    Notes: Basal leaf sections of Triticum aestivum L., cv. ‘Chinese Spring’, and XTriticosecale Wittmack cv.‘Welsh’were cultured on Kao's medium with 2 mg/l of 2,4-D. Callus was induced on the leaf sections at frequencies of 64 and 63 %, respectively, and plant-lets regenerated at 18 and 31 %, respectively. Compared to the controls, the regenerated plants showed higher frequencies of univalent chromosomes at meiosis and low frequencies of trivalents, and quadrivalents; indicating that the callus phase had induced low frequencies of chromosome irregularity.

    Notes: Common wheat × hexaploid triticale hybrids were produced and evaluated for tolerance to barley yellow dwarf virus disease (BYD). The BYD tolerance expression varied with wheat × triticale combination. The selection for BYD tolerance increased the recovery of tolerant genotypes in the next generations. Homozygous tolerant and susceptible lines were obtained in advanced generations. The rye chromosomes 1R, 2R, and 4R with 7R were transmitted as disomic or monosomic, disomic, and double disomic substitution to the late generations of ‘Musala’ (common wheat) בMuskox 658’ (triticale), ‘Encruzilhada’ (common wheat) בNord Kivu’ (triticale) and ‘Encruzilhada’× 12th. International Triticale Screening Nursery 267 (12ITSN267) (triticale), respectively. A clear association was established between the 1R chromosome of the ‘Muskox 658’ triticale line and the tolerance to BYDV. Results suggest that the 2R chromosome may be involved in BYD tolerance of ‘Nord Kivu’ triticale line.

    Notes: Summary The genus Avena consists of at least 23 species composed of three ploidy levels. Cytogenetic analysis has characterised four distinct karyotypes. These are the A, B, C and D genomes. We have isolated a repeated sequence clone that can be used for the detection of the C genome in Avena by filter hybridization techniques. This clone, termed RS-1, is a genomic DNA clone containing at least one highly repeated sequence that is abundant in Avena species containing the C genome. This sequence or a related sequence is also present, but at much reduced levels, in species that do not contain the C genome. Because of its abundance and the characteristic Southern blot pattern, we have termed this clone a C genome specific clone. We have also done similar analysis of the Avena genus using a rDNA clone from wheat. The results of these experiments demonstrate that clearly definable C genome-specific markers can be identified with both probes. These molecular probes can be useful in studying the genomic relationships of Avena and can provide some clues as to the origin of the cultivated Avena species. These results can, therefore, provide breeders with directions for the efficient transfer of desirable traits of wild Avena species into commencal varieties.

    Notes: Summary Hybrids between B. inermis Leyss (2n=8x=56) and B. riparius Rehm. (2n=10x=70) were easily made. The F1 hybrids had a fertility of 20%–50% under open pollination and backcrossing to B. inermis. Chromosome pairing in B. riparius was predominantly as bivalents (29.04–33.85 per cell for plant means). Bivalents also predominated in the F1 hybrid (2n=9x=63) and there was a high level of pairing with no reduction in chiasma frequency. It was impossible to estimate the frequency of auto-versus allosyndetic pairing. Chromosome pairing in a hybrid between B. arvensis (2n=2x=14) and B. riparius confirmed that the B. riparius complement is capable of complete autosyndetic pairing. Chromosome numbers in the F2 progeny ranged from 2n=56 to 72 but they were skewed towards 2n=63 to 70. Backcrosses ranged from 2n=56 to 63, as expected, with the distribution skewed towards 2n=56. Selection towards the 2n=56 level would be difficult in the F2. Empirical observation suggested that cytoplasm had a major influence on morphology in the backcrosses. Additional studies are required to determine the best breeding scheme to introgress germ plasm between B. inermis and B. riparius.

    Notes: Abstract  Broadening of the genetic base and systematic exploitation of heterosis in cultivated lentils requires reliable information on genetic diversity in the germ-plasm. The ability of random amplified polymorphic DNA (RAPD) to distinguish among different taxa of Lens was evaluated for several geographically dispersed accessions/cultivars of four diploid Lens species. This study was carried out to assess whether RAPD data can provide additional evidence about the origin of the cultivated lentil and to measure genetic variability in lentil germplasm. Three cultivars of Lens culinaris ssp. culinaris, including one microsperma, and two macrosperma types, and four wild species (L. culinaris ssp. orientalis, L. odemensis and L. nigricans) were evaluated for genetic variability using a set of 1 11-mer and 14 random 10-mer primers. One hundred and fifty-eight reproducible and scorable DNA bands were observed from these primers. Genetic distances between each of the accessions were calculated from simple matching coefficients. Split decomposition analysis of the RAPD data allowed construction of an unrooted tree. This study revealed that (1) the level of intraspecific genetic variation in cultivated lentils is narrower than that in some wild species. (2) L. culinaris ssp. orientalis is the most likely candidate as a progenitor of the cultivated species, (3) L. nigricans accession W6 3222 (unknown) and L. c. ssp. orientalis W6 3244 (Turkey) can be reclassified as species of L. odemensis and (4) transmission of genetic material in Lens interspecific hybrids is genotypically specific, as identified by the RAPD markers in our study.

    Notes: Summary Chromosome pairing was studied in hybrids of (Hordeum vulgare ×Triticum aestivum) ×Secale cereale. Chiasma frequency per cell varied from 1.94 to 3.16 between the different hybrids. This variation was attributed to genetic variability in rye parents which affected homoeologous pairing. The pairing of rye chromosomes as revealed by Giemsa C-banding was a combination of nonhomologous association between rye chromosomes and associations with chromosomes of wheat and barley.
