    Notes: GALACTINOL SYNTHASE (GOLS) acts on UDP-galactose and myo-inositol to form galactinol. The production of galactinol is thought to be the committed step in the biosynthesis of the raffinose family oligosaccharides (RFOs), a group of carbohydrates hypothesized to play a number of roles in plant development. Three cDNAs for maize (Zea mays L. inbred line B73) GOLS (ZmGOLS) were identified from the Pioneer Hi-Bred expressed sequence database. The recombinant proteins from these cDNAs synthesize galactinol. Transcript amounts in developing and germinating seeds were determined using northern blot, ribonuclease protection assays, and multiplex RT-PCR. ZmGOLS1 mRNA was not detected in any of the tissues examined. ZmGOLS3 transcripts accumulated late during seed development and were present in mature seeds but declined quickly to undetectable amounts following imbibition. Experiments in which germination was interrupted by desiccation, but not by cold, heat, or salt stress, detected ZmGOLS2 transcripts. Soluble sugar amounts and GALACTINOL SYNTHASE activity were determined for developing maize seeds as well as for mature seeds during germination and upon stress during germination. Neither enzyme activity nor galactinol or raffinose quantities could be related to ZmGOLS transcript abundance. Galactinol amounts were most closely related to myo-inositol quantities during development and early germination. Maturation desiccation appears to be a switch delineating a realm of expression for ZmGOLS3, predominant during seed development, from that of ZmGOLS2, principally associated with post-imbibition dehydration stress.

    Notes: Summary Leaflets from 11 L. esculentum x L. hirsutum F2 individuals were bioassayed with spider mites (Tetranychus urticae). Densities of Type I, Type III, Type IV, Type V, and Type VI trichomes and the intensity of red fluorescence of the Type VI trichome tip varied widely among the 11 F2 hybrids. Mite responses were analyzed in relation to these trichome characters and leaflet surface. Variation in Type IV density accounted for most of the variation in mite responses. Densities of Type I, V and VI trichomes also contributed to mite responses but their effects were obscured by high Type IV densities. Type VI tip fluorescence was not associated with responses of mites in bioassays of these hybrids.

    Notes: Summary Density of certain types of trichomes were characterized on leaflets of Lycopersicon hirsutum, L. esculentum and their hybrids. Trichome density covaried with leaflet development, and depending on the species and type of trichome, density also covaried with leaflet surface. Type IV trichomes were not observed on leaflets of L. esculentum and Type V trichomes were not observed on leaflets of L. hirsutum. Type IV and V trichomes were present on F1 hybrids and densities of these two types of trichomes segregated in the F2 population. When corrected for differences of leaflet size between L. hirsutum and L. esculentum, densities of Type VI trichomes were more similar between species than uncorrected densities. There was little difference in Type VI density among F2 individuals. The appearance, lipid and phenol staining properties, and native fluorescence of the multicellular tip of the Type VI trichome differed between L. hirsutum and L. esculentum. F1 hybrids were intermediate for these characters and these characters segregated in the F2 generation.

    Notes: Abstract Bioassays employingTetranychus urticae Koch were used to examine repellency of leaves and trichome secretions of one cultivar of tomato,Lycopersicon esculentum, and 11 accessions ofL. hirsutum consisting of seven accessions ofL. hirsutum f.typicum and four accessions ofL. hirsutum f.glabratum. Leaves of f.typicum were more repellent to mites than were those of f.glabratum orL. esculentum. Removal of trichomes and their secretions by wiping leaves of f.typicum accessions with 95% ethanol rendered them less repellent, indicating that trichomes or their secretions were potentially responsible for repellency. Gas and thin-layer chromatography of leaflet washes obtained by steeping leaflets in hexane indicated that the chemical composition of the washes differed among accessions. The main difference between botanical forms was the presence in leaflet washes from f.typicum accessions of compounds eluting late on the gas Chromatograph and migrating further on reverse-phase thin-layer chromatography; compounds having these characteristics were absent in washes of f.glabratum andL. esculentum. The compositions of secretions taken directly from type I, type IV, and type VI trichomes were similar to that of the leaflet wash obtained from the same f.typicum plant, indicating that leaflet washes contained mainly compounds of trichomal origin. Leaflet washes from f.typicum plants were generally more repellent toT. urticae than were washes from f.glabratum when tested by choice and nonchoice bioassays. Taken together, the data indicate that trichome secretions of f.typicum are likely responsible for potent repellency to two-spotted spider mites observed in this taxon; the likely active compounds are sesquiterpene acids or other polar constituents of trichome secretions.

    Notes: Abstract Lycopersicon hirsutum, a wild relative of the tomato, is highly resistant to arthropod herbivores. Both botanic forms ofL. hirsutum, L. hirsutum f.glabratum (C.H. Mull.) andL. hirsutum f.typicum (Humb. & Bonpl.), are resistant to two-spotted spider mites,Tetranychus urticae Koch. However, leaves and trichome secretions from f.typicum repel mites more so than those from f.glabratum. We have previously demonstrated that trichome secretions from LA 1363 and LA 1927, accessions of f.typicum, repelled mites. In this paper we report the identification of the primary component of trichome secretions responsible for repellency. Leaflet washes having compositions similar to trichome secretions were collected and separated into neutral and acid fractions; repellency was mainly associated with the acid fraction, which, when applied to nonrepellent leaflets of f.glabratum, rendered them repellent. Separation of leaflet washes by HPLC allowed purification and subsequent identification by gas chromatography-mass spectrometry and nuclear magnetic resonance of 2,3-dihydrofamesoic acid (3,7,11-trimethyl-6, 10-dodecadienoic acid) as the primary chemical component responsible for repellency. Application of this acid to leaflets ofL. esculentum rendered them repellent. Other volatile compounds present in minor amounts in the acid fractions were farnesoic acid and 16∶0, 16∶3, 18∶0, 18∶2, and 18∶3 fatty acids. This is the first report of the natural occurrence of 2,3-dihydrofarnesoic acid.
