    Description: In this study, a universal protein expression enhancement RNA tool, termed RNAe, was developed by modifying a recently discovered natural long non-coding RNA. At the moment, RNAe is the only technology for gene expression enhancement, as opposed to silencing, at the post-transcriptional level. With this technology, an expression enhancement of 50–1000% is achievable, with more than 200% enhancement achieved in most cases. This work identified the sufficient and necessary element for RNAe function, which was found to be merely 300 nucleotides long and was named minRNAe. It contains a 72-nt 5' pairing sequence which determines the specificity, a 167-nt short non-pairing interspersed nuclear element (SINE) B2 sequence which enhances ribosome recruitment to the target mRNA, and a poly(A) tail, provided together on a plasmid bearing the appropriate sequences. Cellular delivery of RNAe was achieved using routine transfection. The RNAe platform was validated in several widely-used mammalian cell lines. It was proven to be efficient and flexible in specifically enhancing the expression of various endogenous and exogenous proteins of diverse functions in a dose-dependent manner. Compared to the expression-inhibitory tool RNAi, the RNAe tool has a comparable effect size, with an enhancing as opposed to inhibitory effect. One may predict that this brand new technology for enhancing the production of proteins will find wide applications in both research and biopharmaceutical production.

    Description: Site velocities derived from repeated measurements in a regional GPS network in Southeast Asia help to constrain the motion of tectonic blocks as well as slip rates along major faults in the area. Using 3-D forward dislocation modelling, the influence of seismic elastic loading and unloading on the measured site motions are approximated. Results suggest that the northwestern Sunda arc is fully coupled seismogenically, whereas its eastern part along Java shows localized deformation. Higher horizontal velocity gradients than expected from the modelling of a fully coupled plate interface west of Manila in the Philippines suggest that deformation may be localized there. Assuming that geodetically derived convergence represents long-term rates, accumulated geodetic moments are compared to those derived using seismic data from 1977 to 2000 (Harvard CMT catalogue). If areas displaying localized deformation are dominated by creep processes, the largest difference between accumulated and seismically released deformation is located where the 2000 June 4 Mw = 7.8 Sumatra earthquake occurred.

    Description: We investigate variations of the location and intensity of auroral currents during two magnetic storm periods based on magnetic field measurements from CHAMP separately for both hemispheres, as well as for the dayside and nightside. The corresponding auroral electrojet current densities are on average enhanced by about a factor of 7 compared to the quiet time current strengths. The nightside westward current densities are on average 1.8 (2.2) times larger than the dayside eastward current densities in the Northern (Southern) Hemisphere. Both eastward and westward currents are present during the storm periods with the most intense electrojets appearing during the main phase of the storm, before the ring current maximizes in strength. The eastward and westward electrojet centers can expand to 55° MLat during intense storms, as is observed on 31 March 2001 with Dst=−387 nT. The equatorward shift of auroral currents on the dayside is closely controlled by the southward IMF, while the latitudinal variations on the nightside are better described by the variations of the Dst index. However, the equatorward and poleward motion of the nightside auroral currents occur earlier than the Dst variations. The Space Weather Modeling Framework (SWMF) can capture the general dynamics of the storm time current variations. Both the model and the actual data show that the currents tend to saturate when the merging electric field is larger than 10 mV/m. However, the exact prediction of the temporal development of the currents is still not satisfactory.

    Description: We combine density functional theory (DFT) within the local density approximation (LDA), the quasiharmonic approximation (QHA), and a model vibrational density of states (VDoS) to calculate elastic moduli and sound velocities of γ-(Fex,Mg1−x)2SiO4 (ringwoodite), the most abundant mineral of the lower Earth's transition zone (TZ). Comparison with experimental values at room-temperature and high pressure or ambient-pressure and high temperature shows good agreement with our first-principles findings. Then, we investigate the contrasts associated with the β→γ(Fex,Mg1−x)2SiO4 transformation at pressures and temperatures relevant to the TZ. This information offers clearly defined reference values to advance the understanding of the nature of the 520 km seismic discontinuity.

    Description: Results acquired using global positioning system (GPS) data taken over a large part of SE-Asia, indicate that Sundaland, i.e. Indochina along with the western and central part of Indonesia, constitutes a stable tectonic block moving approximately east with respect to Eurasia at a velocity of 12±3 mm yr-1. With respect to India and Australia this block moves due south. Significant motion has not been detected along the northern boundary to South China i.e. along the Red River Fault, whereas nearly 50 mm yr-1 of right lateral motion has to be accommodated between India and Sundaland in the Andaman–Burma region.

    Description: [1] We present first-principles results for elastic moduli (bulk, K, and shear, G) and acoustic velocities (compressional, VP, shear, VS, and bulk VΦ) of olivine (α) and wadsleyite (β) (Fex,Mg1 − x)2SiO4, at high pressure (P) and temperature (T) with varying iron content (0 ≤ x ≤ 0.125). Pressure and temperature derivatives of these properties are analyzed. We show that adding 12.5% of Fe in forsterite softens VP and VS by ∼3–6%, the same effect as raising temperature by ∼1000 K in dry olivine at 13.5 GPa—the same is true in wadsleyite. This study suggests that Fe is effective in producing seismic velocity heterogeneity at upper mantle and transition zone conditions and should be another key ingredient, in addition to temperature and water content variations, in interpreting seismic heterogeneities in the transition zone. The effect of Fe on density, elastic, and velocity contrasts across the α → β transition is also addressed at relevant conditions. We show that simultaneous changes of composition, temperature, and pressure do not affect significantly the relative density contrasts. We also find that compressional and shear impedance contrasts result primarily from velocity discontinuities rather than density discontinuity.
