Notizen: Abstract— The subject of hypersingular boundary integral equations is a rapidly developing topic due to the advantages which this kind of formulation offers compared to the standard boundary integral method. The hypersingular formulation is particularly well suited for fracture mechanics problems, where there are important gradients of the stress field and singularities. This formulation for time domain antiplane problems has been recently addressed by the authors and in the present paper, the formulation for time domain plane problems is presented and applied for the first time. A mixed Boundary Element approach based on the standard integral equation and the hypersingular integral equation is developed. The mixed formulation allows for a very simple discretization of the problem, where no subregion is needed. Conforming quadratic elements are used for the crack and the external boundaries. The hypersingular integral equation is used for collocation points within the crack elements, while the standard integral representation is used for the external boundaries. Several examples with different crack geometries are studied to illustrate the possibilities of the method. The Stress Intensity Factor (S.I.F.) is very accurately computed from the crack tip opening displacements along the crack tip element. The results show that the proposed approach for S.I.F. evaluation is simple and produces accurate solutions.

Notizen: Polypropylene/polyamide-6 and polypropylene/metallocene polyethylene blends containing 2.5 phrof organophilic modified montmorillonite were prepared in a twin-screw extruder followed byinjection molding. In order to compare, blends without layered clay were also made. Styreneethylene-butylene-styrene copolymer and polypropylene grafted with anhydride maleic were usedas compatibilizers in the ternary blends and in the PP nanocomposite preparation, respectively. Thepresence of tactoids, intercalated and exfoliated structures was observed by TEM in some of thesamples containing layered clay and modified PP materials. Results showed that the compatibilizedblends prepared without clay are tougher than those prepared with the nanocomposite of PP as thematrix phase and no significant changes in tensile moduli were observed between them. However, the binary blend with a nanocomposite of PP as matrix and metallocene polyethylene exhibitedbetter tensile toughness and lower tensile modulus, than those prepared with a nanocomposite of PPand polyamide-6 as dispersed phase. These results are related to the degree of clay dispersion in thePP and to the type of morphology developed in the different blends. Differential scanningcalorimetry (DSC) showed that blends with a finer and homogeneously dispersed morphologydetermined by SEM, the PA component exhibited fractionated crystallization exotherms in thetemperature range of 159-185°C. Also, nucleation of the PP component by PA phase and/or thelayered clay was observed in the blends with PA as dispersed phase

Notizen: Summary The presence of calcitonin gene-related peptide (CGRP) in the skin of pig snout and human fingertip was investigated using immunohistochemical techniques. CGRP immunoreactivity was found in Merkel cells and nerve fibres of both species. In pig snout skin, Merkel cells containing CGRP were seen forming clusters at the tips of rete ridge epidermis and in the external root sheath of sinus hair follicles (vibrissae). Human Merkel cells immunostained for CGRP were found isolated or forming small groups in the basal layer of glandular epidermal ridges. In all cases, immunoreactivity was more intense on the side of the Merkel cell facing the associated nerve terminal (which was never positive for CGRP). This part of the Merkel cell has the greatest density of dense-cored granules, suggesting that CGRP must be stored in these granules. Nerve, bundles containing CGRP-immunoreactive fibres were found at dermal and hypodermal level, and blood vessels were often surrounded by CGRP nerve fibres. In pig snout skin some nerve fibres containing CGRP penetrated the epidermis and terminated as free endings, and in the human fingertip a small number of CGRP-immunoreactive nerve fibres were seen in Meissner's corpuscles.

Notizen: Summary Using immunohistochemical methods, we have investigated the cellular distribution of prothymosin alpha (ProT) in adult rat testis. A policlonal antibody raised against thymosin alpha 1 conjugated to keyhole limpet hemocyanin was used. ProT immunoreactivity was observed in the cytoplasm and nucleus of spermatogonia and primary spermatocytes in initial phases of the first meiotic division, preleptotene, leptotene and zygotene. However, in pachytene phase they already showed a weak or negative staining. On the other hand, secondary spermatocytes, spermatids, spermatozoa and Sertoli cells were not stained. Based on this fact we suggest that ProT is present in the proliferative cycle in the final steps of G1 phase, throughout the S and G2 phases and in initial steps of the prophase.

Notizen: Summary The cellular distribution of prothymosin alpha (ProT) was studied in ovarian follicles of adult cycling rats. We found positive granulosa and theca cells throughout follicular maturation. When both ProT and proliferating cell nuclear antigen (PCNA) immunoreactivity was studied, we observed that both proteins were expressed in the same granulosa and theca cells, although sometimes ProT immunoreactivity was weak or absent in the mitotic (M) phase. Moreover, both peptides share the nuclear distribution, but ProT immunoreactivity was never seen in nucleoli. Therefore, we conclude that in mitotic cells ProT is expressed only in actively proliferating cells, since all ProT-positive cells were also positive for PCNA. ProT and PCNA immunoreactivities during the meiotic division were studied in oocytes. The presence of PCNA was, unlike ProT, constant throughout follicle development (except atretic oocytes). Oocytes expressed ProT from primordial follicles to the eighth generation, but more developed oocytes and atretic oocytes were not immunoreactive. In hypophysectomized rats, all oocytes were immunoreactive. Interestingly, in hypophysectomized rats treated with follicle stimulating hormone (FSH) that promoted follicle development, the more developed oocytes did not show ProT immunoreactivity. Since hypophysectomized rats were not treated with luteinizing hormone we conclude that ProT expression is not required to complete meiotic division I.

Notizen: Summary The genetic similarities of eight closely related rye cultivars were estimated using two molecular marking techniques: restriction fragment length polymorphism (RFLP) and random amplified polymorphic DNA (RAPD). Cultivars were evaluated for variation by 11 random cDNA and genomic clones used in combination with four restriction enzymes and 40 decamer primers. A total of 53 polymorphic RFLP fragments and 94 polymorphic RAPD fragments were observed. Based on the presence/absence of fragments, two genetic similarity matrices were calculated which were then used in cluster analysis. Differences between pair of cultivars were observed in RFLP and RAPD dendrograms. RFLP analysis produced estimates of genetic relationships more in accordance with the partially known pedigree of the cultivars than did RAPD analysis. The use of bulk samples of DNA in these analyses affected the sensitivity of RAPD assays more strongly. Dendrograms which took into account all fragments produced, either by RFLP or RAPD, reflected better the relationships between cultivars than did dendrograms based on only one type of marker. This reflects the importance of the number of markers used in determining the genetic relationships between genotypes.

Notizen: The topic of hypersingular boundary integral equations is a rapidly developing one due to the advantages which this kind of formulation offers compared to the standard boundary integral one. In this paper the hypersingular formulation is developed for time-domain antiplane elastodynamic problems. Firstly, the gradient representation is found from the displacement one, removing the strong singularities (Dirac's delta functions) which arise due to the differentiation process. The gradient representation is carried to the boundary through a limiting process and the resulting equation is shown to be consistent with the static formulation. Next, the numerical treatment of the traction boundary integral equation and its application to crack problems are presented. For the boundary discretization, conforming quadratic elements are tested, which are introduced in this paper for the first time, and it is shown that the results are very good in spite of the lesser number of unknowns of this approach in comparison to the non-conforming element alternative. A procedure is devised to numerically perform the hypersingular integrals that is both accurate and versatile. Several crack problems are solved to show the possibilities of the method. To this end both straight and curved elements are employed as well as regular and distorted quarter point elements.

Notizen: This paper presents a procedure for transient dynamic stress intensity factor computations using traction singular quarter-point boundary elements in combination with the direct time domain formulation of the Boundary Element Method. The stress intensity factors are computed directly from the traction nodal values at the crack tip. Several examples of finite cracks in finite domains under mode-I and mixed mode dynamic loading conditions are presented. The computed stress intensity factors are represented versus time and compared with those obtained by other authors using different methods. The agreement is very good. The results are reliable and little mesh dependent. These facts allow for the analysis of dynamic crack problems with simple boundary discretizations. The versatile procedure presented can be easily applied to problems with complex geometry which include one or several cracks.

Notizen: The present paper deals with the study and effective implementation for Stress Intensity Factor computation of a mixed boundary element approach based on the standard displacement integral equation and the hypersingular traction integral equation. Expressions for the evaluation of the hypersingular integrals along general curved quadratic line elements are presented. The integration is carried out by transformation of the hypersingular integrals into regular integrals, which are evaluated by standard quadratures, and simple singular integrals, which are integrated analytically. The generality of the method allows for the modelling of curved cracks and the use of straight line quarter-point elements. The Stress Intensity Factors can be computed very accurately from the Crack Opening Displacement at collocation points extremely close to the crack tip. Several examples with different crack geometries are analyzed. The computed results show that the proposed approach for Stress Intensity Factors evaluation is simple, produces very accurate solutions and has little dependence on the size of the elements near the crack tip.
