    Notes: Egg white was diluted to 10 times its original volume with 50 mM phosphate buffer (pH 9.2) containing 0.1 M potassium chloride. The aqueous solution was mixed with an equal volume of isooctane containing 50 mM bis-(2-ethylhexyl) sodium sulfosuccinate at 10°C for 50 min. After extraction, the organic phase containing lysozyme was separated from the aqueous phase and mixed with an equal volume of 50 mM phosphate buffer (pH 11.8) containing 1M potassium chloride. Backward extraction was then performed at 30°C for 45 min. The procedures recovered 90% lysozyme from the eggwhite. The specific activity of the extract was near 73,000 units/mg.

    Notes: Environmental factors affecting basidiospore germination were studied with Volvariella volvacea, the edible straw mushroom which is common in South East Asia. A relatively mild heal shock is necessary for spore germination. The spores give best germination at 40°C although early hyphal growth is better at 35°C the germination of spores is affected by temperature. pH. a presoaking treatment and spore density. Higher pH supports more germination but seems unfavourable for early mycelial growth. Presoaking treatment in phosphate buffer solution or distilled water also stimulates germination markedly.

    Notes: Abstract Lentinula (Lentinus) edodes, strain LS4, produces manganese-dependent peroxidase (MnP) and laccase, but not lignin peroxidase, when grown on a defined medium with glucose as sole carbon source. MnP production is suppressed by nitrogen whereas highest levels of laccase were observed when the fungus was grown under high nitrogen (26 mM) conditions. Both the titre and time of appearance of MnP were affected by the concentration of Mn in the culture medium with highest enzyme levels recorded in cultures supplemented with 1.1 ppm Mn. Purified MnP from L. edodes LS4 has an apparent Mr of 59000 and a pI of 5.6, and differs in several respects from a MnP isolated from L. edodes grown on a commercial wood substrate.

    Notes: Abstract This paper presents the concept of a Dielectric-lined Multiwave Cerenkov Generator(DMWCG) producing high power millimeter waves, which is investigated with a two and one half dimensional( $$2\frac{1}{2}{\text{D}}$$ ) electromagnetic relativistic Particle-in-Cell(PIC) simulation code. It is showed that the DMWCG can operate in a lower diode-voltage regime with much higher radiation efficiency as compared with the usual Multiwave Cerenkov Generator(MWCG). The simulation work indicates both the downshift of the wave frequency in the presence of the dielectric liner and the existence of the optima for the permittivity of the liner as well as for the magnitude of the guiding magnetic field. The required intensity of the guiding field is reduced with the introducing of the liner. The radiation is generated at the dominant frequency of 31.5GHz. The power level of 1.5GW is achieved, with radiation efficiency up to 15%. The features of parameter dependency are presented. And reasonable explanation is put forward. In addition, the enhanced propagation of the electron beam is studied in the presence of the dielectric liner.

    Notes: Abstract Volvariella volvacea, the straw mushroom, has long been considered as a primary homothallic species. However, great variations in monosporous isolates have ralsed a considerable debate on the sexuality pattern of this edible Basidiomycete fungus. Because clamp connection is absent in V. volvacea, the production of genetically marked strains offers a possible means of resolving this problem. In this study, ultraviolet irradiation of basidiospores and mycellal fragments was used to induce crystal-violet- and malachite-green-resistant mutants. Cross-resistance to crystal violet and malachite green occurred between both classes of mutant. The difficulties of inducing mutants in this multinucleate fungus and the significance of mutant markers are discussed.

    Notes: Abstract After regeneration of protoplasts of seven heterothallic of edible mushrooms, two types of monokaryons were recovered, being identified by the absence of clamp connections. In regenerated colonies, monokaryons grew more slowly than dikaryons and so could be distinguished from them. In all species, the yield of monokaryons in the regenerated colonies was over 45%. The mating types and auxotrophic markers were the same in the protoplasted monokaryons and parental monokaryons. In comparison with other monokaryotization methods, protoplasting is rapid, simple and effective; its applications in breeding of edible mushrooms are discussed.
