    Beschreibung: On 4 August 2014, a catastrophic breach of the Mount Polley mine tailings impoundment released ~25 M m 3 of tailings and water and scoured an unknown quantity of overburden into the West Basin of Quesnel Lake. We document Quesnel Lake and Quesnel River observations for 2 months postspill. Breach inflows raised Quesnel Lake by 7.7 cm, equivalent to ~21 M m 3 . The West Basin hypolimnion was modified immediately, exhibiting increased temperature (~5°C to 6–7.5°C), conductivity (110 to 160 μS/cm), and turbidity (〈1 to 200–1000 nephelometric turbidity units (NTU)). Cooscillating seiches moved West Basin hypolimnetic water both westward and eastward contaminating the Main Basin. Postspill, high-turbidity water propagated eastward (~1 cm/s), introducing a persistent ~20 m thick layer below the thermocline and an ~30 m thick layer at the bottom. The contaminant introduction, mobilization, and bioaccumulation may pose risks to resident and anadromous fish stocks, which support recreational, commercial, and First Nations fisheries.

Goff Most cancers arise from oncogenic changes in the genomes of somatic cells, and while the cells may migrate by metastasis, they remain within that single individual. Natural transmission of cancer cells from one individual to another has been observed in two distinct cases in mammals (Tasmanian devils and dogs), but these are generally considered to be rare exceptions in nature. The discovery of transmissible cancer in soft-shell clams (Mya arenaria) suggested that this phenomenon might be more widespread. Here we analyse disseminated neoplasia in mussels (Mytilus trossulus), cockles (Cerastoderma edule), and golden carpet shell clams (Polititapes aureus) and find that neoplasias in all three species are attributable to independent transmissible cancer lineages. In mussels and cockles, the cancer lineages are derived from their respective host species; however, unexpectedly, cancer cells in P. aureus are all derived from Venerupis corrugata, a different species living in the same geographical area. No cases of disseminated neoplasia have thus far been found in V. corrugata from the same region. These findings show that transmission of cancer cells in the marine environment is common in multiple species, that it has originated many times, and that while most transmissible cancers are found spreading within the species of origin, cross-species transmission of cancer cells can occur.

    Beschreibung: Phase equilibria modelling, laser-ablation split-stream (LASS)-ICP-MS petrochronology, and garnet trace element-geochemistry are integrated to constrain the P–T–t history of the footwall of the Priest River metamorphic core complex, northern Idaho. Metapelitic, migmatitic gneisses of the Hauser Lake Gneiss contain the peak assemblage garnet + sillimanite + biotite ± muscovite + plagioclase + K-feldspar ± rutile ± ilmenite + quartz. Interpreted P–T paths predict maximum pressures and peak metamorphic temperatures of ~9.6–10.3 kbar and ~785–790 °C. Monazite and xenotime 208 Pb/ 232 Th dates from porphyroblast inclusions indicate metamorphism occurred at c . 74–54 Ma. Dates from HREE-depleted monazite formed during prograde growth constrain peak metamorphism at c . 64 Ma near the centre of the complex, while dates from HREE-enriched monazite constrain the timing of garnet breakdown during near-isothermal decompression at c . 60–57 Ma. Near-isothermal decompression to ~5.0–4.4 kbar was followed by cooling and further decompression. The youngest, HREE-enriched monazite records leucosome crystallization at mid-crustal levels c . 54–44 Ma. The northernmost sample records regional metamorphism during the emplacement of the Selkirk igneous complex ( c . 94–81 Ma), Cretaceous-Tertiary metamorphism, and limited Eocene exhumation. Similarities between the Priest River complex and other complexes of the northern North American Cordillera suggest shared regional metamorphic and exhumation histories; however, in contrast to complexes to the north, the Priest River contains less partial melt and no evidence for diapiric exhumation. Improved constraints on metamorphism, deformation, anatexis, and exhumation provide greater insight into the initiation and evolution of metamorphic core complexes in the northern Cordillera, and in similar tectonic settings elsewhere.     Beschreibung: Breast cancer is the most commonly diagnosed female cancer in the world. Though therapeutic treatments are available to treat breast cancer and in some instances are successful, the occurrence of unsuccessful treatment, or the rate of tumour recurrence, still remains strikingly high. Therefore, novel therapeutic treatment targets need to be discovered and tested. The protein arginine methyltransferases (PRMTs) are a family of enzymes that catalyse arginine methylation and are implicated in a myriad of cellular pathways including transcription, DNA repair, RNA metabolism, signal transduction, protein–protein interactions and subcellular localisation. In breast cancer, the expression levels and enzymatic activity of a number of PRMTs is dysregulated; significantly altering the regulation of many cellular pathways that are implicated in breast cancer development and progression. Here, we review the current knowledge on PRMTs in breast cancer and provide a rationale for how PRMTs may provide novel therapeutic targets for the treatment of breast cancer.

    Beschreibung: It is well-established that the Coriolis force that acts on fluid in a rotating system can act to stabilise otherwise unstable flows. Chandrasekhar considered theoretically the effect of the Coriolis force on the Rayleigh-Taylor instability, which occurs at the interface between a dense fluid lying on top of a lighter fluid under gravity, concluding that rotation alone could not stabilise this system indefinitely. Recent numerical work suggests that rotation may, nevertheless, slow the growth of the instability. Experimental verification of these results using standard techniques is problematic, owing to the practical difficulty in establishing the initial conditions. Here, we present a new experimental technique for studying the Rayleigh-Taylor instability under rotation that side-steps the problems encountered with standard techniques by using a strong magnetic field to destabilize an otherwise stable system. We find that rotation about an axis normal to the interface acts to retard the growth rate of the instability and stabilise long wavelength modes; the scale of the observed structures decreases with increasing rotation rate, asymptoting to a minimum wavelength controlled by viscosity. We present a critical rotation rate, dependent on Atwood number and the aspect ratio of the system, for stabilising the most unstable mode. Scientific Reports 5 doi: 10.1038/srep11706

    Beschreibung: Choice of host plants by phytophagous insects is essential for their survival and reproduction. This choice involves complex behavioral responses to a variety of physical and chemical characteristics of potential plants for feeding. For insects of the order Hemiptera, these behavioral responses involve a series of steps including labial dabbing and probing using their piercing mouthparts. These initial probing and feeding attempts also elicit a rapid accumulation of phytohormones, such as jasmonic acid (JA), and the induced defense metabolites they mediate. When Nicotiana attenuata plants are rendered JA deficient by silencing the initial committed step of the JA biosynthesis pathway, they are severely attacked in nature by hemipteran leafhoppers of the genus Empoasca. By producing N. attenuata plants silenced in multiple steps of JA biosynthesis and perception and in the biosynthesis of the plant’s three major classes of JA-inducible insecticidal defenses, we demonstrate that the choice of plants for feeding by Empoasca leafhoppers in both nature and the glasshouse is independent of the accumulation of major insecticidal molecules. Moreover, this choice is independent of the presence of Candidatus Phytoplasma spp. and is not associated with detectable changes in plant volatiles but instead depends on the plant´s capacity to mediate JA signaling. We exploited this trait and used Empoasca leafhoppers to reveal genetic variation in JA accumulation and signaling hidden in N. attenuata natural populations.

    Beschreibung: Plant glandular trichomes exude secondary metabolites with defensive functions, but these epidermal protuberances are surprisingly the first meal of Lepidopteran herbivores on Nicotiana attenuata. O-acyl sugars, the most abundant metabolite of glandular trichomes, impart a distinct volatile profile to the body and frass of larvae that feed on them. The headspace composition of Manduca sexta larvae is dominated by the branched chain aliphatic acids hydrolyzed from ingested O-acyl sugars, which waxes and wanes rapidly with trichome ingestion. In native habitats a ground-hunting predator, the omnivorous ant Pogonomyrmex rugosus, but not the big-eyed bug Geocoris spp., use these volatile aliphatic acids to locate their prey.
