    Notes: The effect of H2 translational, rotational, and vibrational excitation on the dynamics of the O(1D2)/H2 reaction are explored in a semiclassical trajectory study involving both of the energetically accessible potential energy surfaces of the system. Landau–Zener probabilities determine surface hopping. At low reagent excitation, the deep H2O potential minimum dominates the dynamics, causing the reagents to reorient towards a H–O–H (insertion) configuration and form the H2O intermediate, irrespective of the initial approach geometry. High vibrational excitation enhances the probability for transitions onto the excited state potential during the interaction. Reactions which sample the excited state potential have fundamentally different dynamics from those which remain on the lower state. For reactions involving H2(v=4), the OH product has a bimodal vibrational distribution, peaking in OH(v'=2) and OH(v'=9). The lower peak is due to reactions which access the excited state potential; the higher peak results from those which remain on the lower state for the entire interaction. High translational excitation shortens the interaction time and reduces the effect of the potential minimum to reorient the reagents. Rotational excitation also reduces the effect of the potential minimum by causing the system to rotate out of the insertion geometry before entering the potential minimum.

    Notes: We report the development of a data acquisition system which can be used with any commercial Michelson interferometer to carry out time-resolved Fourier transform spectroscopy with a time resolution of one microsecond. The sensitivity and spectral resolution of the experiment are limited only by the interferometer. The instrument occupies one slot on a conventional VME enclosure (6U height). It includes all hardware and firmware to control the experiment which generates the spectral transients and to collect the time-resolved interferograms. We are presently transferring this design to a single 6U height printed circuit board (with a mezzanine), in order to make it available as a standalone system for use with any VME platform.

    Notes: We use fast time-resolved Fourier transform spectroscopy and low pressure infrared chemiluminescence techniques to determine the product energy distribution in the title reaction. We create the reagent hydrogen atoms with 2.3 eV of translational excitation by photofragmentation of H2S at 193 nm, and observe the time evolution of the infrared chemiluminescence from the product HCl(v',J') under single gas kinetic collision conditions. The initial vibrational distribution, determined from the first observation after creation of the H atoms is P(v'=1:2:3:4:5:6:7:8)=1.0:0.84:0.74:0.59:0.34:0.24:0.17:0.13. The initial HCl rotational distribution in each vibrational level is broad, showing no identifiable maximum. The fraction of the total available energy entering HCl vibration and rotation, respectively, are 0.19 and 0.10. The time evolution of the observed vibrational and rotational distributions gives information about the changes in the reaction dynamics consequent on reduction of the reagent translational energy.

    Notes: Abstract Mistletoe preparations have been shown to express immunomodulatory properties. In order to evaluate the stimulatory potency of different mistletoe extracts, peripheral blood mononuclear cells (PBMC) from healthy and allergic/atopic individuals were exposed to aqueous or fermented extracts derived from Viscum album L. grown on apple trees (Mali-extracts) or on pines (Pini-extracts). None of them had received any mistletoe treatment. Iscador Pini was the only extract which strongly induced proliferation of PBMC in contrast to the other five preparations. On testing these extracts by Western blotting with anti-mistletoe lectin-1 (ML-1) antibody positive sera from mistletoe-treated patients, it became evident that Iscador Pini was almost devoid of lectins. The stimulatory potency of Iscador Pini for PBMC from three different groups was examined: PBMC from 35 normal controls (Group I), 23 patients with drug-induced adverse effects (Group II) and 16 individuals with allergic manifestations (Group III). Cells were exposed in 7-day cultures to the extract at concentrations between 1 and 10,000 μg/ml. PBMC from 63% of Group III individuals showed strong stimulation (SI varying from 6 to 97) in contrast to only 9% from Group I and 22% from Group II individuals. Anti-ML-1 antibodies were detected in 5% and anti-IP antibodies in 11% of subjects in the three groups. They were either of the IgA or IgM type but not of the IgG type. Our findings strongly imply that a non-lectin associated antigen from Iscador Pini is able to activate PBMC from healthy and allergic/atopic individuals, thereby demonstrating sensitization to probably highly conserved plant antigens.

    Notes: Abstract Objective: Mistletoe extracts are used in adjuvant cancer treatment, but little is known concerning their mode of action. There is, however, evidence that antigens in these extracts may stimulate cells of the immune system, thereby modifying the altered immunological reactivity in tumour patients. Methods: In order to find out whether the postulated immunomodulatory properties of mistletoe extracts are mediated by cytokines, a spectrum of different cytokines was analysed in the supernatants of peripheral blood mononuclear cells (PBMC) from healthy (n = 23) and allergic (n = 16) individuals after stimulation with the fermented mistletoe lectin-1 (ML-1) free mistletoe extract Iscador Pini (IP) in vitro, and their cytokine patterns were compared to those from tumour patients with either breast cancer (n = 20) or colorectal tumours (n = 22). Results: PBMC from healthy and allergic individuals produced high levels of TNF-α and IL-6 and to a lesser extent Th1- and Th2-related cytokines. This finding was in contrast to data obtained in tumour patients. Thus, the concentration of TNF-α was significantly lower in the cell cultures from breast cancer patients than in controls, and patients with colorectal tumours released IFN-γ/IL-2 (5%) in the supernatants significantly less frequently than PBMC from healthy controls (26%). Similar results were obtained when the Th1- and monocyte/macrophage-related cytokines were analysed in the unstimulated cell cultures. Conclusions: These in vitro studies provide evidence that there is a reduced immunological reactivity to the fermented ML-1 free mistletoe extract in tumour patients and may give some clues as to how mistletoe-derived antigens could act on immune cells involved in the tumour defence.

    Notes: Summary To evaluate the specificity of the lymphocyte transformation test (LTT) in the diagnosis of drug allergy we studied over 71 days an atopic woman with a past history of frequent adverse reactions to pyrazolone drugs. Rechallenge with the incriminated substances aminophenazone (aminopyrine) and propyphenazone was carried out on Days 11 and 31 respectively. An immediate type of hypersensitivity reaction was seen after 100 mg aminophenazone, while 100 mg of propyphanozone led to a serum sickness-like syndrome. We found two specifically sensitized lymphocyte populations using either the pure substance or sera containing metabolite in cell cultures. Stimulatory responses with indices ranging between 3 and 6 were seen 3–4 days after exposure, and the tests remained positive in both instances for 3–4 weeks. Specific sensitization was proven by positive skin tests and by a small but distinct lymphocyte proliferative response before challenge. Several lymphocyte function tests were performed over a period of 53 days and revealed a large fall in pokeweed mitogen-induced immunoglobulin synthesis and an increase in suppressor cell activity after rechallenge with aminophenazone. We conclude that the proliferative response observed in the presence of the offending drug is due to the activation of T memory cells and therefore highly suggestive of a true allergic reaction.

    Notes: Summary 3,4-dihydro-6-[4-(3,4-dimethoxybenzoyl)-1-piperazinyl]-2(1H)-quinolinone (OPC-8212) is a quinolinone derivative with positive inotropic properties. In order to elucidate the effect of OPC-8212 on the haemopoietic system we studied its in vitro effect on bone-marrow progenitor cells (granulocyte/monocyte colonyforming units [CFU-GM] and erythroid burst-forming units [BFU-E]), on the proliferation and secretion of granulocyte/monocyte colony-stimulating factor (GM-CSF) and interferon-γ (IFN-γ) by peripheral lymphocytes, and on GM-CSF secretion by fibroblasts from healthy individuals. The dose-effect relations of OPC-8212 on CFU-GM proliferation and on lymphocytic GM-CSF secretion showed no effect at very low drug concentrations, with a threshold at the lower end of the therapeutic range and highly significant dose-dependent inhibition at concentrations above that threshold. BFU-E, peripheral lymphocyte proliferation and lymphocytic IFN-γ secretion were depressed, although to a lesser extent, in a linear dose-dependent fashion. OPC-8212 did not affect GM-CSF secretion by one strain of fibroblasts but reduced it at higher concentrations in assays with another strain of cells. We conclude that direct toxic effects on bone-marrow progenitor cells, in combination with the inhibition of cytokines involved in the regulation of haemopoiesis in certain susceptible individuals, may be responsible for idiosyncratic reactions to OPC-8212.

    Notes: Summary Recently, there has been a variety of reports of adverse drug reactions during therapy with the flavonoid Cianidanol (Ci), a cytoprotective radical scavenger, especially involving haemolytic anaemia and drug fever. To elucidate whether the fever was due to a direct, antigen-independent interaction of Ci with immune competent cells, its effect on macrophage (MΦ) function and early biochemical events during lymphocyte activation has been examined. A direct interaction of Ci with MΦ was demonstrated, resulting in increased secretion of interleukin-1 (IL-1). The influence of Ci on lymphocyte activation was assessed by measuring levels of cyclic AMP and GMP. At high concentrations of Ci, cAMP levels were increased, and at low Ci concentrations cGMP levels were elevated. Both findings are correlated with lymphocyte proliferation and function, which is increased at low and decreased at high concentrations of Ci. The synthesis of prostaglandin E2 by MΦ, an important factor in MΦ-mediated suppression, was reduced by increasing doses of Ci, which inhibited MΦ-cyclooxygenase. Ci did not affect phospholipase A2 activity. These findings indicate that flavonoid-induced fever may be due to allergic as well as pseudo-allergic mechanisms, the latter probably caused by increased antigen-independent release of IL-1, the endogeneous mediator of fever.
