    Notes: Melanophore constellation patterns on the heads of 0+ year Atlantic salmon Salmo salar allowed robust identification of individuals, and were probably unique. Reliable identification was possible from shortly after emergence until at least 16 months later. The method for recording the patterns was inexpensive, required no special equipment and had no additional physical impact on the fish.

    Notes: In neutron-diffraction measurements of the orientation of apatite crystals, for studying the stress distribution in bone, it is necessary to remove substantially the collagen in order to reduce the incoherent scattering. Data from X-ray and neutron diffraction indicate the effect of chemical treatment with hydrazine and ethylenediamine and of heat treatment up to 650°C. Crystal growth commences above 500°C, with a slight sharpening of the curve of preferred orientation but without change of the preferred direction. Typically the half-width at half-intensity is reduced from 28° to 24° at 650 °C. Fortuitously, because of the differences of scattering amplitudes for the constituent atoms in apatite, the neutron pattern is much more informative concerning crystallinity than is the X-ray pattern.

    Notes: High-intensity neutron beams provide a means of measuring the integrated orientation of apatite crystals in bulk samples of bone. With a resolution of 2 mm2 in compact bone and 12 mm2 in trabeculae this promises a tool for the study of the load-bearing function of bone in a variety of anatomical sites.

    Notes: Abstract Temperatures of more than 25° C adversely affect the activity of soluble starch synthase (SSS), an amyloplastic enzyme, in endosperm of wheat (Triticum aestivum L. cv. Mardler). Enzyme rate was found to have a temperature optimum between 20 and 25°C. This effect was apparently reversible after a short period of exposure to elevated temperature. We also found that with a prolonged period of exposure to elevated temperature there was another temperature-related phenomenon which caused a loss of enzyme activity that appeared to be much slower to reverse. We have termed this effect of temperature on SSS activity “knockdown”. The knockdown in SSS activity also occurred in-vivo. However, elevated temperature did not affect the activities of several other enzymes in the pathway of starch synthesis (ADP-glucose pyrophosphorylase, UDP-glucose pyrophosphorylase, sucrose synthase, phosphoglucomutase, phosphoglucose isomerase, bound starch synthase or hexokinase). Because the knockdown effect appeared to be specific to the enzyme SSS, we quantified the effect of knockdown on flux of carbon into starch and used these data to calculate the flux-control coefficient for SSS. Using data at 10–20°C the flux-control coefficient was CStarch10–20C = 0.50, whereas at 20–30° C the flux-control coefficient was CStarch20–30C = 1.38, and between 30–40°C the flux-control coefficient was CStarch30–40C = 0.69. Using data at 10–30°C the flux-control coefficient was CStarch10–30C = 1.15, and at 10–40°C the flux-control coefficient was CStarch10–40C = 0.82. In conclusion, we suggest that SSS is a major site of regulation of starch synthesis in developing wheat grain. During periods of high temperature the control point in the pathway of starch synthesis is apparently not associated exclusively with ADP-glucose pyrophosphorylase. In field conditions, in which temperatures are fluctuating, there will likely be periods of control of starch synthesis being exerted predominantly by SSS. During periods at lower temperature, control of flux may be exerted by SSS, perhaps in combination with other flux-controlling enzymes in the pathway. Our data point-out a crucial new aspect of quantifying control strengths of enzymes in plants: the determination of enzyme control strengths should be done in carefully regulated temperature conditions. Thus, since temperature is a major determinant of real flux through a pathway and the individual enzymes can respond differently to changing temperature conditions, the control strengths of individual steps in a pathway may vary with changing environmental conditions. This is particularly pronounced in starch deposition, because of the temperature instability of SSS.

    Description: Tracer investigations were combined with a geographical information system (GIS) analysis of the 31 km2 Girnock catchment (Cairngorm Mountains, Scotland) in order to understand hydrological functioning by identifying dominant runoff sources and estimating mean residence times. The catchment has a complex geology, soil cover and topography. Gran alkalinity was used to demonstrate that catchment geology has a dominant influence on baseflow chemistry, but flow paths originating in acidic horizons in the upper soil profiles controlled stormflow alkalinity. Chemically based hydrograph separations at the catchment scale indicated that ∼30% of annual runoff was derived from groundwater sources. Similar contributions (23-36%) were estimated for virtually all major sub-basins. δ18O of precipitation (mean: -9.4‰; range: -16.1 to -5.0‰) and stream waters (mean: -9.1‰; range: -11.6 to -7.4‰) were used to assess mean catchment and sub-basin residence times, which were in the order ∼4-6 months. GIS analysis showed that these tracer-based diagnostic features of catchment functioning were consistent with the landscape organization of the catchment. Soil and HOST (Hydrology of Soil Type) maps indicated that the catchment and individual sub-basins were dominated by hydrologically responsive soils, such as peats (Histosol), peaty gleys (Histic Gleysols) and tankers (Umbric Leptosols and Histosols). Soil cover (in combination with a topographic index) predicted extensive areas of saturation that probably expand during hydrological events, thus providing a high degree of hydrological connectivity between catchment hillslopes and stream channel network. This was validated by aerial photographic interpretation and groundtruthing. These characteristics of hydrological functioning (i.e. dominance of responsive hydrological pathways and short residence times) dictate that the catchment is sensitive to land use change impacts on the quality and quantity of streamflows. It is suggested that such conceptualization of hydrological functioning using tracer-validated GIS analysis can play an important role in the sustainable management of river basins.
