    Notes: Effects of organized turbulence structures on the propagation of an optical beam in a turbulent shear flow have been analyzed. An instantaneous passive-scalar field in a computed homogeneous turbulent shear flow is used to represent index-of-refraction fluctuations, and phase distortion induced in a coherent optical beam by turbulent fluctuations is calculated. The organized vortical structures ("hairpin-shaped'' eddies) in the turbulent flow give rise to a scalar distribution with elongated regions of intense fluctuation, which have an inclination (about 30°) with respect to the mean flow, similar to that of the characteristic "hairpin'' eddies. Two-point correlations of vorticity and scalar fluctuations support a proposed physical model in which the regions of intense scalar fluctuation are produced primarily by hairpin vortices. It is found that the spatial distribution of the phase distortion has a substantial variation with the direction of propagation. A highly localized distribution of intense phase distortions is produced when the optical beam propagates at an angle (45°) close to the inclination of hairpin vortices; at larger angles of propagation the distribution shows an elongated pattern with smaller phase distortions. It is also found that the root-mean-square phase distortion depends significantly on the propagation direction, and the phase distortion can be minimized at an angle of propagation approximately normal to the inclination of hairpin eddies. This study shows how the characteristics of an optical beam propagating through a turbulent shear flow are affected by the geometrical configurations of organized vortical structures.

    Notes: Quantification of the enhancement in cleavage fracture toughness of ferritic steels following warm pre-stressing has received great interest in light of its significance in the integrity assessment of such structures as pressure vessels. A Beremin type probability distribution model, i.e., a local stress-based approach to cleavage fracture, has been developed and used for estimating cleavage fracture following prior loading (or warm pre-stressing, WPS) in two ferritic steels with different geometry configurations. Firstly, the Weibull parameters required to match the experimental scatter in lower shelf toughness of the candidate steels are identified. These parameters are then used in two- and three-dimensional finite element simulations of prior loading on the upper shelf followed by unloading and cooling to lower shelf temperatures (WPS) to determine the probability of failure. Using both isotropic hardening and kinematic hardening material models, the effect of hardening response on the predictions obtained from the suggested approach has been examined. The predictions are consistent with experimental scatter in toughness following WPS and provide a means of determining the importance of the crack tip residual stresses. We demonstrate that for our steels the crack tip residual stress is the pivotal feature in improving the fracture toughness following WPS. Predictions are compared with the available experimental data. The paper finally discusses the results in the context of the non-uniqueness of the Weibull parameters and investigates the sensitivity of predictions to the Weibull exponent, m, and the relevance of m to the stress triaxiality factor as suggested in the literature.

    Notes: Abstract An optical tomography system that is capable of operating at frame rates of up to 5 kHz has been used to obtain spatially resolved cross-sectional temperature images of a heated round jet. These tomographic images show dynamic details in the evolving vortical flow structures found in the near field of the jet that are consistent with previous studies of low speed jet flow. Reconstructions produced by the system are quantitative temperature distributions of a planar cross section of the jet measuring temperature differences with a spatial resolution of 1.4 mm.

    Notes: Abstract The aim of this study was to characterise the plasma protein binding of tacrine hydrochloride (THA) in vitro. Binding was assessed in the plasma of 11 healthy individuals aged 20 to 27 years using ultrafiltration followed by HPLC assay. At THA concentrations from 10 to 100 ng/ml protein binding ranged from 78.6 to 71.0%. Binding to commercially available human albumin ranged from 41.7 to 38.3% and to human α1-acid glycoprotein from 23.1 to 12.4% over the THA concentrations from 25 to 100 ng/ml. THA binding and total plasma protein, plasma albumin and α1-acid glycoprotein were measured in healthy young subjects (n=13), healthy elderly individuals (n=12) and patients hospitalised with acute illnesses (n=8). There were significant differences between the groups in total plasma protein, plasma albumin and in α1-acid glycoprotein but no differences in the protein binding of THA which remained constant at about 75%. There was no correlation between THA binding and any plasma protein concentration. The THA binding was not high enough to be of major significance clinically or to reduce the validity of total plasma THA measurement in therapeutic monitoring.

    Notes: Abstract Objectives: The purpose of this study was to validate the lower end of the putative therapeutic range of serum tacrine concentrations of 7–20 ng · ml−1 in the treatment of Alzheimer's disease. Methods: The relationship between dose, steady-state serum tacrine concentrations and change in MMSE score (a measure of cognitive function) was examined in 106 Alzheimer's disease patients who had been treated with the drug for 12 weeks. Results: In all, 72% of patients showed some response, but there was no relationship between dose and the chance of a favourable outcome. The proportion of patients with serum concentrations above 7 ng · ml−1 who improved (79%) was significantly greater than that of those with serum concentrations below this level (47%) (P 〈 0.02). Also, a significantly greater proportion of patients with serum concentrations above both 5 ng · ml−1 and 9 ng · ml−1 showed improvement in comparison to those with concentrations below these levels. Conclusions: This study indicates that therapeutic monitoring of serum tacrine concentrations might increase the possibility of responding to tacrine by some 68%. This represents an important contribution to the management of Alzheimer's disease patients with this drug, and may also be relevant to the use of the newer generation of cholinesterase inhibitors.

    Description: ATM (ataxia-telangiectasia mutated) is a phosphatidylinositol 3-kinase–related protein kinase (PIKK) best known for its role in DNA damage response. ATM also functions in oxidative stress response, insulin signaling, and neurogenesis. Our electron cryomicroscopy (cryo-EM) suggests that human ATM is in a dynamic equilibrium between closed and open dimers. In the closed state, the PIKK regulatory domain blocks the peptide substrate–binding site, suggesting that this conformation may represent an inactive or basally active enzyme. The active site is held in this closed conformation by interaction with a long helical hairpin in the TRD3 (tetratricopeptide repeats domain 3) domain of the symmetry-related molecule. The open dimer has two protomers with only a limited contact interface, and it lacks the intermolecular interactions that block the peptide-binding site in the closed dimer. This suggests that the open conformation may be more active. The ATM structure shows the detailed topology of the regulator-interacting N-terminal helical solenoid. The ATM conformational dynamics shown by the structures represent an important step in understanding the enzyme regulation.

    Description: 〈p〉A shock propagating through a gas mixture leads to pressure, temperature, and density increases across the shock front. Rankine-Hugoniot relations correlating pre- and post-shock quantities describe a calorically perfect gas but deliver a good approximation for real gases, provided the pre-shock conditions are well characterized with a thermodynamic mixing model. Two classic thermodynamic models of gas mixtures are Dalton’s law of partial pressures and Amagat’s law of partial volumes. We measure post-shock temperature and pressure in experiments with nonreacting binary mixtures of sulfur hexafluoride and helium (two dramatically disparate gases) and show that neither model can accurately predict the observed values, on time scales much longer than that of the shock front passage, due to the models’ implicit assumptions about mixture behavior on the molecular level. However, kinetic molecular theory can help account for the discrepancy. Our results provide starting points for future theoretical work, experiments, and code validation.〈/p〉
