    Notes: Some routine haematological methods for examining fish blood are described including haemoglobin estimation, haematocrit, erythrocyte counts, erythrocyte sedimentation rate, total and differential leukocyte counts, and cytochemical staining. Descriptions of stained blood cells are given as are the ranges and mean values for the above tests on brown trout Salmo trutta (L.). These methods are suggested as a possible means of assessing fish health but there is a need for establishing values in health, disease and various stress conditions before their value in diagnosis can be evaluated.

    Notes: This is a review of some of the literature on the use of chromosome techniques with fish. There is a need for developing methods which leave the animal alive. Such a test is the leukocyte culture method used in human medicine. Once a reliable test has been developed the results should show more consistent karyotypes and better morphology thus providing information of use in cytotaxonomy, fish-breeding and in studying the effects of sub-lethal levels of water pollutants on fish.

    Notes: A preliminary report is made on the distribution of acid phosphatases and acid esterases in the cells of rainbow trout. A modified technique for the cytochemical demonstration of acid esterases is given, resulting in clearer visualization. Lymphocytes undergoing transformation in the presence of phytohaemagglutinin show a parallel decrease in enzyme localization for acid esterase and acid phosphatase as transformation in culture proceeds.

    Notes: Brown trout (Satmo trutta L) lymphocytes were shown to separate into two fractions on a Percoll discontinuous gradient, with 53% of the cells in the 1.07gl-1 fraction. The cells from the two fractions showed equal enzyme activity when stained for acid esterase and acid phosphatase. About 70% of the lymphocytes gave a positive enzyme reaction, which if the reaction is comparable with mammalian lymphocyte cytochemistry would indicate they were T-lymphocytes. There appears to be increasing evidence among fish for the existence of T- and B-lymphocytes, and cytochemical staining could prove a comparatively convenient method for their demonstration.

    Notes: This investigation was carried out to determine if lymphocyte functional heterogeneity could be demonstrated in teleosts on the basis of density difference. The lymphocytes separated into two fractions on a Percoll discontinuous gradient. The less dense lymphocytes separated at 1.056gml−1 and contained both moderately villous and smooth lymphocytes, whereas the more dense lymphocytes separated at 1.07 g ml−1 and were predominantly moderately villous lymphocytes. The moderately villous lymphocytes had a higher mitochondria content than the smooth lymphocytes. Lymphocytes separating in the 1.056g ml−1 layer were more responsive to stimulation with polyclonal mitogen (PHA) than the lymphocytes separating in the 1.07 g ml−1 layer. This indicates that the lymphocytes separated according to either cell lineage or maturity.

    Notes: A comparison is made of methods currently used in separating mammalian lymphocytes from whole blood when adapted for use with fish blood. The methods give good results but do require minor modifications. The use of Ficoll-Paque, although requiring greater expertise, did result in obtaining good recovery rates of lymphocytes in a viable condition. The separation of cells without the addition of agents other than culture medium, while resulting in good yields of lymphocytes, was much more variable and did not result in the separation of such a pure cell line. The other methods compared were found to be unsuitable for fish blood.

    Notes: No observable differences were noted in electron microscopic studies between brown trout, Salmo trutta L., and carp, Cyprinus carpio L., lymphocytes and thrombocytes separated by the Ficoll-Paque technique. Transmission electron microscopy showed a similar ultra-structure of fish lymphocytes to those of other reported species. Scanning electron microscopy showed that these lymphocytes studied all had a villus structure and the thrombocytes clearly showed their spindle-shaped structure.
