    Notes: We have characterized the interaction of the Neis-seria meningitidis TonB-dependent receptor HpuAB with haemoglobin (Hb). Protease accessibility assays indicated that HpuA and HpuB are surface exposed, HpuB interacts physically with HpuA, and TonB energization affects the conformation of HpuAB. Binding assays using [125I]-Hb revealed that the bipartite receptor has a single binding site for Hb (Kd≈ 150 nM). Competitive binding assays using heterologous Hbs revealed that HpuAB Hb recognition was not species specific. The binding kinetics of Hb to HpuAB were dramatically altered in a TonB– mutant and in wild-type meningococci treated with the protonophore carbonylcyanide m-chlorophenylhydrazone (CCCP), indicating that TonB and an intact proton motive force are required for normal Hb binding and release from HpuAB. Our results support a model in which both HpuA and HpuB are required to form a receptor complex in the outer membrane with a single binding site, whose structure and ligand interactions are significantly affected by the TonB-mediated energy state of the receptor.

    Notes: We have previously described HpuAB, a two-component receptor that mediates binding to haemoglobin (Hb), haemoglobin–haptoglobin (Hb-Hp) and apo-haptoglobin (Hp). In this communication, we constructed non-polar mutations in the hpuA and hpuB loci to examine the individual roles of HpuA and HpuB. Our results indicate that both HpuA and HpuB are required for the acquisition of Fe from Hb and Hb-Hp. We isolated Hb utilization-positive (Hb+) variants of our Hb utilization-negative (Hb−) hpu mutants at a frequency of 10−3 and demonstrated that the Hb+ phenotype resulted from the expression of a second Hb receptor, HmbR. Expression of HmbR in DNM2 was found to be controlled by translational frameshifting involving a polyguanine (G) tract located within the hmbR locus. The hpuA locus also contains a poly(G) tract, which suggested that meningococci could phase vary each Hb receptor independently by slip-strand mispairing in the poly(G) tracts found in hpuA and hmbR. Thus, we isolated a naturally occurring Hb− variant of DNM2, designated DNM2 Hb−, which did not express either HpuAB or HmbR. Hb+ variants of DNM2Hb− were selected and examined for HpuAB and HmbR expression. In each instance, acquisition of HpuAB or HmbR expression was correlated with phase variation in the poly(G) tract of each Hb receptor.

    Description / Table of Contents: Zusammenfassung Die Maillard- oder Bräunungsreaktion genannten Umsetzungen zwischen reduzierenden Zuckern und Eiweiß führen zur chemischen Zerstörung der Aminosäuren und zum Verlust der Proteinqualität während der Lebensmittelbearbeitung und -lagerung. Der vorliegende Beitrag zeigt Befunde auf, daß die Maillardreaktion auch im Gewebe des Menschen bei der Alterung von Proteinen mit langer biologischer Halbwertszeit auftritt. Die Konzentrationen an den sogenannten Amadori-Produkten, die im Initialstadium der Maillardreaktion aus Glucose und den Proteinen der Augenlinse oder dem Kollagen der Haut entstehen, erwiesen sich als relativ konstant, auch mit zunehmendem Alter. Die Produkte der Glycosylierung und nachfolgenden Oxidation der Proteine, auch Glycoxidationsprodukte genannt, häufen sich dagegen im Alter an, und zwar bei Diabetikern in vermehrtem Maße. Zu diesen Produkten gehören die Aminosäurenderivate N-(carboxymethyl)-lysin (CML), N-(carboxymethyl)-hydroxylysin (CMhL) sowie das fluoreszierende Quervernetzungsprodukt Pentosidin. Während diese Glycoxidationsprodukte in den Körpergeweben nur in Spuren vorkommen, gibt es deutliche Hinweise auf die Anwesenheit weiterer Bräunungsprodukte, deren Charakterisierung jedoch noch aussteht. Es werden Möglichkeiten zur „Entgiftung“ der reaktiven Zwischenprodukte aus der Maillardreaktion sowie zum Abbau extrem gebräunter Proteine diskutiert sowie neuere Möglichkeiten zur therapeutischen Modulierung fortgeschrittener Stadien der Maillardreaktion aufgezeigt.
    Notes: Summary The Maillard or browning reaction between reducing sugars and protein contributes to the chemical deterioration and loss of nutritional value of proteins during food processing and storage. This article presents and discusses evidence that the Maillard reaction is also involved in the chemical aging of long-lived proteins in human tissues. While the concentration of the Amadori adduct of glucose to lens protein and skin collagen is relatively constant with age, products of sequential glycation and oxidation of protein, termed glycoxidation products, accumulate in these long-lived proteins with advancing age and at an accelerated rate in diabetes. Among these products are the chemically modified amino acids, Nɛ-(carboxymethyl)lysine (CML), Nɛ-(carboxymethyl)hydroxylysine (CMhL), and the fluorescent crosslink, pentosidine. While these glycoxidation products are present at only trace levels in tissue proteins, there is strong evidence for the presence of other browning products which remain to be characterized. Mechanisms for detoxifying reactive intermediates in the Maillard reaction and catabolism of extensively browned proteins are also discussed, along with recent approaches for therapeutic modulation of advanced stages of the Maillard reaction.

    Notes: Abstract  In soybean [Glycine max (L.) Merr.] heterosis has been reported for seed yield. Molecular markers may be useful to select diverse parents for the expression of heterosis and yield improvement. The objective of this study was to determine if molecular markers could be used to predict yield heterosis in soybean. From each Maturity Group (MG) II and III, 21 genotypes were selected on the basis of high yield (HY), different geographic origin (GO), and isozyme loci (ISO) and for diversity in restriction fragment length polymorphisms (RFLP), and crosses were made within MGs and selection criteria groups to obtain 6 F1 hybrids per group. The 21 parents and the 24 F1 hybrids of each MG were evaluated for yield in replicated tests at two locations in 2 years, and midparent heterosis (MPH) and high-parent heterosis (HPH) estimates were calculated. On the basis of hybrid performance during the first year, 12 parents (3 per selection criteria group) were chosen in each MG to conduct a second RFLP analysis using 129 probes. Genetic distances (GDM) for pairs of the 12 genotypes were calculated with this RFLP information and correlated with MPH and HPH estimates. Significant MPH averages for seed yield were observed in the combined analysis of variance in each of the four selection criteria groups of MG II, and in the HY, ISO, and GO of MG III. Significant HPH averages were observed only in the ISO and GO groups of MG II. The greatest frequency of F1 hybrids with significant MPH was observed in the ISO and GO groups of both MGs. For HPH, the greatest frequency was observed in the ISO group of both MGs. In both MGs, the ISO group had the largest absolute MPH value; the RFLP group had generally the smallest. The observations indicated that the expression of heterosis in seed yield might be associated with diversity in the isozyme loci present in the parents. For the genotypes included in the second RFLP analysis, correlations of GDMs with MPH and HPH values on an entry-mean basis were low and not significant, indicating that heterosis in yield may not be associated with genetic diversity at the molecular level as determined by RFLPs. The results suggest that in soybean, parent selection on the basis of RFLPs and isozyme loci to exploit heterosis in seed yield may not be feasible. There was no association between genetic distance estimated by the RFLP analysis and seed yield heterosis, and in spite of the observed relationship between isozyme loci and heterosis for yield, the practicality of using the isozyme markers to select parents may be limited because of the reduced number of assayable isozyme loci in soybean.
