    Description: Calcareous foraminifer shells (tests) represent one of the most important archives for paleoenvironmental and paleoclimatic reconstruction. To develop a mechanistic understanding of the relationship between environmental parameters and proxy signals, knowledge of the fundamental processes operating during foraminiferal biomineralization is essential. Here, we apply microscopic and diffraction‐based methods to address the crystallographic and hierarchical structure of the test wall of different hyaline foraminifer species. Our results show that the tests are constructed from micrometer‐scale oriented mesocrystals built of nanometer‐scale entities. Based on these observations, we propose a mechanistic extension to the biomineralization model for hyaline foraminifers, centered on the formation and assembly of units of metastable carbonate phases to the final mesocrystal via a non‐classical particle attachment process, possibly facilitated by organic matter. This implies the presence of metastable precursors such as vaterite or amorphous calcium carbonate, along with phase transitions to calcite, which is relevant for the mechanistic understanding of proxy incorporation in the hyaline foraminifers.
    Description: Plain Language Summary: Foraminifers are single celled marine organisms typically half a millimeter in size, which form shells made of calcium carbonate. During their life, the chemical composition of their shells records environmental conditions. By analyzing fossil shells, past conditions can be reconstructed to understand ancient oceans and climate change. To do that correctly, we need to know exactly how foraminifers form their shell. We find that foraminifers build micrometer‐sized mesocrystals which are made of smaller building blocks. This means that the smallest building blocks form first and assemble to form a larger grain, which is oriented in a specific direction. To align all the building blocks, it is possible that they are first unstable and undergo transformation on assembly, during which their composition may change. By understanding and quantifying this process, the composition of the final fossil shell may be understood, ultimately leading to more reliable reconstructions of past environmental change.

    Notes: Abstract. The Everglades Nutrient Removal Project (ENRP) is a 1,545-ha prototype wetland constructed to test a larger system of stormwater-treatment areas that is being used to remove agricultural nutrients and help restore the remnant Everglades. Concerns that the ENRP may lead to mercury bioaccumulation as observed in other newly created reservoirs resulted in an effort to construct a mercury mass budget and measure bioaccumulation in aquatic organisms. For the first 2 years of operation, total mercury (THg) and methylmercury (MeHg) concentrations in water decreased significantly a short distance into the ENRP, probably through particle settling. During both years, THg concentrations in largemouth bass were about 0.1 μg/g throughout the ENRP while the adjacent Water Conservation Area bass were at or above the state health advisory level of 0.5 μg/g. THg levels in mosquitofish were lower than levels found in bass, and mosquitofish mercury concentrations in the interior of the ENRP were significantly lower than the inflow and outflow sites. The major THg pool was the sediment while less than 1% the total mass was stored in vegetation, water, and fish. The major MeHg pool also was the sediment but vegetation, water, and fish were significant fractions. Mass budget estimates showed the inflow pump and rainfall to be the major sources of THg, while the outflow pump was the major output. The inflow pump was the major source of MeHg while the outflow pump was the major output. Evasion of elemental mercury estimated by the stagnant-film model indicated a minor loss, but field measurements over cattails suggested that evasion may be the major pathway. When all inputs and outputs were considered, the ENRP removed about 70% of the THg and MeHg masses in the first 2 years of operation.

    Notes: Abstract La Désirade, a small island east of Guadeloupe, is underlain by the only exposed pre-Tertiary basement rocks in the Lesser Antilles. The basement complex comprises spilitic and keratophyric flows and pillow lavas (with interbedded and overlying radiolarian cherts), swarms of mafic to silicic dikes, and subjacent plagiogranite. These features, and the absence of carbonates, terrigenous clastic sediments, or tuffaceous sediments from the complex indicate that it developed in a deep marine environment beyond the reach of terrigenous sedimentation or emergent island arc pyroclastic deposition. Previous workers have suggested that the Désirade basement complex originated either as oceanic crust or during an early (tholeiitic) stage of island arc growth. The isotopic compositions of Sr and Pb from the complex, and previously reported rare earth data (Johnston and Schilling, 1974) do not provide a clear distinction between these two possibilities. Nor does the presence of siliceous keratophyre in the complex rule out an oceanic crustal origin-such rocks are common in well studied ophiolites that originated as oceanic crust. Hence we turn to the age relationships of the complex, the surrounding ocean floor, and adjacent island arcs in an attempt to resolve this problem. The age of the complex strongly supports an oceanic crustal (ophiolitic) origin. The ages of zircons and a previously reported K-Ar age indicate that the complex is 145±5 m.y. old. The complex predates the next oldest volcanic rocks of the Lesser Antilles arc by ca. 110 m.y., and the oldest known rocks of the Aves Ridge, a possible Mesozoic precursor of the Lesser Antilles arc, by 50–60 m.y. This makes it unlikely that the Désirade complex is related to an early phase of either of these arcs. Instead, the age of the complex falls in the range of ages expected for oceanic crust in the vicinity of the Lesser Antilles prior to the development of any subduction zone and resulting arc. Thus we interpret the Désirade complex to be an uplifted segment of oceanic crust that represents the basement on which the later island arcs grew: first the Aves Ridge, an arc that was active in middle to late Cretaceous time (but whose exact mode of origin is enigmatic, and is considered in four alternate tectonic models), then the Eocene to Recent Lesser Antilles arc.

    Notes: Abstract Tidal inlets along the mesotidal coast of Maine contrast with those from other parts of the world by being dominated by flood-tidal currents. Analysis of the factors responsible for flood or ebb dominance indicates factors external to the backbarrier environment. We suggest that the flood dominance is caused by both a steepening of the tidal wave in the Gulf of Maine and the shallow depth of the ebb-tidal delta and spit platform. Flood currents are typically 10–20 cm/sec stronger than the ebb at the inlet throat. The flood dominance results in a significant net landward transport of sediment into the backbarrier.

    Notes: The cylic undecapeptides cyclosporin (Csp) A, CspD and dihydro CspC (HCspC) were analyzed by high-performance liquid chromatography/thermospray-mass spectrometry (HPLC/TSP-MS) on line with 1-2 pmol injected on column. Mass spectra were characterized by signals corresponding to the [M + H]+ ion as well as fragment ions derived from he loss of 112 (CspA and CspD) or 44, 114 and 114+44 (HCspC) mass units from the parent ion. The same qualitative profile was observed for negative-ion acquisition where the ions were formed by proton abstraction. The application of the technique to the characterization of CspA and its major hydroxylated and dealkylated metabolites in human blood samples is presented.

    Notes: The topography and properties of plasma membrane proteins from mouse L-929 cells are studied by comparing their availability for enzymatic labeling on the external and internal surfaces of the membrane. In order to study the internal surface, phagolysosomes are prepared from cells after they ingest latex particles. The plasma membrane surrounding these seems to have an “inside-out” orientation. The sugars of the membrane glycoproteins in intact phagolysosomes are not available for interaction with lectins or available for periodate-borotritide labeling. A comparison of the lectin-binding proteins lableled by lactoperoxidase-catalyzed iodination on the external cell surface with those labeled on the internal cell surface suggests that a variety of plasma membrane glycoproteins span the lipid bilayer.Using two-dimensional gel electrophoresis it has been shown that selected proteins are labeled at both the internal and external faces of the plasma membrane. Analysis of the 2-D gel electrophoregrams reveals that there are two distinct prominent proteins at 60,000 and 100,000 daltons which are enzymatically iodinated from both sides of the membrane. The partial hydrolysis of the 100,000 dalton protein reveals that different peptides are iodinated when the iodination is performed on intact cells or on the phagolysosomes. These proteins are extensively phosphorylated in cells incubated with inorganic 32P. We conclude that the phagolysosome is probably oriented in an “inside-out” configuration and that this membrane preparation can be used to study the topographic organization of membrane proteins.The use of oriented membranes, selective labeling of proteins, and affinity separation of proteins in combination with gel electrophoresis to define the position and properties of proteins is discussed.
