    Notes: SUMMARY. Ochromonas malhamensis (Pringsheim strain) can be grown above 35.5°C.; below 35°, the previous chemically defined medium supports dense growth. The B12 and thiamine requirements rise steeply with temperature, and growth promotion by folic acid emerges; folic acid spares the enhanced B12 requirement. B12 is spared also, perhaps wholly bypassed, by purines + pyrimidines + amino acids (below 35°, exogenous purines, pyrimidines, and folic acid have little effect). Requirements also emerge for glycine (spared by serine), valine and isoleucine (their ratio is critical; leucine and threonine assist in maintaining a good balance), and, at very slightly higher temperatures, phenylalanine, tryptophan, cystine, and lysine. Requirements for Mg, Fe, Zn, and Mn appear to rise steeply with temperature; metal toxicities have to be circumvented carefully. The proportion of histidine + arginine to carbohydrate has to be increased, and a Krebs-cycle component such as succinic acid becomes stimulatory. At 36.3–36.7°, a further supplement of crude natural materials such as an autoclaved suspension of Ochromonas cells is needed. Relevance of these findings to fever stress in vertebrates, general mitochondrial function, and repair of radiation damage, is discussed.

    Notes: SYNOPSIS. The heterotrophic, phototrophic, and phagotrophic nutrition of the chrysomonad flagellates Ochromonas malhamensis and O. danica were compared. Unlike O. malhamensis, O. danica, which is much richer in chloroplast pigments, grew readily photoautotrophically in a substrate-free medium in 5% CO2. Utilization of bound biotin in bacterial (Thiobacillus) bodies served to demonstrate phagotrophy in both flagellates. This nutritional versatility suggests that this group of flagellates will be exceptionally valuable for studying the evolutionary steps connecting photoautotrophy, heterotrophy, and phagotrophy.

    Notes: Total lipids of several protozoa were determined and found higher than in most other microorganisms. The lipid content in a phytoflagellate, Ochromonas malhamensis, could be increased by excess nicotinic acid or deficiency in nitrogen supply. Cell multiplication and lipid synthesis were unaffected by inhibitors of vitamin synthesis such as penicillamine, pan-toyltaurine, or pyridine-3-sulfonic acid, up to 100 mg%. The phytoflagellates, O. danica, O. malhamensis, Euglena gracilis, and the photosynthetic bacterium Chlorobium limkola contained large quantities of non-saponifiable lipids: ergesterol was identified as a major component of this lipid fraction. Sterols were not found in the bacteria, Streptococcus faecalis, Rhodopseudomonas palustris; the algae. Cyanidium calidarnm. Anacystis nidulans, and Porphyridium cruentum; and the protozoan Tetrahymena pyriformis.

    Notes: 3-Amino-l,2,4-triazole (AT) stops multiplication of the photosynthetic protists, Ochromonas danica, O. mal-hamensis, Euglena gracilis var. bacillaris. and Rhodopseudo-monas palustris; it also stops growth of the multicellular plant, Spirodela oligorhiza. O. danica and S. oligorhiza are the most sensitive. AT is about equally inhibitory to algal flagellates grown in the light or dark. Porphyrin biosynthesis is suggested as the probable site of inhibition by AT. Both AT and streptomycin (SM) bleach all the above organisms. Bleaching is permanent only in SM-bleached E. gracilis; all the other organisms become fully pigmented when placed in inhibitor-free media. The uniqueness of the SM-bkaching of E. gracilis is discussed.

    Notes: SYNOPSIS. Stationary cultures of Ochromonas danica accumulated lipids as they aged. The bulk of the increase in fatty acids was in the unsaturates, particularly the polyunsaturates. The quantity of lipid peroxides (thiobarbituric acid-positive material) also increased with age. Aging was also associated with increase in sensitivity to inhibitory compounds. The implications of lipid accumulation for cell sensitivity are discussed.

    Notes: SYNOPSIS. Changes accompanying aging of stationary cultures of Ochromonas danica were examined with the electron microscope. The cultures included light- and dark-grown populations ranging in age from 3 days to 5 weeks. Cells from the youngest cultures contained minimal amounts of lipid and a distinct leucosin vacuole. After 1 week, the number of lipid globules in the cytoplasm increased. The amount of lipid increased progressively in cells from older cultures until the leucosin vacuole was obliterated by the coalescing spheres. Cells from cultures older than 3 weeks showed a general breakdown of cytoplasmic integrity. An area of pinocytotic activity was also present; a relationship between this anterior region and blebs arising from the cell membrane is suggested.

    Notes: SYNOPSIS. Euglena gracilis strain Z rendered permanently apochlorotic by several chemical and physical agents contains bodies similar in size to normal plastids. Some of these bodies contain lamella-like structures; others contain crista-like structures. The lamella- and crista-like structures are made up of 2 electron-dense layers separated by an electron-lucent layer.These structures are compared to normal lamellae and it is suggested that apochlorotic cells contain lamellae that lack the thicker, inner electron-dense layer, or more likely, that the apochlorotic condition prevents two discs from fusing to produce the thicker inner electron-dense layer which contains the photosynthetic pigments.
