    Description: Cocaine abuse is a world-wide public health and social problem without a US Food and Drug Administration-approved medication. An ideal anticocaine medication would accelerate cocaine metabolism, producing biologically inactive metabolites by administration of an efficient cocaine-specific exogenous enzyme. Our recent studies have led to the discovery of the desirable, highly efficient cocaine hydrolases (CocHs) that can efficiently detoxify and inactivate cocaine without affecting normal functions of the CNS. Preclinical and clinical data have demonstrated that these CocHs are safe for use in humans and are effective for accelerating cocaine metabolism. However, the actual therapeutic use of a CocH in cocaine addiction treatment is limited by its short biological half-life (e.g., 8 h or shorter in rats). Here we demonstrate a novel CocH form, a catalytic antibody analog, which is a fragment crystallizable (Fc)-fused CocH dimer (CocH-Fc) constructed by using CocH to replace the Fab region of human IgG1. The CocH-Fc not only has a high catalytic efficiency against cocaine but also, like an antibody, has a considerably longer biological half-life (e.g., ∼107 h in rats). A single dose of CocH-Fc was able to accelerate cocaine metabolism in rats even after 20 d and thus block cocaine-induced hyperactivity and toxicity for a long period. Given the general observation that the biological half-life of a protein drug is significantly longer in humans than in rodents, the CocH-Fc reported in this study could allow dosing once every 2–4 wk, or longer, for treatment of cocaine addiction in humans.

Greatbatch, Peter Brandt, Dexing Wu, Xiaopei Lin & Lixin Wu Current climate models systematically underestimate the strength of oceanic fronts associated with strong western boundary currents, such as the Kuroshio and Gulf Stream Extensions, and have difficulty simulating their positions at the mid-latitude ocean’s western boundaries. Even with an enhanced grid resolution to resolve ocean mesoscale eddies—energetic circulations with horizontal scales of about a hundred kilometres that strongly interact with the fronts and currents—the bias problem can still persist; to improve climate models we need a better understanding of the dynamics governing these oceanic frontal regimes. Yet prevailing theories about the western boundary fronts are based on ocean internal dynamics without taking into consideration the intense air–sea feedbacks in these oceanic frontal regions. Here, by focusing on the Kuroshio Extension Jet east of Japan as the direct continuation of the Kuroshio, we show that feedback between ocean mesoscale eddies and the atmosphere (OME-A) is fundamental to the dynamics and control of these energetic currents. Suppressing OME-A feedback in eddy-resolving coupled climate model simulations results in a 20–40 per cent weakening in the Kuroshio Extension Jet. This is because OME-A feedback dominates eddy potential energy destruction, which dissipates more than 70 per cent of the eddy potential energy extracted from the Kuroshio Extension Jet. The absence of OME-A feedback inevitably leads to a reduction in eddy potential energy production in order to balance the energy budget, which results in a weakened mean current. The finding has important implications for improving climate models’ representation of major oceanic fronts, which are essential components in the simulation and prediction of extratropical storms and other extreme events, as well as in the projection of the effect on these events of climate change.

    Description: Fracture mechanisms of metallic glasses are fundamentally different from that of crystalline alloys. Nano-voids formation and coalescence inside shear bands were believed to be one of the reasons causing the final failures. Although molecule dynamic simulations have successfully simulated cavitation in shear bands of brittle metallic glasses, direct experimental evidences are still rare. By carefully examining the shear bands of Pd 40.5 Ni 40.5 P 19 , nano-voids and their coalescence have been observed in the center-diffused region of shear bands. It is experimentally confirmed that nano-voids formation and their coalescence into large voids within shear bands is one of fracture mechanisms of metallic glasses.

    Description: Species delimitation through which infers species boundaries is emerging as a major work in modern systematics. Hop-hornbeam species in Ostrya (Betulaceae) are well known for their hard and heavy woods. Five species were described in China and their interspecific delimitations remain unclear. In this study, we firstly explored their distributions in all recorded field sites distributed in China. We then selected 110 samples from 22 natural populations of five species from this genus and one type specimen of O. yunnanensis , for molecular barcoding analyses. We sequenced four chloroplast (cp) DNA fragments ( trnH–psbA , trnL–trnF , rps16, and trnG ) and the nuclear internal transcribed spacer (ITS) region for all samples. Sequence variations of Ostrya from four cpDNA fragments identified three groups that showed no correspondence to any morphological delimitation because of the incomplete lineage sorting and/or possible interspecific introgression in the history. However, phylogenetic analyses of ITS sequence variations discerned four species, O. japonica, O. rehderiana, O. trichocarpa, and O. multinervis while O. yunnanensis nested within O. multinervis . Morphological clustering also discerned four species and showed the complete consistency with molecular evidence. Moreover, our phylogenetic analyses-based ITS sequence variations suggested that O. trichocarpa comprised an isolated lineage different from the other Eurasian ones. Based on these results, hop-hornbeams in China should be treated as four separate species. Our results further highlight the importance of ITS sequence variations in delimitating and discerning the closely related species in plants. Both molecular and morphological evidence discerned four Ostrya species in China. cpDNA fragments identified three groups that showed no correspondence to any morphological delimitation because of the incomplete lineage sorting and/or possible interspecific introgression in the history. Phylogenetic analyses-based ITS sequence variations suggested that O. trichocarpa comprised an isolated lineage different from the other Eurasian ones.

    Description: Urban surface energy fluxes are closely associated with land-cover types (LCTs) and critical biophysical compositions. This study aims to assess the contribution of LCTs, vegetation fractional coverage (VFC) and percentage of impervious surface area (ISA%) to urban surface energy fluxes using remote sensing. An advanced urban surface energy flux algorithm was used to combine satellite imagery and meteorological station data to investigate the thermal environments in the city of Suzhou, China. The land cover abundances retrieved by multiple endmember spectral unmixing analysis (MESMA) were used to retrieve the per-pixel sensible heat flux (H) and latent heat flux (LE). The resultant heat fluxes were assessed using evaporation pan data collected from meteorological stations and ratios of the heat fluxes to the net radiation (Rn). Furthermore, spatial patterns of urban heat energy were investigated using an integrated analysis among land surface temperature (LST), heat fluxes, LCTs, VFC and ISA%. The high values of H and LST were found over the urbanized areas, which also had low values of LE. Conversely, the vegetated area was characterized with high LEs, as well as low LSTs and Hs. Moreover, a statistically-significant correlation (p 〈 0.05; R2 = 0.88) was observed between LE and VFC at the zonal level, and a statistically-significant correlation (p 〈 0.05; R2 = 0.90) was exhibited between H and ISA%. It is concluded that VFC, ISA% and LCTs are promising for delineating urban heat fluxes. Overall, this study indicates that remote sensing techniques can be used to quantify urban thermal environments.

    Description: Routing metric is very important for performance of wireless ad hoc sensor networks. ETX and ETT routing metrics cannot accurately estimate the average transmission time since they do not take back-off scheme of IEEE 802.11 in Mac layer into account. We present a new available transmission time (ATT) routing metric based on IEEE 802.11 DCF, which considers available link bandwidth and also takes physical transmission rates into account. A plugin for OLSR routing protocol daemon is implemented on Linux platform, and experiments are taken to compare ATT with ETX and ETT. Our results show that the ATT metric has the lowest packet loss rate and the lowest jitter rate among the analyzed metrics. ATT also outweigh other two metrics in the network performance, so it is the most appropriate for wireless ad hoc sensor networks.
