nucleus of Meynert, substantia nigra, raphe nuclei, Purkinje cell layer of the cerebellum and in the dorsal motor nuclei of the vagus of the medulla oblongata. The distribution of neurones immunoreactive to endothelin was similar to that of endothelin mRNA, although fewer immunoreactive cells throughout the brain, were noted. Immunoreactive fibres were present mainly in the cortex and hypothalamus, and to a lesser extent in the brain stem. Combined in situ hybridization and immunocytochemistry on the same section revealed the presence of endothelin-1 mRNA and immunoreactivity in the same cortical neuronal cell. Colocalisation studies in the cortex revealed endothelin-1 mRNA and immunoreactivity in a number of cells which also expressed neuropeptide Y mRNA and immunoreactivity. In the hypothalamus and basal nucleus of Meynert endothelin immunoreactivity was colocalised to a subset of neurophysin- and galanin-immunoreactive cell bodies respectively. Endothelin mRNA and immunoreactivity was also seen in some blood vessel endothelial cells. The findings of endothelin mRNAs and immunoreactivity in heterogenous neuronal populations further emphasises the potential role of endothelin as a neuropeptide, probably having diverse actions in the nervous system of man.

    Notes: Summary Poly-l-lysine (PPL) has been used to coat glass slides in the preparation of tissue sections for immunocytochemical staining. The adhesive properties of different molecular weight (m.w.) polymers of l-lysine have been tested on pre-fixed cryostat sections which were subjected to a 3 day washing treatment. It has been found that the higher the molecular weight of the polymer, the greater the adhesive force it provides. PLL (m.w. 350,000) at concentrations in the range of 0.05–0.1% was found to be the most effective polymer.

    Notes: Abstract A class of two-persons zero-sum differential games with the possibility of singular arcs is considered. Under some assumptions, a sufficient condition for the existence of a saddle-point solution is derived. In this approach, the properties of convexity and concavity of the performance index are used to find the sufficient condition. It is observed that, unlike total singularity, the occurrence of partially singular arcs does not cause any problems for the existence of a unique saddle-point solution.

    Description: Stable isotopes of oxygen and hydrogen were measured in water samples collected annually from a representative suite of 50 lakes in northeastern Alberta over a nine-year period, and are interpreted using a steady-state isotope mass balance model to determine water yield and runoff ratios for the lake watersheds, and residence time of the lakes. This isotopic perspective on hydrology of the region provides new insight into the role of land cover, watershed morphometry, climatic drivers and permafrost thaw on lakes. Bog cover, permafrost and presence of thaw features in bogs are found to be the dominant hydrologic drivers, although morphometric properties such as elevation, lake area, and drainage basin area are also influential. In addition to quantifying the hydrologic fluxes, the analysis establishes contrasting conditions in more southerly lakes, located in the Stony Mountains and west of Fort McMurray, as compared to more northerly sites in the Birch Mountains, Caribou Mountains and northeast of Fort McMurray, due mainly to contributions from thawing permafrost at the northerly sites. Distinct hydrologic conditions are also noted for Shield systems north of Lake Athabasca where bogs and permafrost are absent. While permafrost thaw is not directly labelled by oxygen and hydrogen isotope composition, isotope mass balance calculations suggest that contributions of up to several hundred millimetres per year are occurring in 14 of the 50 lake watersheds under study. Several of these lakes have water yields in excess of precipitation in some years and regional groups of lakes display significant correlations between water yield and percentage of bogs that have collapsed.     Description: We present CO, H 2 , H i and HISA (H i self-absorption) distributions from a set of simulations of grand design spirals including stellar feedback, self-gravity, heating and cooling. We replicate the emission of the second galactic quadrant by placing the observer inside the modelled galaxies and post-process the simulations using a radiative transfer code, so as to create synthetic observations. We compare the synthetic data cubes to observations of the second quadrant of the Milky Way to test the ability of the current models to reproduce the basic chemistry of the Galactic interstellar medium (ISM), as well as to test how sensitive such galaxy models are to different recipes of chemistry and/or feedback. We find that models which include feedback and self-gravity can reproduce the production of CO with respect to H 2 as observed in our Galaxy, as well as the distribution of the material perpendicular to the Galactic plane. While changes in the chemistry/feedback recipes do not have a huge impact on the statistical properties of the chemistry in the simulated galaxies, we find that the inclusion of both feedback and self-gravity are crucial ingredients, as our test without feedback failed to reproduce all of the observables. Finally, even though the transition from H 2 to CO seems to be robust, we find that all models seem to underproduce molecular gas, and have a lower molecular to atomic gas fraction than is observed. Nevertheless, our fiducial model with feedback and self-gravity has shown to be robust in reproducing the statistical properties of the basic molecular gas components of the ISM in our Galaxy.

    Description: Herschel far-infrared continuum data obtained as part of the Hi-GAL survey have been used, together with the GLIMPSE 8 μm and MIPSGAL 24 μm data, to attempt the first 3D-decomposition of dust emission associated with atomic, molecular and ionized gas at 15 arcmin angular resolution. Our initial test case is a 2 2 square degrees region centred on ( l , b ) = (30°, 0°), a direction that encompasses the origin point of the Scutum–Crux Arm at the tip of the Galactic Bar. Coupling the IR maps with velocity maps specific for different gas phases (H i 21cm, 12 CO and 13 CO, and radio recombination lines), we estimate the properties of dust blended with each of the gas components and at different Galactocentric distances along the line of sight (LOS). A statistical Pearson's coefficients analysis is used to study the correlation between the column densities estimated for each gas component and the intensity of the IR emission. This analysis provides evidence that the 2 2 square degree field under consideration is characterized by the presence of a gas component not accounted for by the standard tracers, possibly associated with warm H 2 and cold H i . We demonstrate that the IR radiation in the range 8 〈  〈 500 μm is systematically dominated by emission originating within the Scutum–Crux Arm. By applying an inversion method, we recover the dust emissivities associated with atomic, molecular and ionized gas. Using the DustEM model, we fit the spectral energy distributions for each gas phase, and find average dust temperatures of T d,H i  = 18.82 ± 0.47 K, T d,H 2  = 18.84 ± 1.06 K and T d,H ii  = 22.56 ± 0.64 K, respectively. We also obtain an indication for polycyclic aromatic hydrocarbons depletion in the diffuse ionized gas. We demonstrate the importance of including the ionized component in 3D-decompositions of the total IR emission. However, the main goal of this work is to discuss the impact of the missing column density associated with the dark gas component on the accurate evaluation of the dust properties, and to shed light on the limitations of the inversion method approach when this is applied to a small section of the Galactic plane and when the working resolution allows sufficient de-blending of the gas components along the LOS.

    Description: The Southern Parkes Large-Area Survey in Hydroxyl (SPLASH) is a sensitive, unbiased, and fully sampled survey of the southern Galactic plane and Galactic Centre in all four ground-state transitions of the hydroxyl (OH) radical. The survey provides a deep census of 1612-, 1665-, 1667-, and 1720-MHz OH absorption and emission from the Galactic interstellar medium, and is also an unbiased search for maser sources in these transitions. We present here first results from the SPLASH pilot region, which covers Galactic longitudes 334° to 344° and latitudes ±2°. Diffuse OH is widely detected in all four transitions, with optical depths that are always small (averaged over the Parkes beam), and with departures from local thermodynamic equilibrium common even in the 1665- and 1667-MHz main lines. To a 3 sensitivity of ~30 mK, we find no evidence of OH envelopes extending beyond the CO-bright regions of molecular cloud complexes, and conclude that the similarity of the OH excitation temperature and the level of the continuum background is at least partly responsible for this. We detect masers and maser candidates in all four transitions, approximately 50 per cent of which are new detections. This implies that SPLASH will produce a substantial increase in the known population of ground-state OH masers in the southern Galactic plane.
