    Notes: Summary The question of cell death is accessible to study by histochemists and many questions remain to be resolved. From a physiological point of view, the most important are the causal relationships. (1) At what phase in cell death is the synthesis of RNA disrupted and at what phase is the rate of degradation of RNA increased? (2) Does the disruption of synthesis result from a direct genetic command, or does it result indirectly from gradual deterioration of energy resources or optimal ionic conditions? (3) What properties, presumably of the substrate organelles, marks them for specific absorption into autophagic vacuoles? (4) What proteases and other hydrolases operate currently undetected in the cytoplasm? How are they controlled and regulated? (5) Why does the physiologically dying cell shrink and appear more dense? To what extent is a cell in this state able to regulate any metabolic parameter? The advent of newer, more sensitive and quantitative techniques, and greater attention to the controls and causes as opposed to the phenomena, should help to resolve these questions.

    Notes: Summary Calcium stores were cytochemically demonstrated using a combined oxalate—pyroantimonate method in the neuromuscular junctions of the degenerating intersegmental muscles in the giant silkmothAntheraea polyphemus. The elemental composition of punctate precipitates of the reaction product was determined by electron probe X-ray microanalysis of unstained thin sections by energy-dispersive spectrometry and wavelength-dispersive spectrometry. The wavelength-dispersive spectra collected over terminal axons demonstrate a significant calcium signal and a trace of antimony. During the rapid lytic phase of spontaneous muscle degeneration, the calcium punctate deposits were detected in presynaptic terminals in the following sites: the synaptic vesicles and the mitochondria. Calcium precipitates were also found in the dense bodies and the mitochondria encountered in the glial convolutions. No calcium deposit was seen in the synaptic clefts and intercellular spaces of the subsynaptic reticulum of type I and type II. A comparison of calcium to antimony ratios between the terminal axons and the sarcoplasmic lysosomes revealed highly significant differences (P〈0.001). Such a variability of the calcium to antimony ratio may be related to different conditions of precipitation or antimony diffusion in the different cell compartments. It was concluded that such synaptic terminals do not appear damaged in spite of the muscle degeneration and presumably continue to perform vital functions while the muscles are no longer contractile 20 h after adult ecdysis.

    Notes: Summary The intersegmental muscles in the metamorphosing silkmothAntheraea polyphemus were examined by two electron cytochemical procedures for demonstration of calcium compartmentation during the two-day period of degeneration after emergence. Muscle fibres were treated with either oxalate—pyroantimonate, or phosphate—pyroantimonate procedures. The elemental composition of the reaction product arising from the oxalate procedure was determined with electron probe X-ray microanalysis of unstained thin sections by energy dispersive spectrometry and wavelength dispersive spectrometry. The wavelength dispersive data revealed high peaks of calcium and antimony in the electron-dense precipitates. No reaction was obtained in muscles after treatment with the phosphate—pyroantimonate method. Shortly after the emergence of the moth, very few calcium deposits were found in the mitochondria, which also contained amorphous matrix densities. During the rapid lytic phase (17 and 30 h after ecdysis), the mitochondria, autophagic vacuoles sequestering mitochondria, and lysosomal dense bodies issuing from the latter were highly reactive in each muscle fibre. These results demonstrate that the collapse of tracheae (hypoxic conditions) is correlated with the calcium overload of mitochondria when the cell calcium homeostasis is apparently lost. Such calcium overload of the mitochondria appears to cause irreversible damage to these organelles which are then sequestered in autophagic vacuoles. This mitochondrial autophagic process leads to calcium translocation into a lysosomal compartment. We suggest that the calcium lysosomal stores may have a transient function of cell detoxification and stimulation of calcium-dependent degradative processes prior to the final muscle collapse.

    Notes: In studying ultrastructural changes in metamorphosis-related degeneration of intersegmental muscles in Antheraea polyphemus, particular attention was directed to the mechanisms and timing of degradation of organelles and myofilaments. At emergence, the muscles are typical slowly contracting insect muscles, with a few dense body lysosomes and occasional autophagic vacuoles containing mitochondria. During the early phases of degradation the number of autophagic vacuoles, dense bodies, and lamellar bodies increases rapidly, along with an expansion of the Golgi system and the T system. Free glycogen particles and glycogenosomes are demonstrated by the PATAg test.Between 7 and 20 hours after ecdysis the T system continues to expand, the fibers subdivide, and the contractile system is degraded. Myofibrils fragment; myofilaments are not enclosed in isolating membranes at the time of their dissolution. The destruction of individual filaments occurs rapidly, with few intermediate stages being seen, while thick filaments tend to disappear earlier than thin filaments and Z-line material. The process is generalized and not confined to specific regions of the fiber. Autophagy destroys cell organelles in apparent synchrony with the first signs of nuclear pycnosis.By 20 to 30 hours after emergence, the fibers are reduced to lamellae of polynucleate sarcoplasm containing no organized contractile material. The sarcoplasm is filled with autophagic vacuoles containing mitochondria, dense lamellar or residual bodies, and ribosome-rich sarcoplasm. The number of mitochondria is drastically reduced at this time.In the final phases of involution (40-49 hours after emergence) shedding of the residual sarcoplasm precedes the expulsion of the pycnotic nuclei into the hemocoele.These results indicate that autophagy is responsible for the selective destruction of mitochondria, glycogen particles, ribosomes, and other organized sarcoplasmic structures. The one exception is the dissolution of the myofilaments, a process which remains undefined but which appears to be independent of lysosomal activity.

    Description / Table of Contents: Résumé Des expériences d'extraction enzymatique sur coupes ultrafines de glandes prothoraciques incluses en Epon, pratiquées selon la méthode de Monneron (1966a, b), ont montré que certaines structures cellulaires peuvent être attaquées par la pepsine après fixation glutaraldéhydique-osmique. Ce sont les mitochondries typiques dont la matrice est extraite par l'action modérée de la pepsine à 40° C, et les pores nucléaires avec leurs anneaux denses qui sont également digérés par cette protéase. La protéolyse complète de toutes les structures associées à ces pores confirme leur nature protéique suggérée par les expériences de digestion trypsique effectuées par Merriam (1961). Ces résultats sont discutés en relation avec nos connaissances sur la composition chimique de ces structures.
    Notes: Summary Enzymatic extraction, according to Monneron's method (1966a, b), in ultrathin sections of prothoracic glands embedded in Epon has shown that some cellular structures are rapidly attacked by pepsin after glutaraldehydic-osmium tetroxide fixation. These are the typical mitochondria whose matrix is extracted by the moderate action of pepsin at 40° C and the nuclear pores with dense annuli which are digested by this protease. The complete lysis of all the structures associated with these pores confirms their protein nature suggested by the experiments of Merriam (1961) with trypsin. These results are discussed in the light of our present knowledge about the chemical composition of these structures.
