    Description / Table of Contents: Zusammenfassung Zur Mengenbestimmung 14C-markierter Substanzen in einzelnen Zellen wird ein quantitativ-autoradiographisches Verfahren beschrieben. Hierbei kommen Referenzquellen aus 14C-Methacrylat zur Anwendung, die im Anschluß an die Exposition zur Wiederverwendung vom Autoradiogramm abgelöst werden können. Die Dicke der Referenzproben ist größer als die maximale β-Reichweite der 14C-Zerfälle in Methacrylat, und Länge und Breite betragen ein Vielfaches der maximalen β-Reichweite. Daher sind die Bedingungen von Strahlengeometrie, Rückstreuung und Selbstabsorption der β-Teilchen in Referenzquelle und markierten Einzelzellen verschieden. Ein autoradiographischer Vergleich ist aber durch rechnerische Berücksichtigung dieser unterschiedlichen Gegebenheiten möglich. Außerdem muß die Kernspuremulsion über Zellen und Referenzquelle eines Autoradiogramms eine gleichbleibende Dicke aufweisen. Einflüsse von positiver und negativer Chemographie, insbesondere ihre unterschiedliche Wirkung auf die Kernspuremulsion im Bereich von Zellen und der Referenzquelle, werden untersucht. Sie sind durch geeignete Fixation der Zellen und Entwicklung der Autoradiogramme vermeidbar. Der 14C-TdR-Einbau in einzelne Thymuszellen wird unter Anwendung der beschriebenen Methode gemessen und mit den Ergebnissen einer unabhängigen Methode verglichen. Dabei handelt es sich um die Herstellung und Auswertung von Bahnspur-Autoradiogrammen gleicher Zellen unter den Bedingungen einer 100 %-Geometrie. Die Methoden liefern gut übereinstimmende Ergebnisse.
    Notes: Summary A quantitative autoradiographic method is presented for determining amounts of 14C-labeled compounds in individual cells. Reference sources consisting of 14C-metharcrylate are used, which can be removed from the autoradiographs and reused after exposure has been terminated. The thickness of these sources exceeds the maximum β-range of 14C-disintegrations in methacrylate, while length and breadth exceed the maximum β-range many times over. Therefore, conditions of radiation geometry, of backscattering and β-selfabsorption differ in reference source and in individual labeled cells. Autoradiographic evaluations are comparable only if allowance is made for these differing conditions. In addition, the nuclear emulsion covering the cells and the reference source has to be of a constant thickness. Influences of positive or negative chemography are investigated with special attention to different effects on the nuclear emulsion which, on the one hand, covers the cells, and on the other, the reference source. They can be eliminated by appropriate procedures of fixation of the cells, and of the development of the autoradiographs. Using the described method, measurements of the incorporation of 14C-TdR into single cells are made. They are then compared with the results obtained by means of an independent method. This latter method consists basically in the preparation and evaluation of trackautoradiographs of the same cells under conditions of a 100% geometry. The results obtained from the two methods coincide well.

    Description / Table of Contents: Zusammenfassung Nach Hemmung der de novo-Synthese von TMP durch 5-F-UdR läuft die DNS-Synthese in vitro in Anwesenheit von 10−5 M/l mit unveränderter Geschwindigkeit ab. Dadurch ist die DNS-Synthese-Dauer über die quantitative autoradiographische Messung des 14C-TdR-Einbaus bestimmbar. Diese in vitro-Methode wird an Rattenknochenmark und Zellen einer CML geprüft. Erste Ergebnisse zeigen eine gute Übereinstimmung mit in vivo-Messungen durch andere Methoden.
    Notes: Summary Suspended haemopoietic cells are incubated in the presence of 5-fluoro-deoxyuridine (5-F-UdR) as an inhibitor of the de novo synthesis of thymidine monophosphate (TMP). The DNA synthesis rate remains undisturbed, if 10−5 M/l thymidine (TdR) is added. Thus, the DNA synthesis rate of individual cells can be determined by means of a quantitative autoradiographic method in measuring the incorporation rate of 14C-TdR into the DNA of the cells. DNA synthesis rates are then converted into DNA synthesis times. This in vitro method has been checked in cells of rat bone marrow and of peripheral blood in a case of chronic myelocytic leukaemia (CML). Preliminary results correlate well with in vivo estimations obtained by other methods.

    Notes: Abstract Ecological studies on rainforest in the central Amazon combine the analysis of environmental factors, which affect and are affected by the natural rainforest, and the nutrient balance of the forest biome. The natural rainforest is a highly sensitive climax system, whose quasi-steady state conditions are determined by the abundance and availability of micro- and macro-nutrients, the continuous but low flux of energy and bioelements through the various compartments, the pool of energy and nutrients in the standing crop of the rainforest and the multiple interactions and loops among species, matrices and environment. Large parts of the natural rainforest in the Amazon are being transferred to man-managed land-use schemes, mainly ranch land. The vast and uncontrolled clearing of the rainforest will partially disturb and in some cases destroy the potential of the area for long term natural and even man-managed ecosystems.
