    Notes: . The ultrastructure of the cortex beneath the fission furrow of dividing Stentor coeruleus was examined using scanning and transmission electron microscopy. During division, basal bodies, axonemes, and km fibers beneath the furrow were absorbed near the moving primordial oral apparatus, and a circumferential band of microtubules and filaments was formed at the base of the furrow. The location and orientation of this fibrous band suggest that it may be an important component of the cytokinetic machinery. Treatment with vinblastine sulfate (4 × 10-5 M) disrupted the circumferential microtubules and blocked division, which is consistent with this hypothesis.

    Notes: SYNOPSIS. Sexual reproduction of up to 50% of the cells from 9 cultures of Stentor coeruleus was observed repeatedly for several months. Photomicrographs were taken of mating pairs, and a number of new observations were made, including the ability of well fed cells (containing food vacuoles) to conjugate, the ability of a high percentage of cells (approaching 50%) in a culture to mate at one time, a more detailed description of the region of cell attachment as viewed from the frontal fields of the partner cells, and the ability of one partner to twist off and leave its oral area attached to the other member of the pair.

    Notes: SYNOPSIS. Oral regeneration, oral reorganization and cell division in the ciliate Stentor coeruleus can be temporarily blocked at any stage of development by exposure to colchicine. The implications of this observation, presented as an assumption that colchicine exerts its effects by specifically preventing the formation of microtubular elements in the cells, suggest that microtubules are essentially involved in many aspects of these complicated processes of cellular differentiation.

    Notes: To elucidate further the molecular events required for cytodifferentiation in Stentor coeruleus, the effects of several chemical metabolic inhibitors were tested on the outgrowth in situ of the membranellar cilia of the oral feeding organelle. The chemicals used included several inhibitors of cytoplasmic and mitochondrial protein synthesis (cycloheximide, emetine, and chloramphenicol), and an antimitotic agent (colchicine). Ciliary growth was affected only by colchicine, suggesting that a pool of “ciliary proteins” exists in interphase Stentor of sufficient size to permit complete reformation of the membranellar cilia. The implication of these observations to an understanding of the more complicated process of oral regeneration is discussed.

    Notes: SYNOPSIS. Regeneration of missing feeding organelles in the ciliate Stentor coeruleus can be delayed by ultraviolet (UV) irradiation. In an attempt to identify the radiation-sensitive cellular component(s), cells in various stages of regeneration were immobilized, flattened and irradiated with a microbeam of 265 nm UV light. Targets included regions of either the macronucleus or the cytoplasm at varying distances from the ramifying zone of the cortex, which is the region where the new oral organelles are formed. During the early stages of regeneration the macronucleus is more sensitive to UV-induced regeneration delay than the cytoplasm, but it is not possible to distinguish between direct and indirect effects. Damage to cortical primordia in middle or late stages leading to structural abnormalities appears to result only from direct effects.

    Notes: Abstract We present an extensive study of a new Monte Carlo acceleration algorithm introduced by Wolff for the Ising model. It differs from the Swendsen-Wang algorithm by growing and flipping single clusters at a random seed. In general, it is more efficient than Swendsen-Wang dynamics ford〉2, giving zero critical slowing down in the upper critical dimension. Monte Carlo simulations give dynamical critical exponentsz w=0.33±0.05 and 0.44+0.10 ind=2 and 3, respectively, and numbers consistent withz w=0 ind=4 and mean-field theory. We present scaling arguments which indicate that the Wolff mechanism for decorrelation differs substantially from Swendsen-Wang despite the apparent similarities of the two methods.
