Notes: Amorphous silicon containing high concentrations of H was produced by Si evaporation and the simultaneous addition of H from rf discharge. The H content in the films is approximately 60–70% of that found in glow-discharge produced amorphous Si containing H. The optical gap was found to be 1.7 eV. Effectiveness in the reduction of the dangling bond density has been determined by electron spin resonance. It was found that the Si–H bond was primarily formed in the gaseous phase and subsequently condenses into the solid phase. Thermal annealing and infrared spectra indicate that films produced in this manner result in improved thermal stability when compared to samples produced by glow discharge.

Notes: Charge transient measurements have been used to study deep levels in n-type CdTe single crystals. The samples, including In-doped, undoped, and Sn-doped single crystals, were grown by the Bridgman or the travelling heater method (THM). The temperature dependence of the capture cross section was investigated under the partial filling conditions. The activation energies of capture cross section were determined. For In-doped samples, the results indicate both levels Ec −0.66 eV (Bridgman samples) and Ec −0.68 eV (THM samples) are associated with the same indium-defect complex, but levels Ec −0.34 eV (Bridgman) and Ec −0.32 eV (THM) exhibited different capture mechanisms. The unusual capture properties of the trap at Ec −0.61 eV in the undoped sample can be attributed to a doubly charged Cd interstitial. For Sn-doped samples, four levels were observed, and levels Ec −0.89 eV and Ec −0.43 eV can be related to the presence of tin.

Notes: During cell division in Gram-negative bacteria, the cell envelope invaginates and constricts at the septum, eventually severing the cell into two compartments, and separating the replicated genetic materials. In Escherichia coli, at least nine essential gene products participate directly in septum formation: FtsA, FtsI, FtsL, FtsK, FtsN, FtsQ, FtsW, FtsZ and ZipA. All nine proteins have been localized to the septal ring, an equatorial ring structure at the division site. We used translational fusions to green fluorescent protein (GFP) to demonstrate that FtsQ, FtsL and FtsI localize to potential division sites in filamentous cells depleted of FtsN, but not in those depleted of FtsK. We also constructed translational fusions of FtsZ, FtsA, FtsQ, FtsL and FtsI to enhanced cyan or yellow fluorescent protein (ECFP or EYFP respectively), GFP variants with different fluorescence spectra. Examination of cells expressing different combinations of the fusions indicated that FtsA, FtsQ, FtsL and FtsI co-localize with FtsZ in filaments depleted of FtsN. These localization results support the model that E. coli cell division proteins assemble sequentially as a multimeric complex at the division site: first FtsZ, then FtsA and ZipA independently of each other, followed successively by FtsK, FtsQ, FtsL, FtsW, FtsI and FtsN.

Notes: In Escherichia coli, nine gene products are known to be essential for assembly of the division septum. One of these, FtsL, is a bitopic membrane protein whose precise function is not understood. Here we use fluorescence microscopy to study the subcellular localization of FtsL, both in a wild-type strain and in a merodiploid strain that expresses a GFP–FtsL fusion protein. We show that FtsL localizes to the cell septum where it forms a ring analogous to the cytoplasmic FtsZ ring. FtsL localization is dependent upon the function of FtsZ, FtsA and FtsQ, but not FtsI. In a reverse approach, we use fusions of green fluorescent protein (GFP) to FtsZ, FtsA and ZipA to show that these proteins localize to the division site in an FtsL-independent fashion. We propose that FtsL is a relatively late recruit to the ring structure that mediates septation.

Notes: A sulphated glycosaminoglycan-dependent mechanism of microbial infection for mammailan cells was characterized for the Chlamydia trachomatis trachoma and lymphogranuloma venereum (LGV) biovars. We demonstrated that the trachoma and LGV biovars compete for the same receptor(s) on host cells and that their infectivity was inhibited by heparin or heparan sulphate. Using a specific heparan suiphate lyase (heparitinase) to treat organisms, the Infectivity of both biovars was abolished. Furthermore, exogenous heparan sulphate rescued chlamydial infectivity following treatment with heparitinase and the restored infectivity was neutralized by an anti-heparan sulphate monoclonal antibody. These data suggest that heparan sulphate-like-mediated Interactions between C. trachomatis and eukaryotic cells are essential for infectivity.

Notes: Caulobacter crescentus assembles many of its cellular machines at distinct times and locations during the cell cycle. PodJ provides the spatial cues for the biogenesis of several polar organelles, including the pili, adhesive holdfast and chemotactic apparatus, by recruiting structural and regulatory proteins, such as CpaE and PleC, to a specific cell pole. PodJ is a protein with a single transmembrane domain that exists in two forms, full-length (PodJL) and truncated (PodJS), each appearing during a specific time period of the cell cycle to control different aspects of polar organelle development. PodJL is synthesized in the early predivisional cell and is later proteolytically converted to PodJS. During the swarmer-to-stalked transition, PodJS must be degraded to preserve asymmetry in the next cell cycle. We found that MmpA facilitates the degradation of PodJS. MmpA belongs to the site-2 protease (S2P) family of membrane-embedded zinc metalloproteases, which includes SpoIVFB and YluC of Bacillus subtilis and YaeL of Escherichia coli. MmpA appears to cleave within or near the transmembrane segment of PodJS, releasing it into the cytoplasm for complete proteolysis. While PodJS has a specific temporal and spatial address, MmpA is present throughout the cell cycle; furthermore, periplasmic fusion to mRFP1 suggested that MmpA is uniformly distributed around the cell. We also determined that mmpA and yaeL can complement each other in C. crescentus and E. coli, indicating functional conservation. Thus, the sequential degradation of PodJ appears to involve regulated intramembrane proteolysis (Rip) by MmpA.

Notes: This study investigated students' perceptions of work values and occupational choice propensity. A total of 311 hospitality majors at four different universities in Taiwan were analyzed in the study. The work value inventory (WVI) scale, containing 45 attributes, developed by Super, was used to assess students' perceptions of work values. The study found "supervisory relations", "work surroundings", and "way of life" were the most important work values among respondents. Three mutually exclusive clusters of respondents were identified. This research suggests a longitudinal study measuring students' perceptions of work value before and after entering the workforce.
