Notes: Abstract A method was developed for the chromatographic separation of V(V) and V(IV) based on the different sorption forces of these vanadium species in C18 columns in presence of KH-phthalate. The vanadium species were detected with a flame atomic absorption spectrometer with acetylene/N2O flame. The detection limits (3σ) of V(V) and V(IV) were 0.18 μg/mL and 0.15 μg/mL, respectively. The relative standard deviations (N = 5) are 4.2% and 3.4% for 20–20 μg/mL V(V) and V(IV), respectively. The sampling frequency is 75/h. Because of the special interaction occurring between phthalate and V(IV) on the C18 column and the acetylene/N2O flame atomic absorption detection, practically no interferences can be detected even in large inorganic matrix.

Notes: Abstract The sorption loop as a preconcentration unit used so far in the low-pressure flow injection (FI) system was combined with the hydraulic high-pressure nebulizer (HHPN) and a high-pressure liquid chromatographic (HPLC) column. Thus, the sorption preconcentration of Cr(VI) can be coupled to a powerful sample introduction method or preconcentration techniques of Cr(III). The subsequent determination of the Cr(VI) content of the samples was carried out by flame atomic absorption (FAAS).

Notes: Abstract A low molecular mass anticoagulant (17 kDa) was isolated from the salivary glands of prefed female Hyalomma truncatum ticks by means of reverse phase and anion-exchange HPLC. Trypsin digestion and amino acid analysis confirmed the protein nature of the anticoagulant. The inhibitor appears to be uncompetitive with a Ki of 6.9×10−10M. The target of the anticoagulant is factor Xa at the junction of the extrinsic and intrinsic pathways. This may be crucial for the survival of the tick, making it feasible to investigate the possibility of vaccination with this antihaemostatic against tick feeding. In addition, tick anticoagulants may possibly have therapeutic application in controlling thrombosis.

Notes: Abstract Salivary gland extracts of the sand tampan, Ornithodoros savignyi, prolonged the activated partial thromboplastin time (APTT) and prothrombin time (PT) significantly in a concentration-dependent manner. There was also a pronounced inhibition of human activated factor Xa (fXa) by salivary gland extracts. The salivary gland extracts inhibited chromogenic assays specific for both fXa and thrombin. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of the salivary gland proteins followed by elution of specific areas or bands from a polyvinylidene difluoride (PVDF)-membrane, showed that various anticoagulant factors are present when screened by means of the APTT assay. The most active component was associated with a band of M r of 14 kDa. Partial purification of this component was achieved using isoelectric focusing (IEF) and size-exclusion highperformance liquid chromatography (HPLC).

Notes: Abstract An inhibitor of activated coagulation factor X (fXa) was isolated from salivary gland extracts prepared from Ornithodoros savignyi using a two-step procedure, involving reversed-phase high-performance liquid chromatography (RP-HPLC) and diethylaminoethyl (DEAE) ion-exchange chromatography. From its behaviour during DEAE chromatography it could be deduced that it possesses an acidic pI (∼4.6). Capillary zone electrophoresis (CZE) of the purified inhibitor showed it to be homogeneous. The molecular mass was determined as 12 kDa using capillary gel electrophoresis (CGE) and as 7183.4 using laser desorption mass spectrometry (LDMS). The N-terminal amino acid sequence (residues 1–12) was determined and found to share a 66% identity with tick anticoagulant peptide (TAP). The O. savignyi peptide is a slow, tight-binding inhibitor of fXa (K i=0.83±0.10 nM). The interaction of the fXa-inhibitor was found to be competitive and dependent on ionic strength. Preliminary investigations show that the inhibitor may be specific for fXa.

Description: The paguara Chaetodipterus faber (Broussonet, 1782) is a species of economic importance in the South Caribbean. Juveniles were collected near sandy beaches of Margarita Island (Venezuela) and maintained in pompano cage cultures until they reached maturation of their gonads. Adults were selected and spawning was induced by means of gonadotropin hormones. The eggs of Chaetodipterus faber are spherical, transparent and bouyant, with a diameter of 0.9 - 1.1 mm and a single globule oil of 0,25 - 0.35 mm diameter. Hatching of C. faber eggs took place within 26 h at temperatures of 24.5 - 28.5°C (31-32 h at 20-23.5°C) and salinities between 37.5 - 38.5Voo. Newly hatched larvae vary in length from 1.9 to 2.1 mm and have a yolk sac of 1.1. - 1.3 mm in total length which is deploted after two or three days. The larval phase was completed 25 days after hatching. Juveniles 45 days old have 17.17 ± 1.75 mm in total length and weigh 0.21 ± 0.07 g. Development of eggs and larvae are described.

Description: The northeast coast of Venezuela is the most important fishing region in the country, the sardines (Sardinella aurita) and other species abound in its waters, the annual catch exceeds 300.000 mt. Several factors account for this fertility: an upwelling of subsurface waters, the influence of great South American rivers (notably the Orinoco river), the coastallagoons, and the presence of numerous islands. In the island of Margarita, the possibilities of cultivating 16 species or marine fishes have been studied, but so far only two species, the spadefish Chaetodipterus faber (Ephippidae) and the permit Trachinotus goodei (Carangjdae) show any potential of intensive production. Both species reach their commercial weight (300 - 400 p;) in less than one year, show gonadal maturity and spawn in confinement. For C. faber controlled reproduction and the raising of brood have been achieved but fattening should still be optimized. Trachinotus goodei is a very disease resistant species and grows relatively fast. In this paper we discuss the realistic possibilities of cultivation for both species, and the problems that must be solved. Success depends upon the use of adequate dry feed in floating cages in this installations. Experiments with snappers, Lutjanus, and dolphinfish, Coryphaena hippurus (Coryphaenidae), are recommended, because adults of these species are caught in this area all year round, which should facilitate all activities related with reproduction and the raising of juveniles.
