    Description: In the aftermath of a flood, fire ants, Solenopsis invicta , cluster into temporary encampments. The encampments can contain hundreds of thousands of ants and reach over 30 ants high. How do ants build such tall structures without being crushed? In this combined experimental and theoretical study, we investigate the shape and rate of construction of ant towers around a central support. The towers are bell shaped, consistent with towers of constant strength such as the Eiffel tower, where each element bears an equal load. However, unlike the Eiffel tower, the ant tower is built through a process of trial and error, whereby failed portions avalanche until the final shape emerges. High-speed and novel X-ray videography reveal that the tower constantly sinks and is rebuilt, reminiscent of large multicellular systems such as human skin. We combine the behavioural rules that produce rafts on water with measurements of adhesion and attachment strength to model the rate of growth of the tower. The model correctly predicts that the growth rate decreases as the support diameter increases. This work may inspire the design of synthetic swarms capable of building in vertical layers.

    Description: The hot cathode ion source will tend to be unstable when operated with high power and long pulse. In order to achieve stable operation, a new regulation method based on the arc power (discharge power) feedback control was designed and tested on the hot cathode ion source test bed with arc discharge and beam extraction. The results show that the new regulation method can achieve stable arc discharge and beam extraction. It verifies the success of feedback control of arc source with arc power.

    Description: Single cell RNA-seq experiments provide valuable insight into cellular heterogeneity but suffer from low coverage, 3' bias and technical noise. These unique properties of single cell RNA-seq data make study of alternative splicing difficult, and thus most single cell studies have restricted analysis of transcriptome variation to the gene level. To address these limitations, we developed SingleSplice, which uses a statistical model to detect genes whose isoform usage shows biological variation significantly exceeding technical noise in a population of single cells. Importantly, SingleSplice is tailored to the unique demands of single cell analysis, detecting isoform usage differences without attempting to infer expression levels for full-length transcripts. Using data from spike-in transcripts, we found that our approach detects variation in isoform usage among single cells with high sensitivity and specificity. We also applied SingleSplice to data from mouse embryonic stem cells and discovered a set of genes that show significant biological variation in isoform usage across the set of cells. A subset of these isoform differences are linked to cell cycle stage, suggesting a novel connection between alternative splicing and the cell cycle.

    Description: A new CO 2 cryogenic capture and liquefaction system has been proposed previously in order to separate CO 2 from exhausted gases and make it as a resource for industry. This system combines CO 2 cryogenic capture with N 2 /O 2 separation together. Its energy consumption is lower than the traditional amine solution capture process as theoretical analysis. In this study, the simulation of the proposed system with several improvements was carried out aiming to reduce the energy consumption further. Many heat exchangers were introduced and the heat exchanger arrangements were optimized to recycle the refrigeration capacity from the returned N 2 after the N 2 /O 2 separation. The discharge pressure of mixture gas from the compressor was reduced from 10 to 3.493 MPa. The simulation results showed that the compression work could be greatly reduced and the energy consumption of CO 2 capture in this new system after these improvements reached 2.884 GJ/ton CO 2 . The new system is promising because not only liquid or solid CO 2 could be produced but also N 2 and O 2 could be separated.

    Description: The charge trap memory capacitors incorporating a stacked charge trapping layer consisting of double layered ZrO 2 nanocrystals (NCs) and inserted Al 2 O 3 have been fabricated and investigated. It is observed that the memory capacitor with stacked trapping layer exhibits a hysteresis window as large as 14.3 V for ±10 V sweeping gate voltage range, faster program/erase speed, improved endurance performance, and good data retention characteristics with smaller extrapolated ten years charge loss at room temperature and 125 °C compared to single layered NCs. The special energy band alignment and the introduced additional traps of double layered ZrO 2 NCs and inserted Al 2 O 3 change the trapping and loss behavior of charges, and jointly contribute to the remarkable memory characteristics. Therefore, the memory capacitor with a stacked charge trapping layer is a promising candidate in future nonvolatile charge trap memory device design and application.

    Description: Single-well imaging has been a technique increasingly used in the detection of near-borehole geologic structures. The azimuth of a geologic structure, however, cannot be uniquely determined with acoustic signals recorded in the borehole alone, due to the azimuth ambiguity existing in current imaging techniques. We eliminated such ambiguity by revealing the relevant acoustic principle underlying the P-wave reflection behavior. When a P-wave excited by a transducer in the logging tool impinges upon a planar interface, the P-wave reflection coefficient (RC) of the displacement is opposite in sign to that of the normal stress or fluid pressure, regardless of the incident angle and the parameters of the media on the two sides. The derived relation about signs of RCs was validated by finite-difference solutions for reflected waves from a near-borehole plane fault. With this newly discovered reflection principle, one can eliminate the azimuth ambiguity of any interface outside a borehole by checking if the waveforms of pressure and the displacement component are both changed in polarity after reflection. Furthermore, because the pressure and displacement are observable quantities and the waveform of the acoustic source is known in acoustic logging, it is convenient to implement the data acquisition for this technique, which is a major advantage over other techniques. We expounded and exemplified our new technique by numerical simulation.

    Description: Neutral beam injection has been recognized as one of the most effective means for plasma heating. According to the research plan of the EAST physics experiment, two sets of neutral beam injector (NBI) were built and operational in 2014. The paper presents the development of beam diagnosis system for EAST NBI and the latest experiment results obtained on the test-stand and EAST-NBI-1 and 2. The results show that the optimal divergence angle is (0.62°, 1.57°) and the full energy particle is up to 77%. They indicate that EAST NBI work properly and all targets reach or almost reach the design targets. All these lay a solid foundation for the achievement of high quality plasma heating for EAST.

    Description: Hammerhead ribozymes are self-cleaving RNA molecules capable of regulating gene expression in living cells. Their cleavage performance is strongly influenced by intra-molecular loop–loop interactions, a feature not readily accessible through modern prediction algorithms. Ribozyme engineering and efficient implementation of ribozyme-based genetic switches requires detailed knowledge of individual self-cleavage performances. By rational design, we devised fluorescent aptamer-ribozyme RNA architectures that allow for the real-time measurement of ribozyme self-cleavage activity in vitro . The engineered nucleic acid molecules implement a split Spinach aptamer sequence that is made accessible for strand displacement upon ribozyme self-cleavage, thereby complementing the fluorescent Spinach aptamer. This fully RNA-based ribozyme performance assay correlates ribozyme cleavage activity with Spinach fluorescence to provide a rapid and straightforward technology for the validation of loop–loop interactions in hammerhead ribozymes.
