    Notizen: Abstract We describe differences in senescence among populations of grasshoppers that occur along an altitudinal gradient in the Sierra Nevada, California. Experimental males from five populations of the Melanoplus sanguinipes/devastator sibling species complex were reared in each of two thermal culture conditions from eggs of field-caught females. In both culture conditions, we measured the survival of adult cohorts from each population. Differences in the physiological capacity to survive in a sheltered, common environment reveal genetic differences in underlying rates of senescence, providing maternal effects do not affect the rate of aging in offspring. At each temperature, mortality rates increased as a function of age, which indicates that senescence occurs under laboratory conditions. Within each culture condition, cohorts originating from low-elevation populations had greater survival than did cohorts from high elevations. Variation in body size along the altitudinal gradient did not account for the differences in survival. We suggest that high-elevation populations have evolved accelerated senescence as a direct or an indirect result of selection on reproductive schedules, which are potentially truncated by severe winter conditions at the elevated sites.

    Notizen: Summary Clutch size in the pipevine swallowtail of California (Battus philenor hirsuta) varies widely within and among two seasonal flights, early and late spring. Pilson and Rausher (1988) proposed that variation in clutch size in a Texan B. philenor population is due to assessment of host quality with subsequent adjustment of clutch size and to differences in motivational state. The assessment hypothesis and a nonexclusive alternative that clutch size is determined by mature egg load were tested for populations in Sacramento, California. Weak and sporadic correlations between host quality and clutch size were found, suggesting that host assessment with adjustment of clutch size is not a strong factor in this population. Clutch size was highly correlated to mature egg load in the early but not in the late spring flight. Yet, females from both flights had similar loads of mature eggs. It is proposed that females lay clutch sizes proportional to egg load, up to a maximum threshold. The threshold may be lowered in the late spring flight in response to changes in overall host quality or to increased egg predation, reducing both the mean clutch size and the correlation between clutch size and egg load. Potential mechanisms for the change in behavior among flights, based on changes in the threshold, are discussed.

    Notizen: Abstract We are interested in localizing chromosomal regions that extend life span in Drosophila. Using stocks artificially selected for long life by Luckinbill and his colleagues, we have identified marker loci that are highly divergent in allelic frequencies between replicated long-lived lines and controls (Curtsinger et al., 1998). Several of the most divergent loci have been found to be associated with effects on life span in segregating backcross populations. Here we report an independent replication of the backcross test for the N14 marker locus, previously reported to extend male life spans by 12 days. The life span effect successfully replicates in males. N14 accounts for 30% of the total selection response in males. Life span extension occurs by a decrease in age-specific mortality rates at all ages, and is not attributable to modification of the slope of the age-specific mortality curve. The effect in females is small or nonexistent. Sequencing of the N14 locus shows that it is non-coding and not obviously regulatory, suggesting that the phenotypic effect arises from linkage disequilibrium with another locus or loci that directly affect life span. N14 DNA hybridizes to 63F/64A on the left arm of chromosome 3. The location is consistent with previous whole-chromosome substitution studies, and suggests new candidate genes for life span extension in Drosophila, including ras2.

    Notizen: Abstract Two evolutionary genetic models–mutation accumulation and antagonistic pleiotropy–have been proposed to explain the origin and maintenance of senescence. In this paper, we focus our attention on the mutation accumulation model. We re-examine previous evidence for mutation accumulation in light of new information from large-scale demographic experiments. After discussing evidence for the predictions that have been put forth from models of mutation accumulation, we discuss two critical issues at length. First, we discuss the possibility that classical fruit fly stock maintenance regimes may give rise to spurious results in selection studies of aging. Second, we consider evidence for the assumptions underlying evolutionary models of aging. These models assume that mutations act additively on age-specific survival rate, that there exist mutations whose effects are confined to late age-classes, and that all mutations have equal effects. Recent empirical evidence suggests that each of these three assumptions is unlikely to be true. On the basis of these results, we do not conclude that mutation accumulation is no longer a valid explanation for the evolution of aging. Rather, we suggest that we now need to begin developing more biologically realistic genetic models for the evolution of aging.
