    Description: The European CORDEX (EURO-CORDEX) initiative is a large voluntary effort that seeks to advance regional climate and Earth system science in Europe. As part of the World Climate Research Programme (WCRP) - Coordinated Regional Downscaling Experiment (CORDEX), it shares the broader goals of providing a model evaluation and climate projection framework and improving communication with both the General Circulation Model (GCM) and climate data user communities. EURO-CORDEX oversees the design and coordination of ongoing ensembles of regional climate projections of unprecedented size and resolution (0.11° EUR-11 and 0.44° EUR-44 domains). Additionally, the inclusion of empirical-statistical downscaling allows investigation of much larger multi-model ensembles. These complementary approaches provide a foundation for scientific studies within the climate research community and others. The value of the EURO-CORDEX ensemble is shown via numerous peer-reviewed studies and its use in the development of climate services. Evaluations of the EUR-44 and EUR-11 ensembles also show the benefits of higher resolution. However, significant challenges remain. To further advance scientific understanding, two flagship pilot studies (FPS) were initiated. The first investigates local-regional phenomena at convection-permitting scales over central Europe and the Mediterranean in collaboration with the Med-CORDEX community. The second investigates the impacts of land cover changes on European climate across spatial and temporal scales. Over the coming years, the EURO-CORDEX community looks forward to closer collaboration with other communities, new advances, supporting international initiatives such as the IPCC reports, and continuing to provide the basis for research on regional climate impacts and adaptation in Europe.

    Notes: We compute the spontaneous polarization, P, exhibited by a class of ferroelectric liquid crystals (FLCs) with systematically varying structures in an effort to critically test a recently proposed model [D. J. Photinos and E. T. Samulski, Science 270, 783 (1995)] which implies that polarity is indigenous to tilted smectic phases. We evaluate the first and second rank segmental order parameters of the FLC compounds by taking into account the essential features of molec-ular structure and conformation subjected to the general stratification constraints in the model smectic FLC. The results compare favorably with available experimental values of P and bond order parameters obtained from NMR measurements. © 1997 American Institute of Physics.

    Notes: A new method for calculating the ground-state tunneling splitting is presented. It is based on the semiclassical theory including recently derived corrections and it is the first method, which explicitly takes into account the whole conformational space between the minima and the transition state. The density-functional theory is used to determine the qualitative shape of the potential energy surface (PES) and high level ab initio calculations provide information about the stationary points. With a dual level scheme, the low-level energy surface is mapped onto the high-level points to get a good quantitative description of the high-level PES. Therefore, the new method requires no adjustment of additional parameters like scaling of the energy barrier as is necessary in other methods. Once the high-level PES is calculated, the most probable tunneling paths are determined with a global optimization procedure. Along this representative tunneling path, the tunneling splitting is calculated with additional consideration of zero-point vibrational effects. The method is applied to three molecular systems, namely hydrofluoric acid dimer, malonaldehyde, and tropolone. These systems were chosen because their energy barriers differ strongly (1 kcal/mol–7 kcal/mol). The predicted tunneling splittings agree very well with the experimental ones, therefore, we expect our method to be generally applicable, independent of the magnitude of the energy barrier. © 2002 American Institute of Physics.

    Notes: The proton tunneling reaction in malonaldehyde at low temperatures is investigated. The principal aim of this study is to find the optimal tunneling path at 0 K in the framework of the semiclassical theory with a global optimization method. An amount of 11366 ab inito points was determined in the reaction swath (i.e., the conformational space enclosed by the minima and the transition state) of malonaldehyde. With a simulated annealing approach, the path with the smallest integral of the imaginary action through the swath from minimum to minimum was determined. Surprisingly the optimal tunneling path was found to be quite far off the large curvature tunneling path [i.e., the straight connection of the two minima large-current tunneling (LCT path)]. At the beginning, it is following the minimum energy path (MEP) (i.e. the path with the lowest energy connecting the two minima and passing through the transition state), and then it is describing a curved path through the reaction swath. This curve was determined several times with different annealing schemes, which ended up with the same result—the tunneling path is proceeding close to the MEP rather than to the LCT path. Along the optimal tunneling path, the ground-state tunneling splitting was calculated with a new semiclassical method introduced in an accompanying study [C. S. Tautermann, A. F. Voegele, T. Loerting, and K. R. Liedl, J. Chem. Phys. 117, 1967 (2002), following paper]. Another focus of investigation was the influence of deformation of the tunneling paths and a general scheme of determining an approximated optimal tunneling path at 0 K is introduced. © 2002 American Institute of Physics.

    Notes: One requirement for the invasion of, and tight adherence to, human epitheiial cells by Neisseria gonorrhoeae is the synthesis of distinct opacity (Opa) outer membrane proteins, encoded by a family of phase-variable chromosomal genes. However, cloning and surface expression of invasion-promoting Opas in Escherichia coli is not sufficient for the efficient invasion of epithelial cells: additional factors besides Opa may be involved in this process. Using the phoA mini-transposon TnMax4, a library of gonococcal mutants affected in the expression of genes encoding exported proteins was generated through shuttle mutagenesis. Of a total of 608 PhoA+ plasmid clones identified in E. coli E145 approximately 40% were used successfully in transforming N. gonorrhoeae and in activating the corresponding chromosomal genes. Gonococci producing the invasion-promoting Opa50 served as the genetic background to identify 51 mutants unable to enter Chang human epithelial cells. We expect some of these mutations affect the interaction of N. gonorrhoeae with epithelial cells directly, while other mutants may carry defects in general house-keeping, secretory and/or regulatory determinants. In some mutants the loss of invasiveness appears to be due to a negative dominant effect of the PhoA+ fusions produced in these mutants. Some of the identified genes display a phase-variation phenomenon in E. coli and several genes are found in multiple copies in N. gonorrhoeae and/or present only in pathogenic Neisseria species.

    Notes: Abstract An SHV type β-lactamase frequently found in enterobacteria isolated in Greek hospitals was analyzed. The enzyme (SHV-5a) conferred resistance to ceftazidime and aztreonam. The DNA sequence of the structural gene was determined. The deduced amino acid sequence showed that positions 70–73 were occupied by the active site tetrad Ser-Thr-Phe-Lys. As in SHV-5, Ser-238 and Lys-240 were present. However, one deletion (Gly-54) and three substitutions (Arg-140 for Ala, Asn-192 for Lys and Val-193 for Leu) differentiate SHV-5a β-lactamase from SHV-5. Asn-192 and Val-193 have been reported to date only in the R974 plasmid-mediated SHV-1 β-lactamase. Hydrolysis studies with SHV-5a and SHV-5 showed that the enzymes behaved similarly. Additional evidence that they were functionally indistinguishable was provided by the similar MICs of β-lactams when the enzymes were expressed under isogenic conditions. The sequence differences, however, indicate that they are derived from different ancestors.

    Notes: We characterized a novel mutant phenotype (tetrapac, tpc) of Neisseria gonorrhoeae (Ngo) associated with a distinctive rough-colony morphology and bacterial growth in clusters of four. This phenotype, suggesting a defect in cell division, was isolated from a mutant library of Ngo MS11 generated with the phoA minitransposon TnMax4. The tpc mutant shows a 30% reduction in the overall murein hydrolase activity using Escherichia coli murein as substrate. Tetrapacs can be resolved by co-cultivation with wild-type Ngo, indicating that Tpc is a diffusible protein. Interestingly, Tpc is absolutely required for the natural transformation competence of piliated Ngo. Mutants in tpc grow normally, but show a ∼ 10-fold reduction in their ability to invade human epithelial cells. The tpc sequence reveals an open reading frame of ∼1 kb encoding a protein (Tpc) of 37kDa. The primary gene product exhibits an N-terminal leader sequence typical of lipoproteins, but palmitoylation of Tpc could not be demonstrated. The ribosomal binding site of tpc is immediately downstream of the translational stop codon of the folC gene coding for an enzyme involved in folic acid biosynthesis and one-carbon metabolism. The tpc gene is probably co-transcribed from the folC promoter and a promoter located within the folC gene. The latter promoter sequence shares significant homology with E. coli gearbox consensus promoters. All three mutant phenotypes, i.e. the cell separation defect, the transformation deficiency and the defect in cell invasion can be restored by complementation of the mutant with an intact tpc gene. To some extent the tcp phenotype is reminiscent of iap in Listeria, lytA in Streptococcus pneumoniae and lyt in Bacillus subtilis, all of which are considered to represent murein hydrolase defects.
