    Notes: Abstract Gamma-radiation-induced chromosomal interchanges in two inbred lines of rye were analyzed by using the C-banding technique. Neither the doses of radiation nor the time elapsed between radiation and cytological observation influenced the contribution of individual chromosomes to the observed interchanges. Dicentric are more frequent than monocentric interchanges. Neither the individual chromosome contribution to interchanges nor the chromosome combinations fit a random distribution. Chromosomes 6(5R), 4(4R) and 7(1R) of the inbred line Pool and 6(5R), 5(6R) and 4(4R) of line Riodeva, which are carriers of large amounts of heterochromatin, are much more frequently involved in interchanges than expected. Likewise, they are involved in the most frequent combinations observed (6-4, 6-7, 4-5, 4-7 and 6-3 (2R) in line Pool and 6-5, 6-4, 4-5 and 6-7 in line Riodeva). The role of C-heterochromatin in radiation-induced chromosomal interchanges is discussed.

    Notes: Abstract In order to investigate wheter colchicine affects crossing-over, rye anthers of an inbred line of rye forming bridges and fragments at anaphase I produced by erroneous chiasmata, and anthers of plants heterozygous for a conspicuous heterochromatin band, were cultured in a medium with colchicine. Anthers planted at zygotene did not show bridges at AI in the inbred line. In the heterozygotes no difference between associated chromatids in respect to the heterochromatin band, resulting from crossingover, were observed. In anthers planted at pachytene both bridges and chromosomes showing difference between associated chromatids were observed at a stage equivalent to AI with the same frequency as in anaphase I cells of untreated anthers. This demonstrates that crossing-over or a prerequisite to crossing-over is established at zygotene, and also that absence of chiasmate association at later stages is not due to precocious slipping off of chiasmata.

    Notes: Abstract Nucleolar activity was analyzed in wheat (Triticum sp.), rye (Secale cereale) and several types of wheat-rye derivatives using a modified, highly reproducible, silver staining procedure (Lacadena et al. 1984). A comparative analysis of the nucleolar organizer regions (NORs) of somatic metaphase chromosomes was made by phase contrast, C-banding, and silver staining. The frequency distribution of the number of nucleoli visualized at interphase by silver staining was also used to infer the activity of NORs. The results agree quite well with data from in situ hybridization reported by other authors. The behavior of euploid, ditelosomic and nulli-tetrasomic plants of common wheat showed the relative nucleolar activity of the four organizer chromosomes to be: 6B 〉 1B 〉 5D 〉 1A. — Several types of wheat-rye derivatives were analyzed: interspecific hybrid, triticale, addition and substitution lines, and plants with the genome constitutions, AABBDR, ABDR + 5D, ABRR, and ABRRR. In all cases the nucleolar organizer chromosome 1R of rye was suppressed by the presence of wheat chromosomes.

    Notes: Summary Amphiplasty in hexaploid triticale, the artificial amphiploid of tetraploid wheat and diploid rye, is analyzed for the first time using a modified, highly reproducible, silver-staining procedure. A comparative analysis of metaphase somatic cells by phase contrast, C-banding and silver-staining of the hexaploid triticale cv. ‘Cachirulo’ and its parents, namely, the tetraploid durum wheat cv. ‘Enano de Andujar’ and the diploid rye cv. ‘Petkus’ has been made. Two silver-stained nucleolar organizer regions (Ag-NORs) (the chromosome pair 1 R) are observed in all rye plants analyzed, whereas four Ag-NORs (chromosome pairs 1 B and 6 B) are found both in the tetraploid wheat parent and in the triticale. The rye Ag-NORs are absent in the triticale. Since the Agstaining reaction of NORs can be considered as an indication for genetic activity, the silver procedure can be used to visualize gene functionality at the rDNA sites with conventional light microscopy and, consequently, the modified Ag-staining method described can be very useful in analyzing the amphiplasty phenomenon in natural or artificial hybrid combinations and derivatives in the Triticum group and its relatives.

    Notes: Summary The nucleolar organizer activity of wheat (Triticum aestivum, AABBDD) and Aegilops umbellulata (UU) chromosomes have been analyzed in the complete set of the chromosome addition lines by using a highly reproducible silver-staining procedure. Chromosomes 1U and 5U produce the partial inactivation of wheat nucleolar organizer chromosomes 6B, 1B and 5D. The chromosomes D and G from Ae. umbellulata, which are not SAT-chromosomes, seem to specifically influence the activity of wheat NORs. The predominant status of the U genome with respect to nucleolar competition in the Triticeae is confirmed.

    Notes: Summary C-banding patterns and nucleolar activity were analyzed in Dasypyrum villosum, its added chromosomes to hexaploid wheat and the hexaploid amphiploid Triticum dicoccum-D. villosum. Two different populations of the allogamous species D. villosum (2n= 14, VV) from Greece and Italy were analyzed showing a similar polymorphism for C-banding pattern. Six of the seven addition lines were identified by their characteristic C-banding pattern. No polymorphism between both members of each added alien chromosome was found. Furthermore, nucleolar activity and competition were studied by using silver staining procedure. In D. villosum only one chromosome pair, A, was found to be responsible for organizing nucleoli. The results obtained in the amphiploid and in the addition lines demonstrate that nucleolar activity is restricted to SAT-chromosomes 1B and 6B of wheat, while those of D. villosum remain inactive.

    Notes: Summary Meiotic chromosome pairing and Giemsa C-banding analyses in crosses of several European blue-grained wheat strains with Chinese Spring double ditelosomic and other aneuploid lines showed that Triticum aestivum Blaukorn strains “Berlin,” “Probstdorf,” “Tschermak,” and “Weihenstephan” are chromosome substitutions, in which the complete wheat chromosome 4A pair is replaced, whereas the strains “Brünn” and “Moskau” are 4B substitutions. The alien chromosome pair in all of these strains is an A genome chromosome (4A) from diploid Triticum monococcum or T. boeoticum not present in common tetraploid and hexaploid cultivated wheats. The Blaukorn strain Weihenstephan “W 70a86” possesses, in addition to a rye chromosome pair 5R compensating for the loss of part of chromosome 5D, a 4A/5DL translocation replacing chromosome pair 4B of wheat.

    Notes: Summary The nucleolar organizer activity of the Agropyron elongatum, its amphiploid with hexaploid wheat (Triticum aestivum) and the chromosome addition lines is analyzed by the silver-staining procedure. Four Ag-NORs are observed in A. elongatum corresponding to the chromosomes 6E and 7E. In the amphiploid T. aestivum — A. elongatum, eight Ag-NORs are observed which corresponds the wheat chromosomes 1B and 6B and to the elongatum chromosomes 6E and 7E. Thus, there is codominance in the nucleolar organizer activity of the chromosomes of the two species. However, a partial amphiplasty is detected since less than 8 Ag-NORs (7 up to 4) are observed in some metaphase cells; the chromosomes 6E and 7E are occasionally suppressed by wheat chromosomes. This conclusion is confirmed by the behaviour of the addition lines since only in those corresponding to the chromosomes 6E and 7E are the elongatum chromosomes nucleolar active although occasionally they can be suppressed by wheat chromosomes.

    Notes: Summary Meiotic pairing frequencies of the Un and D genomes of Ae. ventricosa and the R of S. cereale could be easily established at metaphase I in Aegilops ventricosa — Secale cereale amphiploid plants as well as in its parental species by using the C-banding technique procedure. The results show a high diminution of chromosome pairing for all genomes in the amphiploid with respect to its parental species probably due to C-heterochromatin content and/or genotypic or cryptic interactions between the three genomes.

    Notes: Summary The Ae. ventricosa and S. cereale genomes were distinguished at meiosis by the C-banding procedure. Only two plants of the amphiploid Ae. ventricosa-S. cereale were found to exhibit the high degree of asynapsis limited to Aegilops ventricosa genomes. In addition, these genomes showed higher homoeologous pairing than homologous pairing frequencies. These results can be explained by the existence of separate genome-specific control of meiotic pairing between the chromosomes of both species in these synaptic mutant plants.
