    Notes: The human pathogenic amoeboflagellate Naegleria fowleri and the nonpathogenic species N. gruberi can be cultivated axenically but usually in different media. Naegleria fowleri 6088 has been adapted to grow in Balamuth H-4 medium, usually used to propagate N. gruberi nB81. and nB81 has been adapted to grow in supplemented Nelson's medium, usually used to propagate N. fowleri. N. gruberi nB81. grown in either medium, enflagellated 135 to 150 min after subculture to non-nutrient amoeba saline, whereas 6088 required 225 min. Naegleria gruberi nB81 grown in either medium was agglutinated by 100 ug concanavalin A/ml, whereas N. fowleri 6088 was not. Naegleria fowleri and N. gruberi grown in Nelson's medium became rounded to a greater extent upon chilling at 5° C and remained rounded longer than Naegleria grown in Balamuth medium. The specificity of the surface antigens was an inherent characteristic of each species and not dependent upon the propagating medium. but Naegleria grown in Nelson's medium was agglutinated more reproducibly and more effectively by antiserum. N. gruberi was somewhat more resistant to acriflavine, actinomycin D, cycloheximide, or tetracycline than N. fowleri, regardless of the culture medium. Naegleria fowleri 6088 grown in Nelson's medium, however, was more resistant to actinomycin D, daunomycin. mithramycin. sulfamethoxazole, or tyrocidine than 6088 grown in Balamuth medium. There are limitations on the validity of comparisons of N. fowleri and N. gruberi based upon cultures grown in different media.

    Notes: Abstract DNA-mediated gene transfer was used to introduce DNA from a methotrexate-resistant mouse fibroblast cell line into mouse bone marrow cells. This cell line contained a methotrexate-resistant dihydrofolate reductase, active at 10−4 M methotrexate, which was electrophoretically separable from the wild-type mouse enzyme. Transformed hematopoietic cells were returned to irradiated mice and selected in vivo by methotrexate administration. Some recipients of transformed marrow cells expressed the electrophoretically distinct, methotrexate-resistant dihydrofolate reductase in hematopoietic cells. These observations suggest that successful transformation of marrow stem cells to methotrexate resistance is accomplished by insertion of a dihydrofolate reductase gene coding for a mutant enzyme that is highly resistant to methotrexate.

    Notes: Abstract Shoot inversion induction of ethylene production was found in inverted shoots of corn, peas, soybean, sunflower, tomato, andPharbitis nil. The increases in ethylene production were found to range from two- to threefold in soybean to eightfold in corn and sunflower. The occurrence of peaks of ethylene production ranged from 16 h following shoot inversion in corn to 72 h inPharbitis. That the enhanced ethylene production was due to activation of 1-aminocyclopropane-1-carboxylic acid (ACC) synthase is supported by the finding of increased ACC content in inverted shoots of all species tested. Shoot inversion inhibition of elongation was found in inverted shoots of pea, soybean, sunflower, tomato, andPharbitis nil. This inhibition is thought to be mediated via increased ethylene production in the inverted shoots. That shoot inversion induction of ethylene is not a persistent effect is supported by the finding that ethylene synthesis could be terminated by reorientation of shoots to the upright position and could be reinitiated by the subsequent inversion of the shoots. The effects of shoot inversion on the enhancement of ethylene production and on the inhibition of elongation of the inverted shoot appear to be general phenomena.

    Notes: A numerical study was performed to investigate chemically reactive flows with sprays inside a staged turbine combustor (STC) using a modified version of the KIVA-II code. This STC consists of a fuel nozzle (FN), a rich-burn (RB) zone, a converging connecting pipe, a quick-quench (QQ) zone, a diverging connecting pipe and a lean-combustion (LC) zone. From the computational viewpoint, it is more efficient to split the STC into two subsystems, called FN/RB zone and QQ/LC zones, and the numerical solutions were obtained separately for each subsystem. This paper addresses the numerical results of the STC which is equipped with an advanced airblast fuel nozzle. The airblast nozzle has two fuel injection passages and four air flow passages. The input conditions used in this study were chosen similar to those encountered in advanced combustion systems. Preliminary results generated illustrate some of the major features of the flow and temperature fields inside the STC. Velocity, temperature and some critical species information inside the FN/RB zone are given. Formation of the co- and counter-rotating bulk flow and the sandwiched-ring-shaped temperature field, typical of the confined inclined jet-in-cross-flow, can be seen clearly in the QQ/LC zones. The calculations of the mass-weighted standard deviation and the pattern factor of temperature revealed that the mixing performance of the STC is very promising. The temperature of the fluid leaving the LC zone is very uniform. Prediction of the NOx emission shows that there is no excessive thermal NOx produced in the QQ/LC zones for the case studied. From the results obtained so far, it appears that the modified KIVA-II code can be used to guide the low-emission combustion experiments.

    Description: Molecular mechanisms responsible for the clinical progression of chronic myelocytic leukemia to its accelerated phase or to blast crisis have not been defined. We found alterations of the p53 gene (p53 is a 53-kDa nuclear protein) including deletions and rearrangements in 8 of 34 patients in blast crisis and 1 of 4 patients in the accelerated phase, but in only 1 of 38 patients in the chronic phase of chronic myelocytic leukemia. Only two other examples of p53 gene alterations were found among 203 patients with hematologic malignancies and solid tumors. Transcripts of the p53 gene were uniformly found in chronic-phase cells, but gene expression was variable in blast crisis, and transcripts were reduced or undetectable in 10 of 16 patients. Heterogeneous alterations in the structure and expression of the p53 gene appear to be relatively frequent in blast crisis and may be involved in the evolution of disease.
