    Description: Mitogenic stimulation of mouse lymphocytes results in two sequential intracellular alkalinizations. The first shift of intracellular pH from 7.18 to 7.35 coincides with early biochemical events following mitogenic stimulation. The second alkalinization begins 12 hours after stimulation and rises in parallel with the rate of thymidine incorporation.     Notes: Abstract The contact angle,θ, formed by a liquid on a solid surface in air depends on the solid-air (γ S ), liquid-air (γ L ) and solid-liquid (γ SL ) interfacial free energies, as described by Young's equation. Critical examination of reported contact angles for numerous liquids and solids leads to an empirical correlation betweenγ sL and bothγ Y andγ S . Combination of this correlation with Young's equation gives an empirical relation allowing calculation ofγ S fromγ L andθ Calculations made with these empirical relations agree well with estimations ofγ S obtained by the method of critical spreading, and are consistent with Young's equation.

    Notes: Summary The effect of dissolved carbon dioxide concentration in the anaerobic growth of Escherichia coli was investigated. E. coli was grown anaerobically with the dissolved CO2 concentration controlled over the range from 8x10-6 M to 3.7x10-2 M in the liquid phase. The maximum specific growth rate was 0.75h-1 at 1.3x10-3 M CO2 and the maximum yield of cells on glucose was 0.32 at 1.75x10-4 M CO2. The maximum specific growth rate occurs close to the concentration of CO2 prevalent in the mammalian gut where E. coli naturally resides.

    Notes: Abstract Four species of Corynebacterium-C. fasciens, C. hydrocarboclastus, C. lepus and C. xerosis, were compared for growth and surfactant production from kerosene. Nitrate as sole nitrogen source resulted in a sharp peak in surfactant concentration during mid-exponential growth phase. Ammonium plus nitrate resulted in growth-associated surfactant production. Surfactant was produced at a concentration of 50–80 times the critical micelle concentration by these species; greatest concentrations were produced by C. lepus. All species were pleomorphic, with greatest cell length occurring in early exponential phase. C. hydrocarboclastus had distinctly longer cells (5 μ) than the other species (3 μ) when grown in shake-flask culture. Ammonium sulfate at concentrations greater than 0.1 % (w/v) increased cell length in stationary phase.

    Notes: A new source of surface-active compounds produced by microbes was investigated. These biologically derived surfactants (biosurfactants) were found to be extracellular products of two newly isolated Corynebacterium species, which were gorwn on kerosene (Imperial Oil No.9). Batch-growth kinetic studies were carried out in a 14 liter fermentor and the production of biosurfactants was found to be cell growth associated. The surface tension of the whole microbial broth was reduced to a minimum of about 30 dyn/cm, as compared with the surface tension for distilled water of 72 dyn/cm. The crude biosurfactants were recovered from the cell-free broth, freeze-dried, redissolved in water, and their surface-active properties were studied. The biosurfactants reported here were found to be nontoxic and their ability to lower the surface tension of water was found to be comparable to that of sodium dodecylbenzene sulfonate, common commercial synthetic surfactant.

    Notes: A Corynebacterium hydrocarboclastus culture isolated in our laboratory (see, Zajic and Knettig, Developments in Industrial Microbiology, 1971, p. 87) has been shown to produce an extracellular biopolymer with emulsifying properties when grown on a mixture of linear hydrocarbons. This microorganism was found to grow well on a variety of carbohydrates and hydrocarbons. However, the best substrates were pure linear hydrocarbons and particularly, n-C12, n-C13, and n-C14. The substrates supporting good growth gave good polymer production. Maximum cell mass of 10-11 g/liter and a maximum amount of polymer of 5-6 g/liter were recorded. The polymers recovered from the different substrates were found to be complex molecules or mixtures with a protein, a lipid, and a carbohydrate moiety. All the polymers are surface active and have two critical micelle concentrations.

    Notes: A control system has been devised for the maintenance of stable ammonium concentrations throughout a fedbatch fermentation. The control system is based on an ammonia gas-sensing electrode that monitors a pH-adjusted effluent stream from the fermentor. To overcome the time lag between the fermentor and the electrode, feedback control included metered flows of ammonium to both the fermentor and the electrode vessel. The system was used to study the growth of Escherichia coli B (ATCC 11303) at controlled ammonium concentrations of 5 to 200mM. Apparent specific growth rates, biomass and protein production, and glucose yields were essentially constant from 5 to 170mM. Above 170mM ammonium growth was inhibited. As ammonium concentration decreased from 170 to 5mM, ammonium yields increased from 1 to 24 g cell dry wt/g ammonium utilized. The results demonstrate that control of ammonium concentrations at levels so low that ammonium would be exhausted in batch fermentations can significantly increase overall ammonium yields.

    Notes: An extracellular polymer was produced by continuous fermentation of Corynebacterium hydrocarboclastus on kerosene in a 24 liter reactor. This polymer was composed of protein, lipid, and carbohydrates. The polymer possesed surface active properties, and had two critical micelle concentrations. Its effectiveness was quite comparable to the effectiveness of synthetic surface active agents such as Tween 80 and Span 20; however, its efficiency was much lower. The polymer also had emulsifying properties. Maximum emulsification was obtained at pH 6. The emulsifying properties were unaffected by high salt concentration [up to 5% (w/v) in Na+], and tolerated a water hardness up to 5,000 ppm. A 2 hr treatment of the polymer at temperatures higher than 65°C resulted in a loss of its emulsifying properties. Two microorganisms, named SLYS and Y, isolated from soil, were able to grow on the polymer as sole carbon and energy source, thus proving its biodegradability. SLYS was tentatively identified as Flavobacterium breve and Y as Flavobacterium devorans.
